
Cancer breakthrough as doctors unveil way to zap tumors into oblivion without chemo
A new cancer treatment that uses light could remove the need for harsh chemotherapy or radiotherapy.
Texas researchers have devised a way to use infrared light as a jackhammer that can tear cancer cells apart from the inside, showing in trials to have eliminated melanomas, the deadliest form of skin cancer.
A synthetic blue dye often used in hospital imaging naturally binds to cancer cells.
Researchers found that when the cancer cells were zapped with infrared light, it triggered violent shaking that tore through the cancer cell's membrane, causing it to die without any heat or harsh drugs.
The latest breakthrough method – known as vibronic-driven action – had a 99 percent efficiency against lab cultures of human skin cancer cells, and half of the mice with melanoma tumors became cancer-free after just one treatment.
'It is a whole new generation of molecular machines that we call molecular jackhammers,' said Rice University nanotechnologist Dr James Tour.
Near- infrared light can penetrate up to 10 cm into the body, reaching organs and bones without surgery. Light particles can also be functionalized to only bind to cancer cells, mitigating damage done to healthy cells.
The treatment itself is highly targeted because the atoms of dye bind to the receptors on cancer cells while leaving healthy cells largely untouched. It has already shown in followup trials to kill colorectal cancer, prostate cancer, and breast cancer cells.
The next step will be determining whether this same molecular jackhammer will destroy cancer cells in humans.
'This study is about a different way to treat cancer using mechanical forces at the molecular scale,' said Ciceron Ayala-Orozco, a Rice research scientist who is a lead author on the study.
If scaled up to human patients, the treatment's impact could be sweeping. An estimated 1.5 million Americans are living with melanoma, 1.4 million have colorectal cancer, and 300,000 men have prostate cancer. In 2025, approximately 316,950 women will be diagnosed with severe, invasive breast cancer.
The unassuming dye found in hospitals nationwide is a key feature of vibronic-driven action. After it binds to receptors on melanoma cells, it is assaulted with infrared light, which triggers an earthquake in the cell. The dye atoms begin to vibrate in unison.
The dye's electrons sync up into a "plasmon"—a wave of collective motion. This energy couples with the molecule's natural vibrations, making the entire structure shudder at 41 trillion times per second.
The violent oscillating energy is transferred directly into the cell's fatty membrane, while nanoscale holes tear open, allowing fluid and foreign molecules to flood in.
This fast-moving mechanical force is such that 'anything around it will be disassembled,' Ayala-Orozco said.
The cancer cell crumbles as the holes poked in its membrane increase, and dies without further use of any drug or radiation treatment.
The Texas team's findings were published in the journal Nature Chemistry.

Ayala-Orozco said: 'These molecules are simple dyes that people have been using for a long time. They're biocompatible, stable in water and very good at attaching themselves to the fatty outer lining of cells. But even though they were being used for imaging, people did not know how to activate these as plasmons.'
After struggling for years to make blue-light-activated motors work against mouse tumors, Ayala-Orozco turned to explore completely different approaches.
'I started to think that maybe what I needed was not necessarily a motor, but a molecule that absorbs near-infrared light, in the hopes that this molecule could be activated and could move in a different way — not necessarily by rotation,' he told Healio.
'That's when I started thinking about some molecules called cyanines and started connecting the dots with a property from photophysics called plasmons, which I studied during my PhD to treat cancer. They are activated by near-infrared light. At some point, I decided I had to try this.'
When researchers turned their microscopes on other cancer types, they made an exciting discovery: the molecular jackhammers proved just as devastating against prostate, breast, and colorectal cancer cells in petri dishes.
'No matter which line, we are able to eliminate 100 percent on cell culture,' Ayala-Orozco said.
The university will need to partner with a private company to scale up, overcome regulatory hurdles, and enter human trials, 'hopefully, within 5 to 7 years,' Ayala-Orozco said.

US FDA puts on hold Rein Therapeutics' lung disease drug trial
June 12 (Reuters) - Rein Therapeutics (RNTX.O), opens new tab has paused patient enrollment and dosing in a mid-stage trial of its lung disease drug in the U.S. after the Food and Drug Administration placed a clinical hold, the drug developer said on Thursday. Shares of the company fell nearly 10% after the bell. The company was testing a drug named LTI-03 to treat patients with idiopathic pulmonary fibrosis (IPF). The disease causes scarring of the lungs that makes it harder for them to work properly. Rein said in a filing that it is actively working with the FDA to remove the clinical hold. The FDA and Rein did not immediately respond to Reuters' requests for comment on why the hold was placed. The drug was well-tolerated and safe in an early-stage study in patients with IPF, the company said in the filing. No drug-related serious adverse events have been reported in any studies treating patients with LTI-03, it added. Rein continues to study the drug and enroll patients in Australia and Europe.

Exclusive: After Trump feud, role of Musk's SpaceX in Golden Dome missile shield in question
WASHINGTON, June 12 (Reuters) - The role of Elon Musk's SpaceX in an ambitious new U.S. missile defense system is in question following the dramatic feud last week between the billionaire entrepreneur and President Donald Trump, according to three people familiar with the project. The White House until recently had considered a plan for SpaceX, Musk's rocket and satellite venture, to partner with software maker Palantir (PLTR.O), opens new tab and drone builder Anduril to construct crucial elements of the project, dubbed "Golden Dome." The administration had instructed the Pentagon to prioritize a network of satellites for the purpose, these people said. But a new framework for the system, which would seek to track and prevent possible missile attacks against the United States, is now being considered that could reduce the role of SpaceX. One possibility, the three people said, could initially forego SpaceX's satellite capabilities and focus on the expansion of existing ground systems for missile defense instead. In a statement, a White House spokesman said "the Trump Administration is committed to a rigorous review process for all bids and contracts." A senior Defense Department official said the Pentagon "has no announcements regarding future contracts associated with the Golden Dome effort." SpaceX, Anduril and Palantir didn't respond to requests for comment. A reduced role for SpaceX would represent the first known setback to Musk's huge volume of business with the U.S. government since his break with Trump last week. The shift in plans, especially for a project that Trump has touted as paramount for U.S. defense strategy, also underscores the highly personalized nature of the president's leadership, aerospace and defense experts said. "That people guiding the program or building it are approved based on their political affiliation signals a real concern that the project itself is very politicized and not being conducted on the technical merits," said Laura Grego, a missile defense expert and research director at the Union of Concerned Scientists, a non-profit based in Cambridge, Massachusetts. In its statement to Reuters, the White House said any decision would be made "prioritizing the best deal for America and leveraging the most advanced and innovative technology." Trump in May said the defense shield should be operational by the end of his presidency, January 2029. But industry experts have said that timeframe, and a projected cost of some $175 billion, could be too optimistic. The change in the proposed "architecture" of the system, the three people said, could have the political advantage of allowing the current administration to deliver at least a portion of it. It isn't clear how soon a final decision on the project could come or whether the ultimate role of any company, including SpaceX, has been determined. Trump's efforts to roll out the project fast have led to uncertainty about the project's details and a scramble by contractors to be involved, industry experts and some of those involved in its development told Reuters. "To this day, no one knows what the requirements are," said one of the people familiar with the process. "There isn't a coordinated effort with a true vision. All of these companies are just grabbing at this pot of money." SpaceX, Anduril and Palantir were all founded by entrepreneurs who have been major political supporters of Trump. The three companies had previously met with top administration officials and decisionmakers from the Defense Department to discuss Golden Dome, according to people familiar with those discussions. Before his high-profile falling out with the president, Musk served as a key Trump advisor and donated more than a quarter of a billion dollars to help elect him. But the recent dispute, which included Musk calling for Trump's impeachment and accusing the president of improper involvement with disgraced financier and sex offender Jeffrey Epstein, triggered the change in direction, the three people told Reuters. "Because of the blowup, the Pentagon has been given the space to look at other alternatives," one of the people said. In recent days, Musk has sought to temper the dispute, saying he regretted some of his comments and taking down some of his social media criticism of Trump, including the call for impeachment. Earlier this week, Press Secretary Karoline Leavitt told reporters that Trump appreciated Musk's apology and that she was unaware of any administration efforts to review Musk contracts because of the dispute. Reuters couldn't determine whether Musk's conciliatory overtures might improve SpaceX's chances of winning Golden Dome contracts or securing further new business with the U.S. government. SpaceX had pitched for a part of the Golden Dome initiative called the "custody layer," a constellation of between 400 and 1,000 satellites that would detect missiles, track their trajectory, and determine if they are heading toward the U.S., Reuters reported in April. In a January 27 executive order, Trump mandated the selection of a proposed "architecture" for Golden Dome and an implementation plan by the end of March. The order called a missile attack "the most catastrophic threat facing the United States."
