"The last stage of the Giro on Sunday will be my last race for Israel Premier Tech," the Dane said in a video released by his team.
"I've had a fantastic career that has given me much more than I ever dreamed of. But all good things come to an end and it's time for me to spend more time with my family and make way for the young riders."
After beginning his professional career on mountain bikes, Fuglsang made a successful switch to the road.
He won the Criterium in 2017 and 2019 when he also won his first Monument, the Liege-Bastogne-Liege. The following year he added the Tour of Lombardy.
In recent years Fuglsang has played the role of road captain for riders such as Canadian Derek Gee, fourth overall in the Giro at the start of stage 19 on Friday.
"When I extended my contract with the team, we agreed on a seven-month contract to support our general classification project with Derek Gee," he explained.
"It's been a fantastic adventure, and I'm happy to finish at a level where I can still play a part and give something back to the team.
While Fuglsang is closing his career with Israel Premier Tech, Fuglsang said he might make a final farewell by riding one last time in the Tour of Denmark, which he has won three times, in August.

Simon Yates on verge of Giro triumph after epic stage 20 effort

Briton Yates will claim a career-defining triumph on Sunday barring disaster after overhauling Del Toro with a brave solo attack on the key Colle delle Finistre climb and finishing the stage third behind Harper and Alessandro Verre. The Visma-Lease a Bike rider now only has to cross the line intact in Rome to win his second Grand Tour, with the final stage largely a procession around the Italian capital and Vatican City. Yates is almost four minutes clear of Del Toro heading into the finale after redemption on the Finistre, the highest climb of this year's Giro and a long 18.5km slog up to 2,178 metres above sea level. He lost the pink jersey the last time the Finistre featured in the Giro, in the 19th stage of the 2018 edition when he ended up finishing over an hour behind eventual victor Chris Froome in the overall standings. Yates trailed both Del Toro and Richard Carapaz at the start of the stage after losing 22 seconds on Friday, but he delivered himself victory after pulling away on the key climb. UAE rider Del Toro spent most of the climb just staying on Carapaz's wheel and ended up unable to bridge the gap to Yates, handing over the pink jersey he had held since the ninth stage. The final stage will honour the late Pope Francis with an unprecedented ride through the Vatican, and new Pope Leo XIV will greet the cyclists as they pass through.

Prodhomme wins Giro stage as Del Toro holds lead

Overall leader Isaac del Toro of Mexico remains 43 seconds ahead of Richard Carapaz of Ecuador while Briton Simon Yates is third at 1min 21s after losing 20sec on the day. Former Giro champion Carapaz was satisfied with his day's work. 'I think we were all very tired, and in the end I just had to give it a try. But I think it was a good stage. I think we can make it with Del Toro so I've got to be happy with my performance.' Prodhomme's face was twisted with the effort as he broke away from a large escape group. "I've waited a long time for a win. I won my first race a week ago and now I win here in the Giro d'Italia." On a scenic route with the temperature tipping 30C (86f) the peloton set a medium pace, keeping their powder dry for a title showdown at Sestrieres ski resort Saturday. Stage 20 ends with a gruelling 20km climb up the Colle delle Finestre -- the moment when the winner is likely to emerge.
