Capricor Therapeutics Announces Completion of Mid-Cycle Review Meeting with FDA on Deramiocel for the Treatment of Duchenne Muscular Dystrophy Cardiomyopathy
-Company remains on track for PDUFA target action date of August 31, 2025-
-Advisory committee meeting to be held in advance of target action date-
SAN DIEGO, May 05, 2025 (GLOBE NEWSWIRE) -- Capricor Therapeutics (NASDAQ: CAPR), a biotechnology company developing transformative cell and exosome-based therapeutics for the treatment of rare diseases, today announced the completion of a mid-cycle review meeting with the U.S. Food and Drug Administration (FDA) for the Company's Biologics License Application (BLA) seeking full approval for deramiocel, an investigational cell therapy, as a treatment for patients diagnosed with Duchenne muscular dystrophy (DMD) cardiomyopathy. During the meeting, FDA stated that no significant deficiencies have been identified by the Review Committee and that the package is on track for a Prescription Drug User Fee Act (PDUFA) action date of August 31, 2025. The FDA has also confirmed its intent to hold an advisory committee meeting, although an official date has not yet been set.
'The successful completion of our mid-cycle review meeting along with the upcoming advisory committee meeting represents major milestones on the path towards approval of deramiocel,' said Linda Marbán, Ph.D., Chief Executive Officer of Capricor. 'Deramiocel is a first-in-class cellular therapy with the potential to halt or slow the progression of DMD-cardiomyopathy, and we are pleased to have the opportunity to present the efficacy and safety data to the advisory committee. We have been actively preparing for an advisory committee meeting, and we look forward to providing the physician and patient perspectives to highlight the weight of evidence supporting the transformative potential of deramiocel in treating DMD-cardiomyopathy.'
The BLA submission is supported by Capricor's cardiac data from its Phase 2 HOPE-2 and HOPE-2 Open Label Extension (OLE) trials compared to patient level data from an FDA-funded and published dataset on the natural history of DMD-cardiomyopathy and potential biomarkers of disease progression. Efficacy from the ongoing HOPE-3 study is not part of this BLA package submission.DMD is a devastating genetic disorder characterized by progressive weakness and chronic inflammation of the skeletal, heart and respiratory muscles with mortality at a median age of approximately 30 years. It is estimated that DMD occurs in approximately one in every 3,500 male births and that the patient population is estimated to be approximately 15,000-20,000 in the United States. DMD pathophysiology is driven by the impaired production of functional dystrophin, which normally functions as a structural protein in muscle. The reduction of functional dystrophin in muscle cells leads to significant cell damage and ultimately causes muscle cell death and fibrotic replacement. In DMD patients, heart muscle cells progressively die and are replaced with scar tissue. This cardiomyopathy eventually leads to heart failure, which is currently the leading cause of death among those with DMD. Treatment options are limited and there is no cure.Deramiocel (CAP-1002) consists of allogeneic cardiosphere-derived cells (CDCs), a rare population of cardiac cells that have been shown in preclinical and clinical studies to exert potent immunomodulatory and anti-fibrotic actions in preservation of cardiac and skeletal muscle function in dystrophiopathies such as DMD. CDCs act by secreting extracellular vesicles known as exosomes, which target macrophages and alter their expression profile to adopt a healing, rather than a pro-inflammatory phenotype. CDCs have been the subject of over 200 peer-reviewed scientific publications and have been administered to over 250 human subjects across several clinical trials.
Deramiocel for the treatment of DMD, has received Orphan Drug Designation from the FDA and European Medicines Agency (EMA). The regulatory pathway for deramiocel is supported by RMAT (Regenerative Medicine Advanced Therapy Designation) in the U.S. and the Advanced Therapy Medicinal Product (ATMP) Designation in the European region. In addition, if Capricor were to receive FDA marketing approval for deramiocel regarding the treatment of DMD, Capricor would be eligible to receive a Priority Review Voucher (PRV) based on its previous receipt of a rare pediatric disease designation.Capricor Therapeutics (NASDAQ: CAPR) is a biotechnology company dedicated to advancing transformative cell and exosome-based therapeutics to redefine the treatment landscape for rare diseases. At the forefront of our innovation is our lead product candidate, deramiocel, an allogeneic cardiac-derived cell therapy. Extensive preclinical and clinical studies have shown deramiocel to exert potent immunomodulatory and anti-fibrotic actions in preservation of cardiac and skeletal muscle function in dystrophiopathies such as DMD. Deramiocel is currently in late-stage development for the treatment of Duchenne muscular dystrophy. Capricor is also harnessing the power of its exosome technology, using its proprietary StealthX™ platform in preclinical development focused on the areas of vaccinology, targeted delivery of oligonucleotides, proteins and small molecule therapeutics to potentially treat and prevent a diverse array of diseases. At Capricor, we stand committed to pushing the boundaries of possibility and forging a path toward transformative treatments for those in need. Zscaler Will Hit $360 Soon: Here's the How and Why
Zscaler's FQ3 results and outlook make it a standout winner compared to its competitors, and analysts are raising their price targets as a result. This story originally appeared on MarketBeat [content-module:CompanyOverview|NASDAQ:ZS] Zscaler's (NASDAQ: ZS) share price is expected to reach $360 due to technical price action, favorable analyst sentiment, and the results driving it. The latest price action has this market breaking out to new highs, confirming not only the near-term uptrend but also breaking the market out of a long-term trading range. The trading range was in place due to valuation concerns and risks of slowing growth. The breakout occurred because the business fundamentals are catching up to the valuation, leading to an improved outlook. That is for sustainable 20% growth, free cash flow, and improving shareholder value. Regarding the soon in this forecast, the technical action suggests this move could be completed by late this year or by early 2026. The target is due to the magnitude of the trading range, which, in this case, is a minimum target. The price action has broken to new highs robustly and may consolidate well above the critical support target, indicating that a bull-case scenario is in place. The market could rise to a new all-time high in that scenario due to the magnitude of the recovery rally currently underway. Using it to project targets puts this market in the range of $375 to $403, sufficient for a new high and a likely precursor to even higher highs. Here's why. Zscaler Wows the Analysts With Fiscal Q2 Results Analyst activity in Zscaler was tepid for more than a year, with price target reductions and generally poor responses to results hampering the price action. Then Bam! Zscaler reports its FQ3 2025 results, and the analyst's attitude shifts. MarketBeat reported 24 revisions from the 36 analysts tracked by its platform, including a few reiterated price targets but more increases than decreases. There were no decreases tracked. The result is a 17% increase in the consensus price target within days of the release, a 25% increase relative to last year, and most revisions leading to the high-end range. The consensus assumes that fair value is near the June opening price levels, but the high-end range adds 15% to it. At $320, it is still well below the $375 to $400 high-end technical target, but a step in the right direction that could easily turn into a trend, assuming upcoming reports are equally good. The takeaway from the analysts' reports is that the results are solid despite macroeconomic uncertainties that negatively impacted guidance for competitors. The strength in Q3 is expected to carry over into Q4, leaving the company with momentum heading into the next fiscal year. New products help the platform carry momentum, and the new Z-Flex is expected to do the same. Z-Flex is a new purchasing model that allows businesses to streamline adjustments to their security stack, reducing costs while improving flexibility and scalability. Zscaler Had a Solid Q3 Despite Slowing Growth The only bad news coming out of Zscaler's Q3 report is that growth slowed to the low 20% range. However, the revenue exceeded the analysts' forecasts, growth topped 20% for another quarter, and was further bolstered by accelerated gains in billings and deferred revenue. [content-module:Forecast|NASDAQ:ZS] Other key highlights for investors to focus on include the 21% increase in net income, an 18% free cash flow margin, and a 25% cash build-up on the balance sheet. Regarding the balance sheet, Zscaler has more than $3 billion in cash and equivalents, and equity is rising, up 41% year-to-date in F2025. Regarding the guidance, the company forecasts billings in alignment with the analysts' consensus forecast, leaving the Q3 strength to carry through to the full-year result, and it may be cautious. The final piece of the puzzle is the sell-side interest, including short-sellers and institutional investors. The institutions, as a group, own a solid but still small 41% of the stock as of early June, but have been buying it on balance throughout the year. That provides a solid support base and tailwind for price action now amplified by short-covering. The short interest ahead of the release wasn't astronomically high at 9%, but it had risen in previous reports and was at a long-term high, setting the market up for a short-covering rally if not a short-squeeze. Ascentage Pharma Releases Promising Clinical Data on Alrizomadlin Monotherapy and Combinations in Solid Tumors
ROCKVILLE, Md. and SUZHOU, China, June 02, 2025 (GLOBE NEWSWIRE) — Ascentage Pharma (NASDAQ: AAPG; HKEX: 6855), a global biopharmaceutical company dedicated to addressing unmet medical needs in cancers, announced that it has released the latest clinical data from its Phase II study of the MDM2-p53 inhibitor alrizomadlin (APG-115) as a single agent or in combination with PD-1 inhibitor toripalimab in patients with advanced adenoid cystic carcinoma (ACC) or other solid tumors in a poster presentation at the 61st American Society of Clinical Oncology (ASCO) Annual Meeting. Alrizomadlin is the first MDM2-p53 inhibitor to enter clinical development in China and a key investigational drug candidate in Ascentage Pharma's apoptosis-targeted pipeline with global first-in-class potential. The ASCO Annual Meeting showcases the most cutting-edge research in clinical oncology and state-of-the-art advanced cancer therapies and is the world's most influential and prominent scientific gathering of the clinical oncology community. Returning to the ASCO Annual Meeting for the eighth consecutive year, Ascentage Pharma has garnered growing interest from the global research community. This year, two studies of the Bcl-2 inhibitor lisaftoclax (APG-2575) and the MDM2-p53 inhibitor alrizomadlin, key drug candidates in the company's apoptosis-targeted pipeline, have been selected for presentations, including an oral presentation, at the ASCO Annual Meeting. These clinical data on alrizomadlin in ACC and other solid tumors demonstrated the antitumor activity of alrizomadlin monotherapy in patients with advanced ACC or malignant peripheral nerve sheath tumor (MPNST). Moreover, alrizomadlin in combination with toripalimab was well tolerated and showed therapeutic potential in MPNST, biliary-tract cancer (BTC), and liposarcoma (LPS). Prof. Ye Guo, MD, a Principal Investigator of the study from the Department of Medical Oncology, Shanghai East Hospital, noted, 'ACC is a rare cancer type that lacks effective treatment options, and the treatment with antiangiogenic tyrosine kinase inhibitors faces certain limitations and safety concerns. At ASCO 2025, our team presented data of alrizomadlin in patients with ACC that demonstrated an objective response rate (ORR) of 16.7% and a disease control rate (DCR) of 100%. These results suggest that the targeted inhibition of the MDM2-p53 pathway has antitumor activity in ACC; therefore, it can potentially offer a new treatment strategy to patients with ACC.' Prof. Ning Li, MD, a Principal Investigator of the study from the Chinese Academy of Medical Sciences Cancer Hospital, commented, 'Currently, there are limited treatment options for patients with sarcomas such as MPNST and LPS. In this study, alrizomadlin both as a monotherapy and in combination with a PD-1 therapy, showed favorable antitumor activity in MPNST. The clinical benefit was particularly notable in patients who received the combination regimen, with two patients with MPNST achieving long-term responses that lasted more than 60 and 96 weeks, respectively. Furthermore, alrizomadlin in combination with a PD-1 therapy also showed clinical activity in LPS. These results suggest that alrizomadlin monotherapy and combination regimens may bring clinical benefit to more patients with sarcoma.' Dr. Yifan Zhai, Chief Medical Officer of Ascentage Pharma, said, 'Alrizomadlin is an investigational drug with global first-in-class potential. The clinical data presented at this year's ASCO Annual Meeting demonstrated the therapeutic potential of alrizomadlin, both as a monotherapy and in combinations, in ACC and other solid tumors, and underscored the synergistic effects between alrizomadlin and immunotherapies, thus suggesting a promising therapeutic strategy for various solid tumors. We are focused on addressing unmet clinical needs in China and around the world, and intend to further accelerate our clinical programs to advance more novel treatment options for patients as soon as possible.' Highlights of this abstract selected for presentation at ASCO 2025 are as follows: A Phase 2 Study of Novel MDM2 Inhibitor Alrizomadlin (APG-115) With or Without Toripalimab in Patients with Advanced Adenoid Cystic Carcinoma (ACC) or Other Solid Tumors Abstract #: 6102 6102 Format: Poster Presentation Poster Presentation Session Title: Head and Neck Cancer Head and Neck Cancer Principal Authors: Ye Guo, MD, Department of Medical Oncology, Shanghai East Hospital, China; Ning Li, MD, Chinese Academy of Medical Sciences Cancer Hospital, China; Xing Zhang, MD, Melanoma and Sarcoma Medical Oncology Unit, Sun Yat-sen University Cancer Center, China; Meiyu Fang, MD, Department of Rare Cancer & Head and Neck Medical Oncology, Cancer Hospital of the University of Chinese Academy of Sciences, China; Shuhang Wang, MD, Chinese Academy of Medical Sciences Cancer Hospital, China, et al. Highlights: Alrizomadlin is a novel investigational oral MDM2 inhibitor that has shown a manageable safety profile with initial clinical activity in ACC. As of the data cutoff date of February 13, 2025, 57 patients with advanced ACC, malignant peripheral nerve sheath tumor (MPNST), liposarcoma (LPS), biliary-tract cancer (BTC), and other tumors were enrolled. Alrizomadlin monotherapy showed encouraging antitumor activity in patients with advanced ACC or MPNST. Alrizomadlin in combination with toripalimab was also well tolerated and demonstrated antitumor activity in patients with MPNST, BTC, and LPS. In the monotherapy arm, 17 patients were efficacy-evaluable. The ORR was 16.7%, and the DCR was 100% in 12 patients with ACC. The DCR was 80% in 5 patients with MPNST, 4 of whom achieved stable disease (SD). In 24 safety-evaluable patients, 33.3% reported grade 3 or higher treatment-related adverse events (TRAEs). Three patients (12.5%) experienced treatment-related serious adverse events (SAEs). One patient discontinued treatment because of TRAE. In the combination arm, 29 patients were efficacy-evaluable. The ORR was 16.7% and the DCR was 100% in 6 patients with BTC. The ORR was also 16.7% and the DCR was 66.7% in 6 patients with LPS. Two patients with MPNST had confirmed partial response (PR) with prolonged progression free survival (PFS) of 60 + weeks and 96 + weeks, respectively. In 27 safety-evaluable patients treated with alrizomadlin at the 150 mg dose level, 12 (44.4%) experienced grade 3 or higher TRAEs. Treatment-related SAEs were reported in 8 patients (29.6%). One patient discontinued treatment because of TRAE. * Alrizomadlin (APG-115) is an investigational compound and has not been approved by the US FDA. Canadian Health Tech Cuts Hospital Emergency Department Triage Time by up to 60%
Photo Courtesy of EmergConnect TORONTO, June 02, 2025 (GLOBE NEWSWIRE) — EmergConnect's digital platform helps hospitals cut ER triage time by up to 60%. The system gives patients real-time wait estimates, supports nursing staff, and is gaining traction as an early solution for Canada's strained emergency departments. 'We've seen ER triage times cut by up to 60% with our platform,' says Ron Galaev, founder and CEO of EmergConnect. 'It's about giving nurses back their time and giving patients the clarity they deserve—because uncertainty shouldn't be part of seeking care.' Hospitals using EmergConnect allow patients to begin their emergency department visit before they even walk through the door, or comfortably in their seat in the waiting room. Individuals can enter symptoms and details through their phones, register in advance, and receive an estimate of how long they might wait. Nurses then review the information in the EmergConnect portal and send it straight into their hospitals' system with just one click. The company worked for eight months with national and provincial health groups to create a tool compatible with current triage standards. The platform also includes tools for paramedics, to reduce offload times, and a redirection system that directs patients to less crowded hospitals nearby or for lower acuity patients alternative care options altogether. 'When we first started analyzing emergency departments, we were shocked to discover nurses had to click 64 times just to intake a single patient, we've reduced that to just four clicks' says Marina Gavrilov, CTO of EmergConnect. Hospitals say the platform reduces staff pressure by cutting out manual steps from the intake process. Hospitals use EmergConnect's tools to direct patients who do not need emergency care to nearby clinics, easing demand. Its AI-based predictions give individualized wait times, tailored to the hospital's current situation. 'By eliminating manual steps and guiding non-urgent cases to nearby clinics, we're helping hospitals ease pressure on staff while ensuring patients get the right care, faster,' Galaev explains. 'Our AI doesn't just estimate wait times,it personalizes them based on what's actually happening inside the hospital.' The company raised $2 million CAD in late 2024 and plans to expand to British Columbia and Quebec. It also considers tackling problems facing the U.S. system next. 'We help hospitals solve one of the biggest day-to-day problems they face,' Galaev said. EmergConnect's leaders believe the tool will serve more hospitals seeking to improve efficiency and patient experience. Four large hospital systems already use the product, and the company says interest is growing quickly, with aims at having the majority of the Canadian healthcare system covered in the coming years. As emergency departments continue to face staffing shortages and surging demand, tools like EmergConnect are becoming part of more conversations between hospital administrators and frontline staff. About EmergConnect EmergConnect is a Canadian digital health company focused on improving same-day care through an integrated triage and registration platform. Designed for patients, nurses, paramedics, and administrators, the system helps reduce emergency room wait times, streamline intake, and improve care navigation.
