The Scottish city has been transformed into downtown New York for filming after becoming an alternate (and cheaper) option to the Big Apple itself.
Tom, 29, smiled as he waved to fans in the superhero's famous suit as he began work on Spider-Man: Brand New Day, set to be released in 2026.
He even appeared to jokingly shoot webs from his suit after first portraying the character in Marvel's Captain America: Civil War in 2016.
While the final casting of the latest instalment in the Spider-Man franchise has not been confirmed, Tom is believed to be starring alongside Stranger Things star Sadie Sink, 23.
Zendaya, 29, will also feature in the film after striking up a romance with Tom on the set of his first feature-length film as the hero, Spider-Man: Homecoming in 2016.
Tom's appearance in Glasgow comes amid a very busy time for the actor after he was named the new face of Prada's Paradigme Fragrance — and posed with some adorable puppies to promote it.
He also recently addressed speculation that he could be stepping into the shoes of James Bond.
Tom's name has been bandied about along with other high-profile stars as a potential new star of the James Bond franchise.
But Tom Holland didn't give any hints about the speculation when he was asked about it in a video posted Thursday.
A screenwriter for the highly anticipated film was also announced on Friday.
While joining chef Gordon Ramsay on his YouTube channel to make fried chicken sandwiches and promote his non-alcoholic beer brand, Tom acknowledged the 'speculation' swirling around him.
'Listen, there's speculation at the minute. We'll keep it to a minimum for now,' he said with a sheepish grin. 'We'll get there one day.'
'Every young British actor, it's the pinnacle of working in our industry,' Tom continued. 'I already consider myself to be the luckiest kid alive. I could never have dreamed to have the career that I've had.'

The next Bond film is coming into focus after the announcement on Friday that Peaky Blinders creator Steven Knight would be joining Villeneuve behind the camera to write the next James Bond film.
While speaking to BBC Radio 5 Live Breakfast, Knight said: 'It has always been on my bucket list and it's fantastic to be invited to do it — I can't wait to get started.'
The movie will be overseen by Amazon MGM Studios, after producer Barbara Broccoli gave up creative control of the Bond franchise.
Even though the creative team is falling into place for the next Bond film, there haven't been any indications of who will replace Craig — or which actors are being considered.
The producers are reportedly looking for a considerably younger actor to take over the role after Craig — who is 57 — presented a more weathered, mortality-focused version of the beloved character.

The huge armoured vehicle sped down the street being pursued by three NYPD police cars with red and blue lights flashing. The vehicle appeared to be 'firing' one of its prop guns, with light being seen coming from the weapon as it soared along the road. Two other cars were speeding behind the tank, one carrying a device used to trigger the blast. As the speeding police cars approached a junction, a huge fireball erupted sending out a ferocious boom as part of the stunt. The pursuing cops cars were sent screeching past the flames, with one weaving and skidding as part of the scene. Another explosion was triggered as the chase moved further down Bothwell Street. The blasts were triggered by devices attached to two motors involved in the chase. The stunt cars had boxes attached to long metal beams running behind them. The Met Office said: 'Gusts of 50 to 70 mph are expected, and are likely to reach 80 to 90 mph on exposed coasts, hills and bridges.' The Met Office has warned that Storm Floris is expected to bring gusts of up to 90mph tomorrow morning. The amber alert for "unseasonably strong and disruptive winds" is in place from 10am until 10pm, covering the central belt, Tayside, Grampian, Strathclyde, Lothian Borders and Highlands. It's the second-highest warning possible, made more dangerous by the fact that trees are still in leaf and more liable to collapse. 'Western coastal areas will see the highest gusts between late morning and early afternoon, the strongest winds then transferring to northeastern Scotland by late afternoon/early evening.' Filming for the new Spider-Man movie has already seen several intense stunts being captured this weekend as locals gathered to watch the action. Earlier today, Spider-Man stood on top of a tank as it flew down Bothwell Street in a chaotic car stunt. TOP FILMING LOCATION THE flick has brought movie-fever to Glasgow once again, with some punters dressed as Spider-Man putting in an appearance. Holland, 29, returns as the titular character, with real-life girlfriend Zendaya, 28, as Michelle Jones "MJ" Watson, Jon Bernthal, 48, portraying The Punisher, and Charlie Cox, 42, as Daredevil. Filming is set to move further along into Merchant City after Wednesday, August 13, with scenes are also expected to be shot on Glassford Street and Virginia Street. It is just the latest lot of filming taking place in Glasgow. Earlier this year, Hollywood hunk Glen Powell filmed scenes for sci-fi flick Ghost Writer on Bothwell Street. It is his second time in the city in a year, as he filmed on the exact same street for the remake of 80's hit Running Man in November. A huge scaffolding structure safety rig was set up around the tank for Spider-Man to be harnessed in as he stood on the roof. Three New York-style police cars followed in hot pursuit as they sped down the road with their lights flashing behind him. And on Saturday, a huge chase scene saw cars thrown into the air as a huge crowd watched. A black film car raced ahead of the tank as it rumbled down the road, and a taxi and a van attached to hydraulic ramps were thrown into the air as the war machine sped past. A high-tech drone with a camera attached was zooming overhead to get aerial shots of the stunt as the tank came to a screeching halt at the bottom of the street. Glasgow now resembles the Big Apple for Holland's latest MCU appearance. Historic street signs have been covered with decor from Manhattan, while hot dog carts and newsstands are scattered among the streets. Movie vehicles are also being stored on West Campbell Street. The road has been lined with an ambulance and New York Fire Department pick-up truck, as well as scores of iconic New York yellow taxis and NYPD police cars. Glasgow City Council told the Scottish Sun that production crews would make the decision on whether filming would go ahead. XM2 could not comment when approached by the Scottish Sun.
