Breakthrough cancer test ‘predicts whether chemotherapy will work'

A breakthrough test can successfully predict whether chemotherapy will work so patients can avoid needless side effects.
The test, developed by Cambridge University, looks at the structure of tumour DNA and forecasts whether it will resist treatment.
It was piloted using data from 840 patients with different types of cancer and found that it could help spot whether treatment was likely to fail for ovarian, prostate and breast cancers.
James Brenton, professor of ovarian cancer medicine at the Cancer Research UK Cambridge Institute, said: 'Sadly, there are too many cases where cancer is resistant to chemotherapy treatment – meaning unpleasant side effects for the patient with limited benefit to them.
'Chemotherapy is a mainstay of cancer treatment and saves many lives. Yet in many cases, it has been administered the same way for over 40 years.
'By understanding who is most likely to respond to it, chemotherapy could become a more tailored treatment across different types of cancer.'
The test can accurately predict resistance to treatment of three common types of chemotherapy – platinum-based, anthracycline and taxane chemotherapy – which are given to nearly 100,000 patients in Britain each year.
Although such treatments can help keep cancer at bay, they are also toxic to healthy cells and can lead to unpleasant side effects, such as fatigue, a weakened immune system, nausea and hair loss.
Experts hope that tests will be used after diagnosis to classify patients as either 'chemotherapy resistant' or 'chemotherapy sensitive' to treatments, to avoid giving patients therapies that will not work.
Dr Iain Foulkes, executive director of research and innovation at Cancer Research UK, which helped fund the research, said: 'The days of chemotherapy being offered as a 'one-size-fits-all' treatment are ending.
'Thanks to this research, we are moving towards a future where personalised cancer treatment is an option for many patients.
'Only by delivering more optimised, successful treatment will more people live longer, better lives, free from the fear of cancer.'
Researchers are now working with the pharmaceutical industry to develop the test and will be applying to regulators for its use in the clinic.
The scientists are also developing further tests for other targeted cancer drugs to find out how widely the technology could be used across different cancer types.
Dr Ania Piskorz, co-lead author and head of genomics at the Cancer Research UK Cambridge Institute, said: 'It was important to us to create a test that could be easily adopted in the clinic, using material we already collect during diagnosis and well-established genomic sequencing methods.
'The test is based on the full DNA sequence that we get from these methods, and we can adapt it to work alongside other genomic sequencing methods that are commonly used to personalise treatment for cancer.'

A deadly fungus dating back to ancient tombs could help treat blood cancer, researchers have revealed. Aspergillus flavus is a fungus that grows on dead plant tissue in soil, spreading to cereal grains, legumes and tree nuts. It's also been nicknamed the 'pharaohs' curse' after researchers studying the tombs of ancient kings over the past several decades have suddenly been struck by deadly respiratory issues. Killing up to 50 percent of those it strikes, the fungus is thought to 'eat people from the inside out,' and experts fear climate change may lead it to spread. But researchers in Pennsylvania and Texas have found polar opposite effects. In a newly released study, they found Aspergillus flavus produces interlocking ringed molecules they named asperigimycins. When tested against human leukemia cells, two of the four asperigimycins showed potent cancer-killing effects But when the researchers enhanced the asperigimycins by adding a lipid to it - a fatty molecule - the asperigimycins treated leukemia cells just as well as cytarabine and daunorubicin, two FDA-approved drugs that have been used to treat the cancer for decades. They believe asperigimycins may attack structures responsible for cell division, which normally causes healthy cells to mutate into cancerous ones. The team compared the surprising benefit of the deadly fungus to crucial advancements in treating diseases, such as the invention of penicillin. Aspergillus flavus has been nicknamed the 'pharaohs' curse' after researchers studying the tombs of ancient kings over the past several decades have suddenly been struck by deadly respiratory issues Dr Sherry Gao, senior study author and associate professor in chemical and biomolecular engineering and in bioengineering at the University of Pennsylvania, said: 'Fungi gave us penicillin. 'These results show that many more medicines derived from natural products remain to be found.' The findings come as 60,000 Americans are struck by leukemia every year and about 23,000 die. Aspergillus flavus was first thrust in the spotlight in the 1920s after a team of archeologists opened the tomb of King Tutankhamun in Egypt and suddenly become ill. After their deaths, rumors circulated of a curse. About 50 years later, in the 1970s, a dozen scientists opened the tomb of Casimir IV, who ruled Poland in latter half of the 15th century. A few weeks later, 10 of the researchers died. Later investigations revealed the presence of Aspergillus flavus. It's unclear how many people worldwide are affected by Aspergillus flavus, but it's believed to kill up to 50 percent of patients it infects by producing spores that attack the liver and lungs, especially in people who are immunocompromised. In the new study, published Monday in the journal Nature Chemical Biology, researchers looking at Aspergillus flavus samples found they had molecules with interlocking rings, which had never been previously described. They dubbed them asperigimycins. Even with no modification, they found two of the four variants of asperigimycins they looked at had potent effects against leukemia cells. They then added a lipid that's also found in royal honey to another variant of asperigimycins and found the method killed just as many cancer cells as the drugs cytarabine and daunorubicin. Both drugs have been linked to a roughly 50 to 80 percent remission rate. The team also found the gene SLC46A3 helps molecules like asperigimycins exit lysosomes, tiny sacs that collect materials that enter cells. Qiuyue Nie, lead study author and a postdoctoral fellow at the University of Pennsylvania, said: 'This gene acts like a gateway. It doesn't just help asperigimycins get into cells, it may also enable other "cyclic peptides" to do the same.' Roughly 2,000 of these cyclic peptides have been shown to treat diseases like cancer and lupus, an autoimmune condition, but most need to be modified in some way. 'Knowing that lipids can affect how this gene transports chemicals into cells gives us another tool for drug development,' Nie said. However, the asperigimycins had no effect on breast, liver or lung cancer cells, siggesting they only disrupt cell division for certain types of cancer cells. Nie cautioned findings are still early but mark the beginning of an 'unexplored region with tremendous potential.' The team will next test asperigimycins in animals with the hope of moving on to human clinical trials. Dr Gao said: 'Nature has given us this incredible pharmacy. 'It's up to us to uncover its secrets. As engineers, we're excited to keep exploring, learning from nature and using that knowledge to design better solutions.'

The Royal Mile has landed at 16th on the list, with the tourist hotspot being singled out for being home to an astronomical number of unhealthy stores. Chemist4U found that, on the Royal Mile, there are 123 stores that it classed as unhealthy. With 546 food-related stores in total in a 500-metre radius, this equates to 22.53 per cent being non-healthy, the Daily Record reports. Obesity experts at Chemist4U investigated which high streets are the unhealthiest by analysing the most populated cities to see where people are most likely to be affected. They noted the number of unhealthy stores and outlets within a 500-metre radius of each one, including fast food outlets and confectioneries. According to the results, Buchanan Street in Glasgow is the most unhealthy high street in Scotland. The iconic city street also placed at number seven on the UK-wide list. The experts found that 28.37 per cent of stores in a 500-metre radius of Buchanan Street are "unhealthy". According to Chemist4U, there are 99 stores with poor diet options and 349 food-related stores overall around the street. Elsewhere, topping the overall list as the unhealthiest high street in the UK is St. Peter's Street in Derby. Within a 500-metre radius, it was found that 46.1 per cent of stores were classified as "unhealthy". Head of Pharmacy at Chemist4U Jason Murphy commented: "It's easy to fall into unhealthy eating habits when fast food and sugary snacks are so readily available on our high streets. "While the occasional treat is fine, regularly relying on processed foods can have long-term effects on your weight, heart health, and energy levels. "Small, sustainable changes, such as swapping fizzy drinks for water or choosing grilled options over fried, can make a real difference. If you're unsure where to start, speak to your pharmacist." To put together their list, the experts narrowed down the results to focus on the most populated cities in the UK. They then counted the number of unhealthy points of interest such as fast food outlets, ice cream parlours, vending machines, confectioneries, bakeries, tortilla shops, and convenience stores. To determine the percentage of unhealthy stores surrounding each high street, they calculated the ratio of unhealthy points of interest to total food-related points of interest. This percentage allowed Chemist4U to identify the high streets with the highest concentration of unhealthy establishments. See below for the list of the UK's 19 unhealthiest high streets. The former BBC and Sky News presenter Dermot Murnaghan has been diagnosed with prostate cancer. The 67-year-old announced his diagnosis on social media, saying it was one of stage four advanced cancer. Murnaghan said he was 'fortunate to have a simply outstanding medical team' looking after him that was 'administering the best possible care with expertise, compassion and sensitivity … I'm responding positively to their excellent treatment, and feeling well.' He added: 'Needless to say, my message to all men over 50, in high risk groups, or displaying symptoms, is get yourself tested and campaign for routine prostate screening by the NHS. 'Early detection is crucial. And be aware, this disease can sometimes progress rapidly without obvious symptoms.' Stage four prostate cancer means that the disease has spread into different parts of the body including nearby organs, such as the back passage or bladder, nearby lymph nodes and other parts of the body outside the pelvis, such as the bones, lungs or liver, according to the Cancer Research UK website. Murnaghan, who spent more than 15 years at Sky News before his departure in 2023, said he was aiming to take part in Sir Chris Hoy's fundraising charity bike ride in September, which will raise awareness and funds for cancer charities across the UK. Hoy, the six-time Olympic gold medallist, announced he had a terminal diagnosis for prostate cancer in October 2024. Murnaghan presents true crime documentary series Killer Britain and the podcast Legends of News. He presented ITV's News at Ten and the BBC Ten O'Clock News – now known as BBC News at Ten – as well as Channel 4 News. Murnaghan also hosted the BBC's quiz programme Eggheads. About one in eight men will get prostate cancer in their lifetime, according to the charity Prostate Cancer UK, and the disease often has no symptoms in its earlier stages.
