They were stopped after they used the boat to travel out to sea where it is believed a cargo vessel had dropped off the cocaine.
The men are suspected of picking up the drugs to bring them to shore where they would be transported to an onward location.
The NCA said in a statement that they are investigating potential links with cargo vessels in the area at the time of the incident.
"This is a fast-moving, intelligence led investigation by the NCA, with the support of partners including Border Force, the South West Regional Organised Crime Unit, Avon and Somerset Police and Devon and Cornwall Police,' NCA Regional Head of Investigations Gavin Webb said
"The NCA is confronting and dealing with at-sea drugs drop-offs as a priority, focusing on protecting the public, and ensuring illegal drugs do not make it to UK shores.
"Our investigation is ongoing and those arrested remain in custody."

Gang who smuggled €60m of cannabis into Ireland and Britain ‘dismantled', say Spanish police

A conspiracy to smuggle cannabis from Spain to Ireland inside bags of animal feed has been uncovered as part of a policing operation led by the Spanish police and involving gardaí and the UK's National Crime Agency (NCA). As well as 13 arrests in Spain following raids in Madrid, Malaga and Seville, one man was detained by An Garda Síochána in Co Louth. The volume of drugs moved through the network's smuggling routes, based on comments by Spain's Policia Nacional, are valued at about €60 million. Spanish police said that at their end of the operation, involving 12 searches, they also seized almost €100,000 in cash, 630 marijuana plants, a firearm and three detonators. Spanish police say they've carried out raids, arrested 13 people (video) re smuggling of cannabis to Ireland, since 2019, inside bags of animal feed Also one arrest in Ireland - Co Louth In Spain, crops of cannabis have been found, a gun, cash, assets seized etc — Conor Lally (@conormlally) The international policing operation started in January, resulting in arrests in Ireland and Spain and the seizure of 300kg of herbal cannabis valued at almost €6 million. Spanish authorities say herbal cannabis had been smuggled from Spain to Ireland and Britain since 2019. They said up to three tonnes of drugs have been shipped in monthly consignments over the past six years. The Policia Nacional said on Thursday its operation with the Garda and NCA had 'dismantled a criminal organisation that shipped marijuana to Ireland hidden in sacks of animal feed'. The international inquiry began when the smuggling operation was discovered in January and involved suspects from Spain, Ireland and Britain, it said. The herbal cannabis was hidden in the animal feed in Spain then transported to Ireland and Britain on pallets. International freight forwarders were used to arrange the logistics. The 12 searches in Spain involved armed police officers breaking their way into properties, carrying out searches and arresting suspects. There were 10 searches in Malaga, one in Seville, both southern Spain, and another in the capital Madrid. The Policia Nacional described the suspect arrested by the Garda in Co Louth as 'another person belonging to the criminal structure'. It was unclear when the arrest took place. As well as a semi-automatic pistol found in one of the searches in Spain, three blank-calibre pistols were also discovered with 11 vehicles and six luxury watches valued at more than €200,000. Electronic devices were also taken for analysis.

Man (23) admits to damaging more than 60 cars while wearing bunny dressing gown

Dillon Fullerton damaged dozens of cars while wearing an Oodie complete with bunny ears. A man today admitted causing thousands of pounds of damage to more than 60 cars in one night. As Dillon Fullerton stood in the dock of Ballymena Magistrates Court, defence counsel Thomas McKeever confirmed the 23-year-old 'is pleading guilty to everything.' On an indictment once described by District Judge Nigel Broderick as the largest he had ever seen Fullerton, from the Station Road in Castledawson, faces a total of 67 charges - 66 of criminal damage and one of attempted criminal damage. The charges arise from a number of incidents on 13 February this year when Fullerton, wearing an Oodie complete with bunny ears, damaged dozens of vehicles. Ballymena Courthouse (Photo by Liam McBurney/PA) News in 90 Seconds - Aug 7th The 23-year-old was captured on CCTV ripping windscreen wipers from vehicles, cracking and smashing windows, scoring paintwork and breaking wing mirrors off multiple cars and vans. Giving evidence at the first appearance a police officer told the court it was around 4.30 that Thursday morning when police received a report of a male causing damage to vehicles in the Grange Road area of the town. Officers who responded received further reports of criminal damage and when they arrived at Rosses Avenue, they spotted a man wearing an orange Oodie, in the process of trying to rip windscreen wipers from a vehicle. Fullerton was arrested and taken to custody and while he was being held, police 'received numerous reports' of vehicles being damaged. The court heard that during police interviews, Fullerton 'made partial admissions' but also gave officers 'non-sensical answers,' claiming that 'the window wipers were drugs and he needed drugs.' At the first appearance, District Judge Nigel Broderick commented that 'I do not think I have ever seen a charge sheet as long as this'. Today, he said he would not be dealing with the case until the Probation Board had compiled a pre-sentence report. Freeing Fullerton on bail, he adjourned the case to 25 September.
