Clearing Brain Waste Dramatically Improves Memory in Aging Mice

As we get older, the essential cleaning processes that our brains need to keep functioning start to break down and fail. In new research, scientists have figured out how to boost waste removal cycles in the brains of mice – with dramatic effects on their memory.
Led by a team from Washington University in St. Louis, the research focuses on vessels around the brain called meningeal lymphatics, the brain cleaners in chief. These vessels are part of the larger lymphatic system in the body, responsible for waste disposal and helping the immune system.
The researchers used a targeted protein treatment on older mice to help these meningeal lymphatics grow and operate. In subsequent experiments, the treated mice were shown to have improved memory function compared to untreated animals.
There's a clear link to neurodegenerative conditions like Alzheimer's here, conditions in which aging brains lose memory functions and cognitive abilities. This new work could potentially offer some clues as to how to slow down or prevent dementia.
"A functioning lymphatic system is critical for brain health and memory," says neuroscientist Kyungdeok Kim, from Washington University in St. Louis.
"Therapies that support the health of the body's waste management system may have health benefits for a naturally aging brain."
The team also discovered that the protein interleukin 6 is used as a kind of distress signal by overwhelmed immune cells called microglia – a distress signal that's sent out when the brain's cleaning apparatus gets overwhelmed.
As well as boosting the mice's memory, the lymphatics treatment reduced interleukin 6 levels, restoring order to this part of the immune system and preventing some of the damage in the brain caused by stressed microglia.
Another important aspect of the research: the meningeal lymphatic vessels are just outside the brain, so they can be targeted without the complexity of having to get through the blood-brain barrier that helps keep the brain protected.
"The physical blood-brain barrier hinders the efficacy of therapies for neurological disorders," says neuroscientist Jonathan Kipnis, from Washington University in St. Louis.
"By targeting a network of vessels outside of the brain that is critical for brain health, we see cognitive improvements in mice, opening a window to develop more powerful therapies to prevent or delay cognitive decline."
These are all important insights into how normal brain communication networks get disrupted when toxic material isn't cleared away, and allowed to build up. It's not unlike trash building up on railway lines and stopping trains from traveling.
The new findings fit in neatly with previous research too, including a 2022 study in which mouse memories were boosted through injections of cerebrospinal fluid – the same fluid meningeal lymphatic vessels clean waste from.
"We may not be able to revive neurons, but we may be able to ensure their most optimal functioning through modulation of meningeal lymphatic vessels," says Kipnis.

Want to reduce your risk of dementia and Alzheimer's disease? Get on your bike and ride, a new study recommends. Biking regularly for transportation appears to lower risk of dementia by 19% and Alzheimer's by 22%, according to results published Tuesday in JAMA Network Open. The results also suggest that cycling might even help increase the size of a brain region important for memory, researchers noted. "Cycling is a moderate- to high-intensity workout, and also requires balance," said Dr. Liron Sinvani, director of geriatric services at Northwell Health in Manhasset, N.Y., who reviewed the findings. "It requires more complex brain function than walking, which is why maybe it was a better reducer of dementia risk." "It's not about just doing exercise and making that part of your routine, but thinking about the way you live your life," she added in a news release. "So instead of having to drive somewhere, taking that bike and using active travel modes to get around as part of your lifestyle becomes very important." For the study, researchers analyzed data on nearly 480,000 people participating in the UK Biobank, a long-term health study of people living in England, Scotland and Wales. As part of the study, participants noted the forms of transport they used most often to get about, not including travel to and from work. During an average follow-up of 13 years, more than 8,800 participants developed dementia and nearly 4,000 developed Alzheimer's. Results showed a lower risk for dementia and Alzheimer's among those who cycled or included cycling among other forms of travel like walking, driving or using public transportation. "Our findings suggest that promoting active travel strategies, particularly cycling, may be associated with lower dementia risk among middle-aged and older adults, which carries substantial public health benefits by encouraging accessible, sustainable practices for cognitive health preservation," concluded the research team led by Liangkai Chen, an associate professor at Tongji Medical College at the Huazhong University of Science and Technology in Wuhan, China. MRI brain scans showed that cycling also was associated with a larger hippocampus, a part of the brain involved in memory formation and learning, researchers noted. However, cycling's benefits appeared to extend mostly to people without a genetic risk for Alzheimer's, results showed. People without the APOE E4 genetic variant had a 26% lower risk of dementia and 25% lower risk of Alzheimer's. Results were not statistically significant among APOE E4 carriers, but indicated less protection from cycling. Interestingly, the results also indicate that driving offered some protection against dementia compared to taking the bus or subway. "Even when using inactive travel like car or public transportation, it seemed like driving had a little bit better impact [on brain health] than public transportation," Sinvani said in the news release. However, researchers noted that this observational study could not draw a direct cause-and-effect link between cycling and healthy brain aging. "What I tell my patients and family and friends whenever they ask me what they can do to reduce their risk of dementia, is whatever gets you outside doing stuff is what you should be doing," she said. "I think we see that it's not just the physical activity, but it's also in the balance, and it really engages different parts of your brain." If you're able to get on a bike, Sinvani said, do that. If you're aren't, use your feet. "I think if it's, 'should I walk or take a bike?' I would say take a bike, but if it's 'should I walk or just stay home?' I would say definitely walk," she said. U.S. senators grilled National Institutes of Health (NIH) director Jayanta Bhattacharya at a hearing on 10 June about how his professed support for science squares with unprecedented funding delays and research-grant terminations at the agency this year, as well as enormous cuts that have been proposed for its 2026 budget. What would normally be a routine hearing about government spending was anything but: hundreds of scientists and advocates for Alzheimer's disease research packed into a cramped room on Capitol Hill to denounce US President Donald Trump's 2026 budget request, which calls for cutting the NIH's budget by about 40% and collapsing its 27 institutes and centres into 8. Such a cut 'would stop critical Alzheimer's research in its tracks,' Tonya Maurer, an advocate for the Alzheimer's Association, a non-profit group based in Chicago, Illinois, told Nature at the hearing. Bhattacharya defended his leadership at the agency — the largest public funder of biomedical research in the world — noting that there is a 'need for reform at the NIH' and that to restore its reputation, the NIH 'cannot return to business as usual.' (The NIH has been accused by Trump and his Republican allies of funding 'woke' science and research on coronaviruses that they say could have sparked the COVID-19 pandemic.) To help fix the agency, Bhattacharya told the senators that he wants to focus on increasing reproducibility in biomedical research, upholding academic freedom and studying the cause of autism, which US health secretary Robert F. Kennedy Jr has pledged to find an answer to by September. Letters of dissent The hearing comes the day after more than 300 NIH staff members sent Bhattacharya a fiery letter decrying the mass termination of jobs at the agency and its cancellation of thousands of research projects on a growing list of topics that the Trump team has said are 'politicized', including those investigating the biology of COVID-19, the health of sexual and gender minorities (LGBT+) and reasons that people might be hesitant to receive a vaccine. 'We are compelled to speak up when our leadership prioritizes political momentum over human safety and faithful stewardship of public resources,' the staff members wrote. They named their letter the 'Bethesda Declaration,' after the Maryland community and Washington DC suburb where most of the NIH is located. The title also alludes to the 'Great Barrington Declaration', an open letter that Bhattacharya co-signed in October 2020 that argued against COVID-19 lockdowns except for the most vulnerable citizens, instead allowing for children and others to be infected so that 'herd immunity' could be reached ― a proposal that numerous scientists and NIH officials called dangerous at the time. At the hearing, Patty Murray, a Democratic senator from Washington, implored Bhattacharya to 'heed their warning,' and said that she expects that 'none of them face retaliation for raising those concerns.' Bhattacharya didn't respond to this comment at the hearing but said in a statement on 9 June that the Bethesda Declaration 'has some fundamental misconceptions about the policy directions the NIH has taken in recent months,' but that 'respectful dissent in science is productive.' Gavin Yamey, a global-health researcher at Duke University in Durham, North Carolina, who signed the latest declaration, said, 'he can talk about freedom, but his own staff are decrying his censorship. How he's actually acting and what he says are not one in the same.' Taking ownership Several senators, including Tammy Baldwin, a Democrat from Wisconsin, questioned who was in charge at the NIH, given reports that billionaire Elon Musk's Department of Government Efficiency ordered agency employees to cut hundreds of specific grants. 'The changes in priorities, the move away from politicized science, I've made those decisions,' Bhattacharya responded. The mass terminations of awards at institutions such as Harvard University in Cambridge, Massachusetts, 'that's joint with the administration', he said. (The Trump administration has alleged that universities such as Harvard have allowed discrimination, including antisemitism, on their campuses, and has cut or frozen research funding as a result.) The drastic 40% cut to the NIH's budget proposed for the fiscal year 2026 is not yet set in stone: the US Congress has the ultimate say over government spending, and during Trump's first presidency, when he proposed a huge cut to the biomedical agency in 2017, it instead approved a slight increase. Nevertheless, the composition of the body has changed significantly since then — far more of its members are now loyal to Trump. Comments made at the hearing by the senators weren't entirely divided down party lines. Susan Collins, a Republican from Maine who voted to confirm both Bhattacharya and RFK Jr, said she was disturbed by the budget proposal. 'It would undo years of congressional investment in the NIH,' she said.

CHARLESTON, S.C. (WCBD) – About 112,500 people in South Carolina are living with Alzheimer's disease, according to the Alzheimer's Association. To advance research, more than 50,000 participants are needed in clinical trials examining potential new treatments. Hundreds of thousands are needed in broader dementia research studies. Alzheimer's Association TrialMatch is a free, easy-to-use service that connects interested individuals living with Alzheimer's and other dementias, caregivers, and healthy volunteers with appropriate trials. Experts believe there isn't a single cause of Alzheimer's, and research shows that adopting healthy habits can improve brain health and reduce the risk of cognitive decline, according to the Alzheimer's Association. They suggest proactively taking care of brain health, which includes challenging the mind, getting regular exercise, protecting the head from injuries, living a smoke-free life, controlling blood pressure, eating healthier foods, and getting quality sleep. 'We want people to know there are steps they can take to keep their brains healthy. We also want to encourage anyone experiencing memory or thinking problems to talk to their doctor. There are many possible causes — and if it is Alzheimer's disease, there are numerous benefits to getting a timely diagnosis,' said Ashton Houghton, executive director of Alzheimer's Association, South Carolina chapter. To get a timely diagnosis, the Alzheimer's Association lists some of the early signs and symptoms of Alzheimer's. Some include memory loss that disrupts daily life, challenges in planning or solving problems, difficulty completing familiar tasks, confusion with time or place, trouble understanding visual images and spatial relationships, new problems with words in speaking or writing, changes in mood and personality. Those interested can volunteer with the Alzheimer's Association to help those in the community take steps to reduce their dementia risk and recognize the signs of Alzheimer's, and can find volunteer opportunities on their website.
