
Healthy babies born in Britain after scientists used DNA from three people to avoid genetic disease
Most DNA is found in the nucleus of our cells, and it's that genetic material — some inherited from mom, some from dad — that makes us who we are. But there's also some DNA outside of the cell's nucleus, in structures called mitochondria. Dangerous mutations there can cause a range of diseases in children that can lead to muscle weakness, seizures, developmental delays, major organ failure and death.
Testing during the in vitro fertilization process can usually identify whether these mutations are present. But in rare cases, it's not clear.
Researchers have been developing a technique that tries to avoid the problem by using the healthy mitochondria from a donor egg. They reported in 2023 that the first babies had been born using this method, where scientists take genetic material from the mother's egg or embryo, which is then transferred into a donor egg or embryo that has healthy mitochondria but the rest of its key DNA removed.
The latest research 'marks an important milestone,' said Dr. Zev Williams, who directs the Columbia University Fertility Center and was not involved in the work. 'Expanding the range of reproductive options … will empower more couples to pursue safe and healthy pregnancies.'
Using this method means the embryo has DNA from three people — from the mother's egg, the father's sperm and the donor's mitochondria — and it required a 2016 U.K. law change to approve it. It is also allowed in Australia but not in many other countries, including the U.S.
Experts at Britain's Newcastle University and Monash University in Australia reported in the New England Journal of Medicine Wednesday that they performed the new technique in fertilized embryos from 22 patients, which resulted in eight babies that appear to be free of mitochondrial diseases. One woman is still pregnant.
One of the eight babies born had slightly higher than expected levels of abnormal mitochondria, said Robin Lovell-Badge, a stem cell and developmental genetics scientist at the Francis Crick Institute who was not involved in the research. He said it was still not considered a high enough level to cause disease, but should be monitored as the baby develops.
Dr. Andy Greenfield, a reproductive health expert at the University of Oxford, called the work 'a triumph of scientific innovation,' and said the method of exchanging mitochondria would only be used for a small number of women for whom other ways of avoiding passing on genetic diseases, like testing embryos at an early stage, was not effective.
Lovell-Badge said the amount of DNA from the donor is insignificant, noting that any resulting child would have no traits from the woman who donated the healthy mitochondria. The genetic material from the donated egg makes up less than 1% of the baby born after this technique.
'If you had a bone marrow transplant from a donor … you will have much more DNA from another person,' he said.
In the U.K., every couple seeking a baby born through donated mitochondria must be approved by the country's fertility regulator. As of this month, 35 patients have been authorized to undergo the technique.
Critics have previously raised concerns, warning that it's impossible to know the impact these sorts of novel techniques might have on future generations.
'Currently, pronuclear transfer is not permitted for clinical use in the U.S., largely due to regulatory restrictions on techniques that result in heritable changes to the embryo,' Williams, of Columbia, said in an email. 'Whether that will change remains uncertain and will depend on evolving scientific, ethical, and policy discussions.'
For about a decade, Congress has included provisions in annual funding bills banning the Food and Drug Administration from accepting applications for clinical research involving techniques, 'in which a human embryo is intentionally created or modified to include a heritable genetic modification.'
But in countries where the technique is allowed, advocates say it could provide a promising alternative for some families.
Liz Curtis, whose daughter Lily died of a mitochondrial disease in 2006, now works with other families affected by them. She said it was devastating to be told there was no treatment for her eight-month-old baby and that death was inevitable.
She said the diagnosis 'turned our world upside down, and yet nobody could tell us very much about it, what it was or how it was going to affect Lily.' Curtis later founded the Lily Foundation in her daughter's name to raise awareness and support research into the disease, including the latest work done at Newcastle University.
'It's super exciting for families that don't have much hope in their lives,' Curtis said.

'Miracle' five years since twins' Covid coma birth
A doctor whose twins were delivered prematurely while she was in a coma says the five years since have been like a series of "miracles unfolding". Dr Perpetual Uke, a rheumatology consultant at Birmingham City Hospital, was placed into an induced coma after catching Covid-19 before her twins were delivered by Caesarean section at 26 weeks on 10 April 2020. She came around 16 days later, convinced her twins were dead, before hospital staff told her they were alive. The twins turned five years old in April and both have learned to walk and talk, which Dr Uke said was something she "cherished". "I just hope for the best because everything about it has been miracles unfolding, just like how you turn the pages of your book," she said. Dr Uke said she was at high-risk of becoming seriously ill with Covid but continued to work during the pandemic because she felt a duty of care to her patients. After catching the disease, Dr Uke was admitted to the hospital's critical care unit, placed on a ventilator and put in an induced coma to help her recover. When they were born, her daughter weighed just 770g (27oz) while her brother, weighed 850g (30oz). Dr Uke's husband Matthew said he had conflicting emotions when his children were born. "If the twins are there - where is my wife? Because by then, it was not certain she was going to come out of her coma," he said. "Am I going to have to care for the twins alone? It was a very difficult moment." Dr Uke recalled the "surreal" experience of waking and how she found it difficult to believe her children were alive, until she saw them for the first time. Despite what her husband described as the "grind" of the past five years, Dr Uke said her outlook had become more positive. "We are very glad, happy and joyous because of what they've achieved and are going to achieve in the future," she said. Mr Uke described 2020 as like "being in a dark moment" and 2025 was "like being in the sunshine". "We are very happy and we're very thankful to the NHS, to all the people who cared for them and that continue to care for them," he said. "They're wonderful. Their dedication and passion are off the charts. All the people who work in the NHS - they are special." Dr Uke has since written a book about the family's story called Covid Coma to Twin Birth: Threads of Miracles, which she hoped would help others going through difficult times. Europe working to launch 'Invictus' hypersonic space plane by 2031 (video)

The European Space Agency (ESA) is funding the development of a hypersonic space plane pathfinder, which will start flying by 2031 if all goes according to plan. The work is being done via a research program called Invictus, which is led by the consulting firm Frazer-Nash. Invictus will leverage technology developed by the English company Reaction Engines Ltd., which aimed to build a huge space plane called Skylon but went bankrupt last year. The key piece of Invictus tech is a "pre-cooler," which Reaction Engines built and tested for its Synergetic Air-Breathing Rocket Engine (SABRE). SABRE combined aspects of jet and rocket propulsion; it was designed to pull oxygen out of the air during flight at lower levels of Earth's atmosphere, reducing the need to carry propellant and therefore increasing efficiency. "Aircraft that fly at hypersonic speeds — more than 5 times the speed of sound — face extremely high temperatures due to shock heating and the friction from the air. Typical aircraft engines cannot operate in these conditions, as the air is too hot to handle," Frazer-Nash representatives said in a statement. This pre-cooler solves this problem, cooling "the air before it reaches the engine, allowing conventional aircraft engines to travel at hypersonic speeds," Frazer-Nash added. Invictus isn't a huge project; its funding is 7 million pounds (about $9.4 million US at current exchange rates), according to the company. But it could have a big impact on European spaceflight, according to ESA. "Hypersonic flight is not just the next frontier of aerospace — it is the gateway to a new paradigm of mobility, defense, and space access," Tommaso Ghidini, head of the Mechanical Department at the agency, said in the same statement. "With Invictus, Europe is seizing the opportunity to lead in technologies that will redefine how we move across the planet and reach beyond it," he added. "By mastering reusable, air-breathing propulsion, we are laying the foundation for aircraft that take off like planes and reach orbit like rockets — revolutionizing both terrestrial and orbital transportation." The "full flight system" will be a reusable vehicle that takes off from, and lands on, a runway like an airplane. It will be up and running by early 2031, if all goes to plan, and could have a variety of uses and applications. "We look forward to seeing how the work develops and the opportunity it presents for boosting economic growth and national security," Tony Forsythe, head of space technology at the U.K. Space Agency, said in the same statement. Invictus isn't the only European space plane in development. Last month, for example, the French government and the French company Dassault Aviation announced plans for a demonstrator called VORTEX. Space planes are experiencing something of a resurgence after the retirement of the most famous such vehicle — NASA's space shuttle — in 2011. The U.S. military operates a robotic orbital space plane called the X-37B, for example, and China has a similar vehicle, called Shenlong. Virgin Galactic flies a suborbital space plane for tourism and research purposes. A number of other companies are developing space planes as well, including Sierra Nevada Corp., Dawn Aerospace and Radian Aerospace.

'Powerful story' of triple amputee shared at RAF Brize Norton event day
An event held at RAF Brize Norton heard from a triple amputee injured in the line of duty as he told his "powerful story". Mark Ormrod MBE, a veteran who lost both legs and an arm while serving as a Royal Marine in 2007 and who is now an athlete, author and motivational speaker, spoke at the Carterton base's Resilience Day hosted by the Tactical Medical Wing. His "powerful story of resilience" resonated with the team and encouraged reflection and conversation on how personnel can better support themselves and each other.
