ASCO25: J&J's trispecific antibody shows 100% response in Phase I multiple myeloma study
Johnson & Johnson (J&J) has unveiled first-in-human data on its trispecific antibody that demonstrates a 100% response rate in a small group of patients with relapsed or refractory multiple myeloma (r/r MM). The initial data from the Phase I trial (NCT05652335) of JNJ-5322 shows that the trispecific antibody led to a 100% overall response rate (ORR) in 27 patients who were naïve to anti-BCMA/-GPRC5D therapies at a median follow-up of 8.2 months. The patients were treated with the proposed Phase II dose of 100mg four times weekly (Q4W). JNJ-5322 simultaneously targets BCMA (B-cell maturation antigen), GPRC5D (G protein coupled receptor family C group 5 member D), and CD3 (cluster of differentiation 3). The drug engages both malignant B cells and T cells, inducing anti-cancer cytotoxicity and T-cell activation, while overcoming tumour heterogeneity. All responders remain in response as of a median 8.5 month follow-up, with a median time to first response of 1.2 months. The Phase I trial, initiated in November 2022, has enrolled 126 patients as of 15 January 2025 with r/r MM previously exposed to proteosome inhibitors, immunomodulatory drugs, and anti-CD3 monoclonal antibodies (mAb). After dose escalation, 36 patients were administered 100mg JNJ-5322 subcutaneously Q4W as the putative recommended Phase II dose. The data was presented at the 2025 American Society of Clinical Oncology (ASCO) conference held in Chicago, Illinois, on 3 June. However, 99% of the collective cohort experienced at least one adverse event (AE), most commonly cytokine release syndrome (59%), while 75% reported infection. In the case of five patients, toxicities proved dose-limiting, and four patients died as a result of AEs. J&J's candidate was granted an orphan drug status to treat r/r MM by the US Food and Drug Administration (FDA) on 27 November 2024. The data presented at ASCO 2025 represent the first clinical insight into the drug's potential. Abstract authors stated, 'first data with JN-5322 suggest a paradigm shift, offering ORRs similar to CAR-Ts (chimeric antigen receptor-Ts) but as an off-the-shelf therapy.' JN-5322 is developed using J&J's antibody discovery platform OmniAb technology. J&J licenced the technology to develop antibodies for multiple myeloma through a partnership with OmniAb's parent company Ligand Pharmaceuticals, which was spun out from Ligand in 2022. "ASCO25: J&J's trispecific antibody shows 100% response in Phase I multiple myeloma study" was originally created and published by Pharmaceutical Technology, a GlobalData owned brand. Report: Nets may trade Cam Johnson and 19th pick to get Lottery pick
Report: Nets may trade Cam Johnson and 19th pick to get Lottery pick The Brooklyn Nets came away from the 2025 NBA Draft Lottery hoping to get the first overall pick that would allow them to take Duke forward Cooper Flagg to begin their rebuild with a hot start. Unfortunately for Brooklyn, they came away from the Lottery with the eighth overall pick, but it looks like they are trying to take the Draft into their own hands. Per NetsDaily on Wednesday via X, the Nets "may want to move up from #19 to the lottery, offering CamJ and #19 to Raptors or Rockets for either Raps 9th or Rockets 10th plus a bad contract." Since the Lottery passed, there has been consistent reporting alluding to the possibility of Brooklyn trying to shake things up by either using their best pick to move up or by getting another Lottery pick. "That would give Nets two picks in top 10 (presupposing neither team would need picks for Giannis trade.)," NetsDaily's post continued. Nets forward Cam Johnson has been consistently brought up in trade rumors ever since last offseason when the Nets began their rebuild by trading Mikal Bridges to the New York Knicks in exchange for draft picks, but it seems like these reports about Brooklyn and the Draft are increasing in frequency. Brian Lewis of the New York Post reported in May that "there is a sentiment around the league the Nets could look to land a second lottery pick by consolidating and packaging some combination of Johnson, those picks at the end of the first round and/or future draft capital." This report followed one by NBA insider Jake Fischer in which the belief among rival teams is that the Nets are going to attempt to trade up in the Draft. When it comes to the term "bad contract," it will be interesting to see what that means for either the Toronto Raptors, who have the ninth overall pick, or the Houston Rockets, who have the 10th overall pick. Johnson, who is slated to make $20.5 million for the 2025-26 season, could potentially get Brooklyn someone like center Jakob Poeltl or forward RJ Barrett from the Raptors or players like forwards Jabari Smith Jr. or Dillon Brooks from the Rockets.

Mike Johnson Says Trump Was ‘Not Delighted' by Musk Criticism Over His Big Beautiful Bill
Republican leaders championed the 'One Big Beautiful Bill' backed by President Trump, lauded Elon Musk for his work leading the Department of Government Efficiency and blasted Democrats for allowing 'tens of millions' of illegal aliens — like the attacker in Boulder, Colorado — into the country during a Wednesday press conference. House Speaker Mike Johnson praised Musk during most of his time speaking, saying the Tesla boss had done 'extraordinary' work finding needless government spending while leading DOGE. Musk, Johnson noted, was inside the 'belly of the beast' that is the U.S. government and helped shine a light on waste that was 'unknowable until now.' Still, Johnson also said President Trump was 'not delighted that Elon did a 180' on the bill. His comments came after Musk on Tuesday blasted the 'pork-filled' bill as a 'disgusting abomination.' The speaker's comments also stand out, considering White House Press Secretary Karoline Leavitt said on Tuesday the president had shrugged off Musk's criticism of the bill. 'The president already knows where Elon Musk stood on this bill, it doesn't change the president's opinion,' she said. Johnson on Wednesday said DOGE was 'the beginning of making government more efficient' and that it is 'going to take us a while' to make all the cuts Musk wants to see, considering it took decades for the deficit to reach the point it is at. He also said he called Musk on Tuesday night and was unable to reach him, but was hoping to the two would speak today. The 'One Big Beautiful Bill' passed the House last month and has moved to the Senate for consideration. Also on Wednesday, an analysis from the nonpartisan Congressional Budget Office noted the same bill could add $2.4 trillion to the nation's debt. GOP leaders then ripped Democrats and the Biden Administration for doing nothing to curb illegal immigration at several points on Wednesday morning. Rep. Lisa McClain of Michigan said the Biden Administration's 'open border policies' paved the way for the attack against Jews in Colorado last weekend while rallying for the release of hostages held by Hamas. The suspect, Mohamed Sabry Soliman, was seen on video yelling 'we have to end Zionists' in a video that went viral on X. The Egyptian national was in the country illegally, Fox News reported; Soliman entered the U.S. in 2022 and his work permit to remain in the country expired in March. 'I mean, honestly, what's next? Is a Democrat going to fly to Boulder to visit the illegal who burned Jews alive? Is that what's next for the Democratic Party?' McClain said on Wednesday. 'The contrast between Republicans and Democrats couldn't be more clear.' Shen also said more terrorist attacks could occur because of the 'tens of millions' of people who are in the country illegally thanks to Biden. Johnson then said one main reason the bill needs to pass the Senate is because it 'includes $45 billion to expand ICE detention capacity.'
