Tens of thousands of 'Clunatics' - the nickname for fans of the show and star Martin Clunes - flock to the peaceful fishing village annually.
During the summer months, the town's narrow winding lanes are so crowded that locals steer clear of the historic harbour.
Fans from the UK, USA, Australia and mainland Europe has turned the once quiet village into an international tourism hub.
To some in Port Isaac, this has brought new opportunities including walking tours, cafes and merchandise shops.
Rubber-necking tourists reportedly flock to view iconic landmarks such as the home of Martin Clunes' character Dr Martin Ellingham, and Aunty Ruth's cottage above Squeezy Belly Alley.
But locals have suggested that their home has become a "victim of its popularity".
Over the past two decades, house prices in the area - renamed Port Wenn on the show - have quadrupled.
Mum-of-two Anne Wilce, 44, a lifelong Port Isaac resident, told how it had become "unrecognisable".
She fumed: 'It is horrific, there is no affordable housing available for anybody now.
'My sons are 19 and 22 and they won't ever be able to afford to buy a house at this rate.
'Without more affordable housing in the village the younger generation will move away because there is nothing here for them.
'My house is 30 years old and it's part of the last affordable housing that has been built here.
Martin Clunes announces sad death of Doc Martin co-star live on air - two years after hit ITV show ended
'I don't go down to the village centre, I have no reason to because it's all second homes.
"Even when something does come up for sale it's ridiculously priced - nobody around here can afford it.
'Doc Martin brought a whole load of money into the village which has created jobs for people and helped this community.
"But it is also a victim of its popularity and it doesn't feel like our village any more.'
Tourism has long since overtaken fishing as Port Isaac's main industry.
Local businesses have thrived as a result of the town's worldwide fame.

John Cleave, who was brought up in the village and runs the Boathouse fashion and gift shop, agrees that the show has changed village life.
"The locals talk about BDM – that's Before Doc Martin.
"It was always favoured by comparatively few people who simply saw it as a beautiful, typical Cornish fishing village.
"As a business you could close your doors from October to Easter and it wouldn't make a lot of difference.
"Thanks to Doc Martin we now have all-year-round trade."
The British comedy-drama first aired in 2004.
On the outer edge of the village - next to Port Isaac's real GP surgery - sits a small housing estate containing some of the village's only affordable homes.
They were constructed around 30 years ago and according to residents they were the last affordable homes built in the village.
There are now just two active fishing boats remaining in the village - compared with 174 holiday lets on Airbnb alone.
In the past year, semi-detached properties in Port Isaac sold for an average of £810,000, with terraced properties fetching £616,250, according to Rightmove.
Shop worker Elisha Scott, 19, and her family have lived in the area for 10 years.
She said: 'Nobody my age can afford to rent let alone buy a house here, all my friends still live with their parents.
'I'm not really thinking about house prices though because I don't think I'll stay here for a long time, there just aren't the job opportunities.
'Doc Martin definitely made a difference to Port Isaac, there are always fans of the show looking around the village.
"What has come with that is a lot of people buying second homes here which has made it too expensive for locals.'
Earlier this month a new plan was unveiled to build more affordable homes for local people.
Tre Haven Community Land Trust (CLT) has started negotiations with Cornwall Council to purchase land for 25 affordable housing developments.
The proposal has support from the area's Lib Dem MP Ben Maguire and local celebrity chef Nathan Outlaw.
But critics point out the site sits on a main road several miles from the village - with almost no public transport links to shops or schools.
Unemployed Ciaran Patching, 20, said: 'It took me two years of searching and I feel incredibly lucky to be here, before that I was living with my mum in the town.
'I would love to be able to buy my own house here but it doesn't feel like that will ever be an option at the moment.
'Port Isaac is definitely very popular in summer but in winter it can be dead.
"The popularity of Doc Martin has helped a lot of businesses around here but it has made it hard for year round residents.'

Emmerdale's Bear Wolf in dramatic new storyline as Lisa Riley reveals shock developments for the Dingles

EMMERDALE'S Bear Wolf will find himself in a dramatic new storyline, Lisa Riley has revealed. In recent episodes of the hit ITV show, Lisa's character Mandy Dingle and partner Paddy, played by Dominic Brunt, have been voicing their concerns about Paddy's dad, Bear. 4 4 4 The couple have tried to help him - from arranging a doctor's appointment to giving him an old motorbike to restore, but he simply puts his strange behaviour down to 'being a pain in the bum.' Now, after an incident at the garage that saw Bear getting very frustrated with the bike, and an altercation with Kammy Hadiq, Emmerdale fans have begun to question whether Bear has a secret health condition. Dropping some hints about the storyline, Lisa Riley, 48, spoke to The Sun and said: 'Whatever's going on with Bear, it'll bring Mandy and Paddy closer together. 'Next week, people are going to see Paddy and Mandy getting their new home and it's all very exciting. And as new homeowners, they want to take advantage of it because they love each other and can't keep their hands off each other. 'It's their passion pad. They like to call it Paddy Towers. But Paddy is also very aware about mental health issues, so what does that mean for Bear moving forward? 'Is he a grumpy old man? Is he suffering from mental health issues? Will he have something severe as we go on? Of course, it's going to go off.' The talented actress, who will celebrate a huge 30 years on the show next month, also spoke about the Dingles and serial killer John Sugden being part of the family. 'Mandy loves John. She's very flirty with him, and she thinks he's great for Aaron. Lisa Riley opens up on the grief of losing her mum 'She thinks that he'll look after Aaron, yeah, and obviously with the viewers, it's so hilarious, honestly. 'When I'm in Waitrose, people go, 'Mandy, Mandy, be careful of that John' and I'm going, 'yeah, OK,' but how do you say it in a non-patronising way? 'I know as a viewer, but obviously Mandy loves him and doesn't see anything wrong with him and has no idea. And if Mandy does find out in so many months to come, oh God help him. 'I mean, her brick would come out - she has a brick in her bag, and you'd see the crazy side of Mandy if she had any inkling that John had something to do with Nate's death.' Lisa added: 'She's the Queen of the family, and you don't cross the Queen.' Away from her life portraying Mandy Dingle on hit soap Emmerdale, Lisa has been supporting Sue Ryder with their latest campaign. Lisa was incredibly close to her mum Cath, but sadly lost her to cancer in 2012. Now, Lisa has joined forces with their Back for a Moment campaign, to help bereaved people get the support they need. And in doing so, the star chatted to The Sun about her mum, and how she sometimes finds herself hiding behind her character Mandy, to disguise the pain of losing her. But Lisa says it's her Emmerdale cast mates who get her through those tougher times. She said: 'On the tough days, I do think Mandy Dingle has been a great person to hide behind. I do hide behind the bravado. 'Because there are some days where you want to sort of roll over on your pillow and cry. But I am so grateful to do what I love, and I am very lucky. 'I am guilty of pressing the bravado button, but I have my wonderful co-stars around me, including Dom - who plays Paddy - and the incredible Bradley Johnson, who plays my son. 'We're all so in tune with each other, they've got my back and I've got theirs, and honestly we're so lucky we do, because sometimes I watch my scenes back and think 'if only people knew what was really going on in my head.'' But the star said she recognises that, for some people, it's hard to find others to turn to when losing someone, especially as 88% of bereaved people feel alone in their grief. Lisa continued: 'There are thousands of people who need to know that Sue Ryder is there, and that they don't have to be by themselves. 'My dad was a bit in denial when my mum died, and that is what the campaign is there for.' Bomb threat panic on Air India flight from UK as jet forced to divert in latest air drama 11 days after Ahmedabad crash

The threat came after the horror Air India disaster in Ahmedabad earlier this month, which killed all but one passengers and crew. The jet had departed at 8:26pm on June 21 from Birmingham Airport. But passengers were left confused when the Boeing 787-8 Dreamliner plane diverted from its original destination. The aircraft went on to land safely, and all those on board underwent standard security checks on Saturday. READ MORE WORLD NEWS CRASH CHAOS Air India slammed for 'repeated violations' as three officials sacked A spokesperson for the airliner confirmed to local media on Sunday that precautionary procedures were followed, and the situation was handled swiftly and securely. They said: "Flight AI114 from Birmingham to Delhi on June 21 received a bomb threat and was consequently diverted to Riyadh, where where it landed safely and completed security checks." No explosive material was found on board the jet after its inspection. And all passengers on the Boeing 787-8 Dreamliner were provided with hotel accommodation. This incident comes amid a recent string of chilling Air India issues, marked by a series of flight cancellations and disruptions since the deadly crash in Ahmedadbad on June 12. Just hours after the deadly smash, another Air India flight was forced to make an emergency landing in Thailand after a bomb threat. At least eight killed in horror hot air balloon crash in Brazil 'after aircraft caught fire and plummeted to the ground' Flight AI379 was travelling from Phuket to India's capital New Delhi - before it suddenly landed again after an explosion warning, according to authorities. The latest bomb threat comes as Air India continues to be hit with criticism for "repeated violations" - after three officials were sacked over "systemic failures" following the tragic crash. Cracks within the airline have started to show after flight AI171 smashed into a doctors' hostel and exploded into a huge fireball. Investigators are continuing their probe into what caused the airline's London-bound plane to plunge to the ground moments after takeoff in Ahmedabad. Air India said last Thursday that the doomed Boeing 787-8 Dreamliner plane was "well-maintained" and that the pilots were accomplished flyers. The airline's chief N Chandrasekaran also hit back at 'speculation' on what caused the London-bound flight to crash a minute after take-off. And it was revealed that the black box recovered from the flight could be sent to the US for analysis after being rescued from a blazing 1,000 degree inferno. The Indian government has the final say on who probes the device, but reports say the National Transportation Safety Board (NTSB) in Washington, D.C. is being considered for the analysis. A team of Indian investigators is expected to accompany the device to ensure all protocols are followed during the advanced data extraction. Investigators warned last Tuesday that the recovered black boxes may have melted due to the intense heat of the raging explosion. 52 Brits died on board the flight, but London local Vishwash Kumar Ramesh managed to cheat death when he escaped the blazing inferno following the crash. Theories have swirled over how the sole survivor managed to narrowly escape death when he was sat in seat 11A during the horror smash. Leading theories on Air India Flight AI171 THESE are some of the leading theories explaining the tragic Air India disaster which killed at least 270 people. Emergency power system: A small turbine generator was seen deploying as the Boeing 787 went down, experts said. Footage showed a "protrusion on the belly of the aircraft" with a "little grey dot" beneath it. Commentators say this was a system called Ram Air Turbine (RAT) poking out from the fuselage of the plane. Bird Strikes: A bird strike could have taken out both of the jet's giant General Electric engines. While a bird taking down something the size of a commercial airliner might sound fanciful, there are numerous examples. Wing flap position: Aviation experts have suggested the position of the aircraft's wing flaps could have played a role in the disaster. Video evidence suggests the flaps were either fully retracted or on a very minimal setting, which would have provided very little lift. The flaps provide crucial extra lift at low speeds during take-offs and landings. Pilot error: The state-of-the-art Boeing 787-8 is highly automated, with human pilots making only key decisions - but human error cannot be ruled out. Co-pilot Clive Kundar had more than 3,400 hours of flying experience and made the mayday call but Captain Sumeet Sabharwa at the helm had 11,500 hours - making him one of Air India's most experienced pilots. Heat: Planes get less lift on a hot day due to lower air density, and therefore they need to go faster to get as much lift as on a cooler day. This is particularly important when an airliner is heavy with fuel, passengers and baggage, as the Air India flight was. Technical error: Catastrophic technical or engineering issues have not been ruled out. The jet's complex design mean it could take months for a design or engineering fault to be pinpointed.
