Fermenting legumes, pulses can boost antioxidant, antidiabetic properties: Study

New Delhi: Love to consume legumes and pulses? Fermenting them can help raise their antioxidant levels, as well as raise their ability to fight diabetes, according to a study.
Food scientists at the University of Illinois Urbana-Champaign, US, identified the optimal fermentation conditions for pulses --the dried edible seeds of legumes -- that increased their antioxidant and antidiabetic properties and their soluble protein content.
In the study, the team fermented pulses obtained from varying concentrations of black beans, black-eyed peas, green split peas, red lentils, and pinto bean flour. The fermentation was done using the bacteria Lactiplantibacillus plantarum 299v as microorganisms.
The results showed that antioxidant activity increased by up to 83 per cent and their capacity to regulate Type 2 diabetes markers increased by 70 per cent. Fermentation also increased the amount of soluble protein in these food items.
Red lentils and green split peas showed the greatest improvements in antioxidant scavenging activity and protein solubility. These also demonstrated the greatest modulation of two enzymes that improve insulin metabolism.
Lp299v is a probiotic strain "containing microorganisms that support gut health," explained first author Andrea Jimena Valdés-Alvarado, a graduate student at the varsity.
"After fermentation, it remains in the digestive process. It will not only preserve the fermented product that you're consuming, but it will also produce these peptides or amino acids that are more easily absorbed than the intact protein in the pulses," Valdés-Alvarado said.
Further, Lp299v is also known to reduce inflammation, boost immunity, and enhance iron absorption, the team said, in the paper published in the journal Antioxidants.
"These pulses contain between 18 per cent and 25 per cent good-quality proteins that can be used alone or as ingredients in other food products. We need to find adequate processing conditions and motivate the food industry to use them in dairy beverages or meat substitutes," said Elvira Gonzalez de Mejia, Professor of food science from the varsity.
The researchers stressed the need to explore the sustainability of plant-based diets amid global food insecurity, shortages of natural resources, and climate change.

Nanoplastics can make E. coli infections worse: study

Nanoplastics are bad news even though each one is only as small as a smoke particle. Research has shown that micro- and nano-plastics are present at the top of the tallest mountains, at the bottom of the deepest trenches, and even in our bloodstream, tissues, and in newborns as well. Their build-up has many toxic effects, including damage to cells and the genetic material inside. Now, a new study from researchers at the University of Illinois, Urbana-Champaign, has revealed that nanoplastics aren't just risky on their own: they also increase the risk from pathogens. The researchers found that nanoplastics with positively charged surfaces could make Escherichia coli, a foodborne pathogen, more virulent. The findings were published in the Journal of Nanobiotechnology. How bacteria work E. coli bacteria have a negative charge on their outer membranes. This can attract positively charged nanoplastics and increase the stress on the bacterium, making it produce more Shiga-like toxins. These are the proteins E. coli secrete and which cause disease. In older studies, scientists have looked at the impact of charged surfaces on non-pathogenic bacteria. The new study examined how charged surfaces affected a particular strain of E. coli. Similarly, while previous studies focused primarily on free-floating cells, the new one studied both free-floating cells and biofilms — a community of bacteria sticking to each other over a surface. Biofilms are hotspots where bacteria exchange genetic material with their neighbours. The research process The team members used a pathogenic E. coli strain that was resistant to the antibiotic rifampicin. They cultured the strain on agar plates and a nutrient-rich liquid medium called LB broth. Then they exposed the bacteria to polystyrene-based nanoplastics with three kinds of charges: positive, negative, and neutral. The team also cultured a second set of E. coli cells in LB broth for use as a control. This group wasn't exposed to nanoparticles. Finally, the researchers observed the growth of the free-floating cells and the biofilm at seven- and 15-day intervals, and used different tests to quantify the growth. For the study's purposes, the researchers used an environmental scanning electron microscope (ESEM) to track interactions between nanoparticles and free-floating cells. Unlike microscopy techniques that require samples to be prepared in protracted processes, ESEM can image 'wet' samples without too much preparation. It also operates in a low-vacuum or controlled gas atmosphere that prevents natural fibres and cellulose from becoming charged. The scientists conducted two tests to measure the concentration of carbohydrates and proteins in the biofilm. They performed a catalase assay to determine the cells' response to oxidative stress. E. coli produce the catalase enzyme to protect themselves against oxidative stress. Higher stress would mean more catalase activity. The team also extracted RNA from the bacteria for study, because gene transfers take place across biofilms. Changes in RNA may indicate such transfers have happened. What the study found The investigations revealed that nanoplastics with surface charges did add to the toxicity of bacterial cells. At first, charged nanoplastics kept cells from growing. But some cells eventually overcame the stress and began to grow. The development and viability of cells exposed to charged nanoplastics were different from those of cells that weren't. Changes in the bacterial RNA suggested genes were changing to ensure the E. coli survived better and became more able to cause disease. The study said both positively and negatively charged nanoplastics caused these changes. Beena D.B., assistant professor of biology at Azim Premji University's School of Arts and Sciences, has conducted preliminary work on the effect of microplastics on soil microbes and said her team had observed similar results. She cautioned that microplastic-induced biofilms are a significant health hazard and that increasing horizontal gene transfer between microbes raised the risk of them developing antibiotic resistance. Study shows how blood sugar response to carbs can signal prediabetes risk

New Delhi: Blood sugar responses after food, especially carbohydrates, may indicate metabolic health status as well as signal the risk of prediabetes, according to a study. Researchers from Stanford University in the US explained that the differences in blood sugar response patterns among individuals were associated with specific metabolic conditions such as insulin resistance or beta cell dysfunction -- both can lead to diabetes. The findings, published in Nature Medicine, suggest that this variability in blood sugar response could lead to personalised prevention and treatment strategies for prediabetes and diabetes. "This study suggests that not only are there subtypes within prediabetes, but also that your subtype could determine the foods you should and should not eat," said Michael Snyder, Professor in Genetics at Stanford Medicine. In the study, 55 participants without a history of Type 2 diabetes underwent metabolic testing for insulin resistance and beta cell dysfunction. The participants were also subjected to multi-omics profiling, which included tests for triglyceride levels, metabolites in the plasma of the blood, measures of liver function, and gut microbiome data. Just under half of the participants, 26 in total, had prediabetes. The results showed that many participants had a blood glucose spike after eating rice or grapes, regardless of their metabolic health status. The blood glucose responses to foods containing the highest amounts of resistant starch -- potatoes and pasta -- varied depending on the participants' metabolic dysfunction. Participants whose blood sugar spiked after eating bread were more likely to have hypertension or high blood pressure. The comparison of the blood glucose responses to potatoes versus grapes was associated with insulin resistance, suggesting that this ratio could serve as a real-world biomarker for insulin resistance in the future. "Such a biomarker would be useful because insulin resistance is amenable to lifestyle and medication interventions that can reduce the risk for diabetes in high-risk individuals. At present there is no easy way to diagnose it in the clinic," said Tracey McLaughlin, professor of endocrinology at the varsity. The researchers also found that eating fibre or protein before the rice lowered the glucose spike, and eating fat before the rice delayed the peak of the spike. However, these changes in blood glucose response occurred only in the metabolically healthy participants who were insulin-sensitive or had normal beta cell function, said the team calling for further research.
