ERA Congress: Long-term data show sustained efficacy and safety of zigakibart in patients with IgA nephropathy
VIENNA, June 5, 2025 /PRNewswire/ -- New 100-week data from the ongoing Phase 1/2 study of zigakibart, an investigational anti-APRIL monoclonal antibody, reinforce its potential as a disease-modifying treatment for IgA nephropathy (IgAN). Findings presented today at the 62 nd ERA Congress demonstrate sustained proteinuria remission, stable kidney function, and a reassuring safety profile. IgAN is the most common form of glomerular disease worldwide and a frequent cause of chronic kidney disease. Its pathogenesis is marked by inflammation and progressive kidney damage, which can lead to kidney failure. Many patients are unaware they have the condition until significant kidney damage has occurred, and 50% of IgAN patients will ultimately develop kidney failure. By targeting the APRIL pathway and reducing production of pathogenic galactose-debecause ofgA1), zigakibart addresses a key driver of disease progression. "Zigakibart is designed to intercept the initiating factor in IgAN pathogenesis, offering a new approach that may halt or significantly delay progression", explained lead investigator Professor Jonathan Barratt. The ADU-CL-19 trial included 40 adults with biopsy-confirmed IgAN and persistent proteinuria despite stable supportive therapy. Patients received zigakibart every two weeks via intravenous infusion or subcutaneous injection, in addition to maximally tolerated renin–angiotensin system inhibitors (RASi) unless RASi-intolerant – demonstrating efficacy beyond standard care. At Week 100, proteinuria was reduced by 60% from baseline. Over half of patients (55%) reached <500 mg/24 h, and 31% achieved <300 mg/24 h, indicating deeper remission. Estimated glomerular filtration rate (eGFR) remained stable across subgroups. "The consistency of eGFR stabilisation over 100 weeks, even across proteinuria response groups, is particularly encouraging," said Prof. Barratt. Treatment also led to sustained reductions in serum immunoglobulins, including a 74% drop in IgA and pathogenic Gd-IgA1, consistent with APRIL pathway inhibition. Zigakibart was well tolerated throughout. Most adverse events were mild or moderate, with no treatment-related serious infections or discontinuations. Infections were the most common AEs; the study coincided with a high prevalence of COVID-19. This is the longest duration of eGFR stabilisation reported for an anti-APRIL agent in IgAN. "These long-term results build confidence in zigakibart as a potential cornerstone therapy for IgAN," said Prof. Barratt. "We're excited to see how the upcoming Phase 3 trials will further define its role." The global Phase 3 BEYOND study is now evaluating zigakibart in a broader population, with primary proteinuria endpoints at 40 weeks and long-term kidney function through 104 weeks.

India's Chess Star D Gukesh Confident Of Meeting Expectations
Star Indian chess player D Gukesh is embracing the "new level" of competition where he is confident of handling the challenges and "meeting the expectations". Gukesh became the youngest chess world champion when he defeated Ding Liren of China last year in Singapore at just 18 years of age. Set to compete in the elite Norway Chess tournament beginning on Monday, the Chennai youngster said the only thing he needed to gain more success was to keep random thoughts at bay and just concentrate on the game. "It is a new level (for me). But when I see it as a challenge to prove to myself that I can handle this, even though the expectations are high, I can meet them if I keep working hard and doing my best," said the Indian Grandmaster. He said that playing five hours of classical chess can make one's mind wander, but the key to winning those tiring games is to not lose focus. "In a classical game, you end up playing five hours and those are the times you cannot fully be thinking about the game. I think there are random thoughts that come to our mind but the important thing is to stay in the game and come back to those thoughts later. "This is the purpose of the note that you're making. And you can't afford to allow weak moments," said Gukesh, who along with Arjun Erigaisi will compete against the likes of world No.1 Magnus Carlsen and No.2 Hikaru Nakamura in the USD USD 1,62,681 tournament, once termed as "the Wimbledon of Chess" by the legendary Garry Kasparov. The 12-day tournament, which concludes on June 6, will see Gukesh compete against Carlsen in his own backyard, which will be the highlight of the event given that the Indian is the world champion and the Norwegian is the world No. 1. This will be the first time since being crowned world champion that the 18-year-old will face off against Carlsen in the classical format. "I feel great being here (in Stavanger). I've had good memories from Norway. I am sure playing Magnus here in a classical game will be a fun challenge. Also, for the spectators, it is about the world champion versus the No.1 player in the world." The last time Gukesh competed in Norway Chess was in 2023 where he finished third. He had secured the crucial FIDE Circuit points needed to qualify for the 2024 Candidates Tournament. That achievement made him the third-youngest player in history to qualify for a Candidates tournament, following in the footsteps of Bobby Fischer and Carlsen. Gukesh hoped the games against Carlsen would live up to expectations, given the hype built around them. "It surely sounds very exciting so it's surely something I am very happy to give, myself and the chess fans. And I hope it will be a treat," added Gukesh.

Sol de Janeiro's 'best fragrance yet' is out in time for summer
'I love this!! This is the smell of summer, it's so floral and fruity. I recommend this for anyone who loves beach days!' Sol de Janeiro is known for sweet scents that get heads turning, with Cheirosa '62 having proudly been the scent of the summer year after year, but they've got a new limited edition release that might be the brand's best fragrance yet. Água Mística is part of the latest release from the brand made up of three scents. All three share Mistica in the name but it's one that's standing out to us above all. Água Mística opens up with a refreshing trio of tangerine, neroli and pineapple before deepening to a heart of jasmine and peony. It's got a base reminiscent of the beach with salt, skin and white cedar extract which gives an overall juicy and moreish mist. Our perfume expert Jada Jones tried it and said: "There are a few brands that come with a level of hype that can be hard to live up to and this is definitely one of them but Água Mística deserves more. I wasn't expecting this to be my favourite of the trio but as soon as I smelled it, it was clear this is Sol de Janeiro's best fragrance yet. Ohio legislators want to increase penalties for passing stopped school buses
COLUMBUS, Ohio (WCMH) — Ohio legislators are trying to increase penalties and awareness when it comes to school bus safety. State Sen. Theresa Gavarone (R-Bowling Green) introduced Senate Bill 62, the School Bus Safety Act. State Reps. Bernard Willis (R-Springfield) and Cecil Thomas (D-Cincinnati) introduced House Bill 3 under the same title. Both bills would increase penalties for passing school buses illegally. See previous coverage of lawmakers hoping to increase school bus safety in the video player above. Under the S.B. 62, illegally passing a stopped school bus would spur a civil penalty of $300, $50 of which would go to a newly created school bus safety fund. The remaining $250 would be paid to the organization in charge of the bus, such as a local school district, which would have to use the funds for bus safety initiatives. Ohio lawmakers want to allow parents to claim conceived children on income taxes H.B. 3 goes even further, designating passing a stopped school bus as an unclassified misdemeanor. This would raise fines for violations to a minimum of $250 and a maximum of $1,000. Repeat offenders could be eligible for fines between $350 and $2,000. Currently, state law permits fines up to $500. All fines collected would go to the school bus safety fund. Both bills would establish the school bus safety fund within the state treasury, which would award grants to school districts to improve bus safety further. The fund would also be used by the Department of Education and Workforce to promote school bus safety awareness. Both S.B. 62 and H.B. 3 would also designate August as 'School Bus Safety Awareness Month.' The bills state funds generated by this legislation could go to eligible school bus safety features. Current Ohio law says nothing about external bus cameras, but S.B. 62 would encourage their use. It does clarify that districts using camera technology would have to use them across all routes. H.B. 3 states that repeat offenders could also face class five suspensions of their drivers licenses, which could range from six months to three years. S.B. 62 had its first Senate committee hearing on Feb. 19, during which Gavarone addressed the Judiciary Committee and reminded members that the State Highway Patrol issued more than 16,000 citations for school bus and zone violations between 2018 and August 2023.
