PALO ALTO, Calif.--(BUSINESS WIRE)--Genascence Corporation ('Genascence'), a clinical-stage biotechnology company revolutionizing the treatment of prevalent musculoskeletal diseases with gene therapy, today announced positive 12-month safety and biomarker results from the Phase 1b DONATELLO clinical trial evaluating GNSC-001, a potential first-in-class gene therapy blocking interleukin 1 (IL-1) for the treatment of knee osteoarthritis (OA). Results from the 12-month analysis showed the study met the primary endpoint, demonstrating continued safety and tolerability across all doses tested, as well as the key secondary endpoint showing sustained IL-1Ra expression in synovial fluid, building on data reported through the six-month visit.
GNSC-001 is a genetic medicine – a recombinant adeno-associated viral vector expressing an optimized human interleukin-1 receptor antagonist (IL-1Ra), a naturally occurring protein that blocks IL-1 signaling. IL-1 is considered one of the key mediators involved in the pathogenesis of OA, causing inflammation, joint pain, and cartilage destruction. GNSC-001 is designed to offer long-term, sustained inhibition of IL-1 following a single intra-articular injection into the affected joint.
The U.S. Food and Drug Administration (FDA) granted GNSC-001 Fast Track designation in the fourth quarter of 2024. Genascence recently completed a successful meeting with the FDA on the design of the Phase 2b/3 clinical trial of GNSC-001 focused on clinical efficacy and plans to initiate the Phase 2b/3 study in 2026.

About the DONATELLO Clinical Trial
The DONATELLO Phase 1b clinical trial (NCT05835895) is a double-blind, placebo-controlled dose-ranging study designed to evaluate the safety, tolerability, and pharmacodynamics of a single intra-articular injection of GNSC-001 in patients with OA of the knee. The study enrolled 67 participants with OA at 10 centers across the U.S. The first five groups were randomized to receive GNSC-001 at doses of 110 12 vg or 110 13 vg, with or without a short course of oral steroids for immune-conditioning, or a placebo (saline) injection. The trial was expanded to enroll an additional, non-randomized arm. In this arm, pre-treatment synovial fluid sampling was required for entry, and subjects received 110 13 vg GNSC-001 with an abbreviated three-day course of oral steroids plus a local, intra-articular steroid injection.
Data and Safety Summary
The primary endpoints of the DONATELLO clinical trial are safety and tolerability. Through 12 months of follow-up, data show that GNSC-001 was well tolerated, with no treatment-emergent or treatment-related deaths, serious adverse events (SAEs), or adverse event (AE)-related withdrawals reported. The most common target knee AEs included arthralgia, joint swelling, and joint effusion.
The study's secondary endpoints include expression levels of interleukin-1 receptor antagonist (IL-1Ra) in synovial fluid at Month 12, as well as change from baseline to Month 12. Results revealed that mean expression of IL-1Ra reached target therapeutic levels in multiple arms of the study and remained above the target threshold throughout the 12-month follow up period. Immune-conditioning with a short course of steroids generally supported higher levels of prolonged IL-1Ra expression.
The DONATELLO clinical trial was supported by a $12 million award from the California Institute for Regenerative Medicine (CLIN2-14265).

U.S. FDA approves third indication of darolutamide for patients with advanced prostate cancer Darolutamide is the first and only in the U.S. and FDA-approved androgen receptor inhibitor (ARi) for the treatment of patients with hormone-sensitive prostate cancer (mHSPC), in combination with androgen deprivation therapy (ADT), with or without chemotherapy. This third approval is based on positive results from the pivotal Phase III ARANOTE trial and broadens the indication profile of darolutamide in mHSPC, enabling its use in combination with ADT, with or without chemotherapy (docetaxel). Darolutamide plus ADT reinforces the established safety and tolerability profile of darolutamide for prostate cancer patients across all approved indications. Orion's collaboration partner Bayer announced today that the U.S. Food and Drug Administration (FDA) has approved the oral androgen receptor inhibitor (ARi) darolutamide in combination with androgen deprivation therapy (ADT) for use in patients with metastatic castration-sensitive prostate cancer (mCSPC), which is also known as metastatic hormone-sensitive prostate cancer (mHSPC). The approval is based on positive results from the pivotal Phase III ARANOTE trial, which showed that darolutamide plus ADT significantly reduced the risk of radiological progression or death by 46% compared to placebo plus ADT (HR 0.54; 95% CI 0.41–0.71; P<0.0001) in patients with mHSPC. With this approval, darolutamide plus ADT is indicated in the U.S. for the treatment of adult patients with mHSPC either with or without docetaxel. In addition, darolutamide is approved for the treatment of adult patients with non-metastatic castration-resistant prostate cancer (nmCRPC) who are at high risk of developing metastatic disease. Prostate cancer is the second most common cancer in men and the fifth most common cause of cancer death in men worldwide.1 In 2022, an estimated 1.5 million men were diagnosed with prostate cancer, and about 397,000 died from the disease worldwide.1 Prostate cancer diagnoses are projected to increase to 2.9 million by 2040.2 Darolutamide, under the brand name Nubeqa®, is already approved in mHSPC in combination with ADT and docetaxel in over 85 markets around the world. It's also approved in combination with ADT for the treatment of patients with non-metastatic castration-resistant prostate cancer (nmCRPC) who are at high risk of developing metastatic disease in more than 85 countries around the world. An approval process in the EU for the treatment of mHSPC in combination with ADT (without docetaxel) is already underway by Bayer. Nubeqa achieved blockbuster status in September 2024, with annual sales reported by Bayer reaching EUR 1.52 billion for the full year of 2024. Darolutamide is developed jointly by Orion and Bayer. About the ARANOTE trial The ARANOTE trial is a randomized, double-blind, placebo-controlled Phase III study designed to assess the efficacy and safety of darolutamide plus ADT in patients with mHSPC. 669 patients were randomized 2:1 to receive 600 mg of darolutamide twice daily or matching placebo in addition to ADT. The primary endpoint of this study is rPFS, measured as time from randomization to date of first documented radiological progressive disease or death due to any cause, whichever occurs first. Secondary endpoints include overall survival (time to death from any cause), time to first castration resistant event, time to initiation of subsequent anti-cancer therapy, time to prostate-specific antigen (PSA) progression, PSA undetectable rates, time to pain progression, and safety assessments. Results from the Phase III ARANOTE trial presented at ESMO 2024 and published in The Journal of Clinical Oncology showed that darolutamide plus ADT significantly reduced the risk of radiological progression or death by 46% compared to placebo plus ADT (HR 0.54; 95% CI 0.41–0.71; P<0.0001), in patients with mHSPC. Consistent benefits in radiological progression-free survival (rPFS) were observed across prespecified subgroups, including patients with high-volume (HR 0.60, 95% CI: 0.44-0.80) and low-volume (HR 0.30, 95% CI: 0.15-0.60) mHSPC. The incidence of adverse events in the treatment group with darolutamide plus ADT in the ARANOTE study was comparable to placebo plus ADT. Darolutamide plus ADT was generally well tolerated and showed lower discontinuation rates due to adverse events compared to placebo plus ADT. About darolutamide Darolutamide is an oral ARi with a unique chemical structure that binds with high affinity to the androgen receptor and exhibits a strong antagonistic effect against the androgen receptor inhibiting the receptor function and the growth of prostate cancer cells. Additionally, preclinical models and neuroimaging data in healthy humans support darolutamide's low potential for blood-brain barrier penetration. Darolutamide (plus ADT or plus ADT and docetaxel) demonstrated a side effect profile in both registrational studies in mHSPC where the incidences of adverse events were roughly similar to the respective comparator arm. Darolutamide is a treatment option for doctors and patients, considering its tolerability and low risk of drug interaction. A robust clinical development program is underway investigating darolutamide across various stages of prostate cancer. The program includes the Phase III ARASTEP trial evaluating darolutamide plus ADT compared to ADT alone in hormone-sensitive high-risk biochemical recurrence (BCR) prostate cancer, who have no evidence of metastatic disease by conventional imaging and a positive PSMA PET/CT at baseline. Furthermore, darolutamide is also being investigated by Bayer in the collaborative Phase III DASL-HiCaP (ANZUP1801) trial led by the Australian and New Zealand Urogenital and Prostate Cancer Trials Group (ANZUP). The study evaluates darolutamide as an adjuvant treatment for localized prostate cancer with very high risk of recurrence. About metastatic hormone-sensitive prostate cancer At the time of diagnosis, most men have localized prostate cancer, meaning their cancer is confined to the prostate gland and can be treated with curative surgery or radiotherapy. mHSPC is a stage in the disease where the cancer has spread outside of the prostate to other parts of the body. Up to 10% of men will present with mHSPC when first diagnosed.3,4,5 For patients with mHSPC, ADT is the cornerstone of treatment, in combination with chemotherapy docetaxel and/or an androgen receptor inhibitor (ARi). Despite treatment, most men with mHSPC will eventually progress to castration-resistant prostate cancer (CRPC), a condition with limited survival. Fresh off of his recent departure from the White House, Tesla CEO Elon Musk gave CBS News an exclusive interview over the weekend. He sported the same blackened right eye he had in the White House when President Donald Trump thanked him for his work with the Department of Government Efficiency on his way out the door. Musk is returning to work full time at Tesla, leaving the side quest he has been on since January as the head of DOGE. Related: Elon Musk explains DOGE mission, takes shot at government While Trump was effusive in his praise of his efforts, Musk's time in Washington was, at times, incredibly rocky. While DOGE wasn't able to deliver on the cuts he initially promised, the work of non-government governmental agencies to root out fraud, waste, and abuse will continue as long as the president wishes it to. DOGE expects to cut at least $150 billion by 2026. A couple of Trump's policies run counter to Elon Musk's own best interests, and Musk has had to learn an old Washington, D.C. maxim about toeing the party line. Automotive and automotive parts tariffs directly affect Tesla's bottom line, and as a political neophyte, Musk originally addressed them the best way he knows how: on X. Musk went after Trump advisor Peter Navarro, who returned fire on cable television. Before the weekend was over, the two men had a full-blown war of words, with Trump taking Navarro's side. This was Musk's first public spat with the administration, and for a while, Musk didn't mention any of the administration's policies he'd disagreed with in the past. Although Musk entered the U.S. on a J-1 visa, which is often given to foreign nationals looking to further their studies in the States, he has been silent regarding Trump's attack on international students seeking to enter the country. But as his time in Washington has concluded, he has become slightly more vocal on some issues. Last week, he criticized the Republican spending bill making its way through Congress. I think a bill can be big or it can be beautiful, but I don't know if it could be both," Musk said. This weekend (June 1), fresh off of an exit interview in the Oval Office with the President, Musk was still sticking with the party line. "It's not like I agree with everything the administration does...I mean, I agree with much of what the administration does, but we have differences of opinion on the things that I don't entirely agree with," he said in an interview with CBS News. "But it's difficult for me to bring that up in an interview, because it creates a buildup of tension. So I'm stuck in a bind where I don't want to speak up against the administration, but I also don't want to take responsibility for everything the administration is doing." A month ago, Tesla reported one of its worst quarters on record. The company reported a 9% decline in first-quarter revenue to $19.3 billion, missing analyst estimates by $2 billion. Earnings of 27 cents per share fell short of Wall Street expectations by 34%. One of the issues during the period was cratering demand in Europe. While only a small percentage of sales were made in Europe, it was a challenging region in the quarter. Sales in Germany reportedly fell 62%, and numbers in Norway, the UK, and France weren't much better. Just this week, it was reported that Tesla's European sales dropped another 49% in April to 7,261, despite overall EV sales rising 34% in the region during the month, according to the European Automobile Manufacturers Association. But he also says that his activism has garnered him even more new fans from the opposite end of the political spectrum. He recently spoke at the Qatar Economic Forum, saying that any politically left-leaning buyers who abandoned the company have been replaced by people who align more with his politics. He punctuated his point by emphasizing that Tesla has no problem with demand.
