Scientists Take Major Step Towards Creating Synthetic Life

For the first time ever, a synthetic eukaryotic genome has been created.
By taking yeast cells and rebuilding their genomes, scientists were able to create a yeast that more resilient and produced more spores—something that could mean more food on a much larger scale
Creating synthetic genomes could eventually make sustainable manufacturing processes that use eukaryotic bacteria more efficiently.
While the concept of creating synthetic life from nothing is still firmly in the science fiction category, synthetic genomes are now reality. Researchers—led by synthetic biologist Hugh Goold of Macquarie University in Australia—have created the first synthetic eukaryotic genome for a species of yeast. Previous experiments had been done on prokaryotes (single-celled organisms that lack a nucleus), but a full synthetic genome had not been reconstructed for any single-celled eukaryotic organisms (which have a nucleus and organelles with distinct functions).
This synthetic genome tech means that life-forms can be customized depending on what aspects need to be enhanced. As part of the Sc2.0 Project, for example, the yeast Saccharomyces cerevisiae was redesigned and reconstructed to produce more spores and keep from mutating (since this yeast is prone to spontaneous mutations) as it grew.
This genomic makeover was a decade in the making. Also known as brewer's yeast, Saccharomyces cerevisiae has a long history of being used in brewing, winemaking, and baking. Reconstructing its genome would not only give it the qualities it needs to resist disease and survive climate change, but allow it to yield a higher quantity and quality of food while maintaining sustainability.
'The technology of synthetic genomics can help to secure supply chain in future situations where changing climates, further global pandemics, and conflict threatens the availability of critical and conventional feedstocks of food and pharmaceuticals,' the researchers said in a study recently published in Nature.
To create a specific synthetic chromosome within the synthetic genome, Goold started with several strains of S. cerevisiae, which had already had synthetic pieces of DNA inserted. These were backcrossed—a process which involves mating genetically different parent cells to create hybrid offspring, which are then mated with the parent cells. Backcrossing may sound like inbreeding, but actually makes it easier to isolate any characteristics that may require a closer look.
The strain with the newly created synthetic chromosome, SynXVI, suffered impaired growth and often mutated. More backcrossing allowed the team to identify which areas of SynXVI were glitching. Why were there issues with a genome that was supposed to be version 2.0 of the original? It turned out that errors in placing genetic markers—genes or short sequences of DNA that are used to identify genes and chromosomes inside a genome—sometimes interfered with the function of yeast cells.
Genetic glitches in the chromosome were identified and edited with CRISPR D-BUGS (which sounds like some sort of futuristic pest killer) and other genetic tools that were able to zero in on specific areas of the genome and correct them. Boosting yeast growth and resilience meant getting it to grow on glycerol (a carbon source) to absorb the carbon it needed at a certain temperature. It turned out that one section of the genome was causing a deficit of copper, but this was easily solved by adding copper sulfate to the glycerol.
Despite the glitches, this technology opens up opportunities for creating more than better crops. Eukaryotic microbes already used in sustainable manufacturing processes could be modified to become even more efficient.
'S. cerevisiae holds great potential in libraries of new chromosomes,' the researchers said in the same study, '[including] neochromosomes, and genomes for subsequent transfer into cells to create life forms tailored to humankind's needs.'
Someday, maybe even artificial mammalian genomes could be created, But for now, just one yeast is offering so many possibilities.

But most weed mats are placed closer to the surface where heat and microbes slowly break them up. 'The whole issue with plastics in the environment is they shed. Once they start breaking down they release microplastics. And that's the issue, they don't go away they just disintegrate,' he said. 'They just break up, not down. Experts Found 6,000-Year-Old Human Remains With No Relatives

Here's what you'll learn when you read this story: An analysis of bones from ancient people who once lived in Colombia has discovered DNA that does not directly connect them to any other ancient or modern population in South America. It is thought that these people might somehow be related to speakers of Chibchan languages, which are spoken in the area where they once lived. More genomic research will be needed to demystify who these unknown people were and whose ancestors they might be. Around 6,000 years ago, hunter-gatherers who migrated south settled in the Bogotá Altiplano of what is now Colombia, transitioning to an agricultural society over the next 4,000 years. Then they vanished. Whoever these people were, they disappeared from the genetic record. The team of researchers who discovered them through fragmented DNA in their skeletal remains have not been able to find any ancient relatives or modern descendants. They are strangely not related to Indigenous Columbians, having more of a connection to people who now live on the Isthmus of Panama and speak Chibchan languages. It could be possible that they spread through the region, mixing with local populations for so long that their genes were diluted, but no one can be sure. Genetics tell the story of how the Americas were populated by the ancestors of modern Indigenous people. Their origins lie in Siberian and East Asian groups who are thought to have first mixed 20,000 years ago, during the Late Paleolithic, later crossing a bridge of ice to North America some 16,000 years ago. It was then that this lineage split into northern Native American and southern Native American lines. While the northern Native American ancestry is mostly made up of populations that stayed in North America, three more lineages branched into southern Native American ancestry, which would reach further south. 'The Isthmo-Colombian area, stretching from the coast of Honduras to the northern Colombian Andes, is critical to understanding the peopling of the Americas,' the researchers said in a study recently published in Science Advances. 'Besides being the land bridge between North and South America, it is at the center of the three major cultural regions of Mesoamerica, Amazonia, and the Andes.' Each southern Native American lineage can be traced back to its earliest ancestors. There is one line descending from the Anzick-1 individual discovered in 1968, when construction workers unearthed the 12,700-year-old skull of a child belonging to the Clovis people, one of the earliest known peoples in the Americas. This young boy is related to modern Indigenous people from North, Central and South America. Another lineage that is found in the Central Andes comes from ancient people living in California's Channel Islands. Yet another lineage, also descended from the Clovis population, is associated with the oldest Central and South Americans from Belize, Brazil and Chile. When exactly these populations appeared in Central and South America is still mostly unknown, but they must have arrived by traveling over the land bridge that connects the southernmost part of Central America to the South American subcontinent. There is also a linguistic connection. Speakers of Chibchan languages share some genetic and cultural aspects with the mysterious people who cannot be traced directly to any population. When and where the ancestral Proto-Chibchan language originated is unclear, but distinct languages are thought to have started evolving from it several thousand years ago, possibly in southern Central America. Chibchan speakers in this region have kept the largest number of these languages alive. While genetic analysis of local Indigenous people has shown that they are related to more ancient speakers of Chibchan languages, some findings suggest that they are not directly descended from the first people to settle in that part of Colombia. By studying both mitochondrial DNA (mtDNA) and genome-wide data from 21 ancient individuals who lived in this region from 6,000 to 500 years ago, the researchers were able to find out some information about who they were, but not all the answers. More ancient Panamanians were found to be related to modern Chibchan speakers than ancient Colombians. However, Indigenous Chibchan speakers from Central America are the modern population genetically closest to ancient Colombians who lived after 2,000 years ago. Many groups who were around at the same time and spoke similar languages to the unknown people still need further investigation. 'Ancient genomic data from neighboring areas along the Northern Andes that have not yet been analyzed through ancient genomics,' the researchers said, 'such as western Colombia, western Venezuela, and Ecuador, will be pivotal to better define the timing and ancestry sources of human migrations into South America.' Cannabis Compound Could Protect Us From Deadly Fungal Disease
A dangerous fungal pathogen has proven no match for what may be one of the most useful plants in nature. Scientists studying the chemical properties of cannabis have found it kills one of the most dangerous fungal pathogens in the world – in a laboratory setting, at least. Cryptococcus neoformans, a species of fungus behind cryptococcosis and cryptococcal meningitis, appears to be vulnerable to topical treatment with cannabidiol and cannabidivarin, compounds found in the plant Cannabis sativa. "When Cryptococcus neoformans gets to your central nervous system, it causes life-threatening meningitis," explains biologist Hue Dinh of Macquarie University in Australia. "The mortality rate is very high, and it's really hard to treat." Fungal pathogens pose a pretty significant threat to human health, with around 300 species known to cause diseases in humans, with varying levels of severity. Because pathogens such as fungi and bacteria continually develop resistances to drugs, new treatment options are continually needed to keep them at bay. One strategy is to look at medications that have already been approved for human use for other ailments. Dinh and her colleagues turned their research to cannabis, isolating five compounds to test on C. neoformans and a range of other pathogens. They isolated the fungal species in a lab, and applied the compounds. Cannabidiol (CBD), which is non-psychoactive, and cannabidivarin (CBDV), which is psychoactive, both killed C. neoformans adroitly, acting even faster than current antifungal treatments. They were also effective at eradicating the fungal pathogens responsible for such conditions as jock itch and athlete's foot. "Proteomics analysis revealed that the antifungal activity of CBD and CBDV was linked to destabilization of the membrane, alterations in ergosterol biosynthesis, disruption of metabolic pathways, as well as selective involvement of mitochondrial-associated proteins," the researchers wrote in their paper. It's one thing to kill a fungus in a petri dish, but quite another to observe the treatment working in a living system. To test their findings further, the researchers turned to Galleria mellonella, the greater wax moth. The larvae of this moth possess an innate immune response that is similar to that of mammals. They are also inexpensive to obtain, have short lifespans, and require no special equipment to keep, making them an excellent model for large-scale studies of infectious pathogens and the treatments thereof. Moth larvae were given small burn wounds, and then divided into groups. One group was left alone with just the burn wounds as a control; the remainder were infected with C. neoformans, and treated with different medicaments. One group was treated with CBD dissolved in dimethyl sulfide. Another group was treated with dimethyl sulfide without the CBD. Finally, the last group was treated using Amphotericin B, a medication used to treat serious fungal infections in humans. The results were striking. The survival rate of the larvae treated with CBD was significantly higher than the survival rate of the larvae treated with the dimethyl sulfide alone, and also higher than the survival rate of the Amphotericin B group. In fact, it was nearly as high as the survival rate of the control group – the larvae that had not been infected with the fungus at all. Although the treatment of infections that reach the lungs and brain is a bit more complicated, the result suggests that, at the very least, the topical application of cannabidiol might be effective at treating a range of fungal skin infections. "If we can demonstrate that these ones work well for common infections," Dinh says, "you could actually just get some CBD oil and then rub it on your skin to treat it."
