New blood test screens for thousands of rare inherited diseases at once

ST. PAUL, Minn., May 25 (UPI) -- A new rapid blood test for newborns could potentially detect genetic mutations linked to thousands of rare diseases all at once, greatly improving on current inefficient detection methods, according to a study to be presented Monday.
The new test developed by Australian scientists has proven highly accurate in identifying gene mutations associated with many rare, inherited diseases, all from just a minimally invasive blood sample taken from infants and children, the authors say.
The study, being unveiled at the European Human Genetics Conference in Milan, Italy, demonstrated that a single, untargeted test capable of analyzing 8,000 human proteins at once was able to correctly identify 83% of people with confirmed rare, inherited diseases.
The "proteomics" test was also able to differentiate between parental carriers of the mutations, who only have one copy of the defective gene, and the affected child, who carries two copies.
Those encouraging results are raising hopes for a new era in which screening infants and children for suspected inherited rare diseases can be accomplished quickly and efficiently for the first time, and that testing can be extended to many more of the estimated 300 million people worldwide affected by these genetic mutations.
Testing for suspected inherited diseases has traditionally been a time-consuming, costly and sometimes painful process that required different procedures for different suspected mutations. But that paradigm could be about to change, the study's lead author says.
Daniella Hock, a senior postdoctoral researcher at the University of Melbourne, told UPI that if the test is implemented in clinical labs, "it can potentially replace multiple functional tests. This can potentially reduce the diagnostic time for patients and families and healthcare costs.
"The test only requires only 1 milliliter of blood from infants, and results can be achieved in less than three days for urgent cases," she said.
One area in which the new proteomics test could have immediate benefits is in the battle against mitochondrial diseases, which are defined by the Cleveland Clinic as a group of genetic conditions that affect how mitochondria in human cells produce energy. Mitochondrial diseases render cells unable to produce enough energy, which can lead to life-threatening complications.
About 1 in 5,000 people worldwide have a mitochondrial disorder. Examples of such illnesses are Leigh's syndrome, which primarily affects the nervous system; Kearns-Sayre syndrome, which primarily affects the eyes and heart; and Leber hereditary optic neuropathy, which can cause a sufferer to quickly and unexpectedly lose their vision.
Hock said the current "gold standard" for screening for these rare maladies starts with genetic testing -- either whole exome or whole genome sequencing -- that currently provides a diagnosis to about half of patients.
"The remaining half often endure years of functional testing trying to identify which genetic change, that is, genetic variant, is causing the disease," she said, adding that more than 7,000 types of disease caused by mutations exist in more than 5,000 known genes.
Some of these current tests are invasive, requiring skeletal muscle or liver biopsies, and are often targeted to a single disease or a few diseases.
"If we take the example of mitochondrial disease, which is a type of rare disease that can be caused by over 350 different genes, the current clinical test to confirm mitochondrial disease is called respiratory chain enzymology," she said. "This enzyme test typically requires a skeletal muscle or skin biopsy and has a turnaround time of a few weeks."
The new proteomic test, which sequences proteins rather than genes, can reduce invasiveness and time to a diagnosis compared to other kinds of current clinical functional tests, its developers say.
"For about half of the individuals where DNA sequencing results are inconclusive, typically due to the identification of variants of uncertain significance, this single test can potentially be used to provide functional evidence to these genetic variants," Hock said.
Study co-author David Thorburn, one of Australia's foremost experts on genetics and co-group leader for brain and mitochondrial research at Murdoch Children's Research Institute in Melbourne, said the current process for determining if a child has one of the many possible genetic variances is a daunting one.
While there are many hundreds of different functional tests tailored to specific genes, "relatively few of these are available in clinical labs, so it is often a matter of contacting researchers who work on that gene, of which there may only be one or a handful internationally to see if they can assist," he told UPI in emailed comments.
"That often requires paperwork like material transfer agreements to be put in place, shipping costs of hundreds or even thousands of dollars if dry ice shipment is needed and frequently a timeframe of months to multiple years depending on whether they have a student or staff to do testing in a research context, where the rigor of the test may be variable," he added.
Thorburn said the new proteomics test, however, "provides a single pipeline that can potentially provide that evidence for about half the known rare disease genes and we are working hard to move it into a clinical test in Melbourne at our Victorian Clinical Genetics Services and working with expert clinical labs in the U.K., U.S.A. and elsewhere to support them doing that."
Having the test available clinically "could allow hundreds of families each year in Australia alone to get confirmed genetic diagnoses, so potentially thousands per year in the U.S.," he said. "For urgent cases, [for example], kids in ICU this can be done in as few as three days from sample receipt."
For those many thousands of patients, the new test could mean their "diagnostic odyssey is ended, unnecessary investigations are no longer needed, targeted therapies may be available, patients may qualify for a clinical trial and parents can be offered reproductive options," he said.

The scrolls could be centuries older than initially thought, according to a study that combined radiocarbon dating with AI to better analyze the remnants of ancient documents, The Times of Israel reported. "The Dead Sea Scrolls ... completely changed the way we think about ancient Judaism and early Christianity," said Mladen Popovic, lead author of the study that was published Wednesday in the journal PLOS One. "Out of 1,000 manuscripts, a bit more than 200 are what we call biblical Old Testament," Popovic told CNN. "They are the oldest copies we have of the Hebrew Bible." Popovic is the dean of the Faculty of Religion, Culture and Society at the University of Groningen in the Netherlands. Archaeologists recovered thousands of remnants of scrolls that were first discovered in 1947 in the Judean Desert by Bedouin shepherds in an area that has become the West Bank. Instead of dating the scrolls based on the form of their lettering, researchers used carbon dating to analyze samples from 30 of the Dead Sea Scrolls that were provided by the Israel Antiquities Authority. They also created high-resolution copies of the scripts and used an AI-powered model called "Enoch" to analyze the textual characters contained in 135 scrolls. The study revealed the scrolls are older than initially thought, which is from the 3rd century B.C. to the 1st century A.D. A paleographic study of the text within the scrolls narrowed their origin to that timeframe in 1961, but little else was done to analyze their origin until now. The new study pretreated pieces of parchment to remove any chemical traces from prior studies before undertaking carbon dating, and AI analysis corroborates the results. It suggests some of the scrolls were one or two centuries older than originally thought, including Old Testament books like Ecclesiastes. The study also suggests literacy was much more widespread in the region. "These manuscripts are not just the earliest copy of these [Old Testament] books that survived," IAA Dead Sea Scrolls Unit leader Joe Uziel told The Times of Israel. They are "one of the oldest copies of these compositions ever written," he said. Only about 10% of the scrolls were studied, which Popovic said means there is a lot more to learn through more studies of the Dead Sea Scrolls.

The Veterans Administration counters the new proposed budget is higher than last year, processing of claims have sped up and it's easier to get benefits. Veterans, military families and others participated in the Unite for Veterans, Unite for America Rally on the 81st anniversary of D-Day, which was the Allies' amphibious invasion of German-occupied France. The protests, which were organized by a union, took place at 16 state capitol buildings and more than 100 other places across 43 states. "We are coming together to defend the benefits, jobs and dignity that every generation of veterans has earned through sacrifice," Unite for Veterans said on its website. "Veteran jobs, healthcare, and essential VA services are under attack. We will not stand by." Speakers in Washington included Democrats with military backgrounds: Sen. Tammy Duckworth of Illinois, former Rep. Conor Lamb of Pennsylvania and California Rep. Derek Tran. There were signs against President Donald Trump, VA Secretary Doug Collins and Elon Musk, the multi-billionaire who ran the Department of Government Efficiency. They said those leaders are betraying the country's promises to troops. "Are you tired of being thanked for our service in the public and stabbed in our back in private?" Army veteran Everett Kelly, the national president of the American Federation of Government Employees, asked the crowd. "For years, politicians on both sides of the aisle have campaigned on their support of veterans, but once they get into office, they cut our benefits, our services. They take every opportunity to privatize our health care." The Trump administration plans to cut 83,000 VA staffers and shift more money from the federal health care system to private-sector clinics. The administration's proposed budget for the VA, released on Friday, slashes spending for "medical services" by $12bn - or nearly 20% - an amount offset by a corresponding 50% boost in funding for veterans seeking healthcare in the private sector. The Department of Veterans Affairs employs approximately 482,000 people, including 500,000 workers at 170 hospitals and 1,200 local clinics in the nation's largest health care system. In all, there are 15.8 million veterans, which represents 6.1% of the civilian population 18 years and older. VA officials said the event was misguided. "Imagine how much better off veterans would be if VA's critics cared as much about fixing the department as they do about protecting its broken bureaucracy," VA press secretary Peter Kasperowicz said in a statement to UPI. "The Biden Administration's VA failed to address nearly all of the department's most serious problems, such as rising health care wait times, growing backlogs of veterans waiting for disability compensation and major issues with survivor benefits." Kasperowicz told UPI disability claims backlog is already down 25% since Trump took office on Jan. 20 after it increased 24% during the Biden administration. He said VA has opened 10 new healthcare clinics around the country, and Trump has proposed a 10% budget increase to $441.3 billion in fiscal year 2026. The administration's proposed budget for the VA reduces spending for "medical services" by $12 billion - or nearly 20% - which is offset by a 50% boost in funding for veterans seeking healthcare in the private sector. Kasperowicz said the "VA is accelerating the deployment of its integrated electronic health record system, after the program was nearly dormant for almost two years under the Biden Administration." The event was modeled after the Bonus Army protests of the 1930s, when veterans who served in World War I gathered in the nation's capital to demand extra pay denied after leaving the service. Irma Westmoreland, a registered nurse working at a VA hospital and the secretary-treasurer of National Nurses United, told the crowd in Washington: "It's important for every person to keep their job, from the engineering staff to the housekeeper to the dietary staff. When cuts are made, the nursing and medical staff will have to pick up all their work that needs to be done."

There's an Invisible Line That Animals Don't Cross. Here's Why.

The animal kingdoms of Asia and Australia are worlds apart, thanks to an invisible line that runs right between the two neighboring continents. Most wildlife never cross this imaginary boundary, not even birds. And so it has been for tens of millions of years, shaping animal evolution in different ways on each side. It all started about 30 million years ago, when the Australian tectonic plate bashed up against the Eurasian tectonic plate and created an archipelago, rerouting ocean currents and creating new regional climates. On one side of the map, in Indonesia and Malaysia, monkeys, apes, elephants, tigers, and rhinos evolved; while on the other side, in New Guinea and Australia, marsupials, monotremes, rodents, and cockatoos flourish. Very few species are abundant on both sides. The curious faunal divide is named Wallace's Line – after the naturalist Alfred Russel Wallace, who first noticed the stark difference in animal life (mostly mammals) while exploring the region in the mid-19th century. "We may consider it established that the Strait of Lombok [between Bali and Lombok] only 15 miles wide [24 kilometers] marks the limit and abruptly separates two of the great zoological regions of the globe," Wallace wrote. The naturalist later went on to independently develop a theory of evolution around the same time as Charles Darwin. The line he drew on a map more than a century ago is still considered a hypothetical evolutionary barrier, although debates continue as to its exact location and mechanisms. Generally speaking, Wallace's line separates a shelf of the Asian continent from a shelf of the Australian tectonic plate. It is a geological line, but it is also a climatic and biological one. Deep ocean channels like the Lombok Strait separate each shelf, which makes it difficult for animals to cross. Even when sea levels in the distant past were much lower than today, this chasm would have still existed. While Wallace's invisible line is most obvious when comparing mammals in Asia and Australia, it also exists for birds, reptiles, and other animals. Even creatures with wings don't typically make the trip across Wallace's line, and in the ocean, some types of fish and microbes show genetic differences on one side of the border compared to the other, indicating very little mixing between populations. Scientists have yet to figure out what invisible barriers are holding these species back. Habitat and climate, however, are probably factors accentuating the evolutionary divide. In 2023, an analysis of more than 20,000 vertebrate species found that Southeast Asian lineages evolved in a relatively tropical ancient environment that allowed them to spread out toward New Guinea on humid island "stepping stones". Wildlife on the Australian continental shelf, meanwhile, evolved in distinctly drier conditions, which dictated a different evolutionary path. This meant that Australian wildlife was at a disadvantage in the tropical islands nearer the equator. The more researchers study the Wallace line, however, the less clear it becomes about where the line should be drawn and how 'porous' the barrier might be – at least to some animals that can swim, float, or fly, like bats, beetles, monitor lizards, or macaques. Wallace's divide isn't an absolute border, but more of a gradient, scientists say. Even still, the blurry line helps us make sense of animal evolution for thousands of species. "Darwin's and Wallace's mental and actual maps were the table on which the evolutionary scheme was played out, comparable in importance to the geological time scale," argued science historian Jane Camerini in 1993 for the History of Science Society. What started as a single, roughly placed line, drawn more than a century ago, has now helped shape a bigger and more complicated picture of the natural world and its mysteries.
