New case study reveals common virus could cause skin cancer

Skin cancer is typically caused by damage to the skin's cells from ultraviolet radiation. But a recent case study has just shed light on another potential cause: human papillomavirus.
The report, which was published in the New England Journal of Medicine, focused on the case of a 34-year-old woman who had been diagnosed with over 40 squamous cell carcinomas (SCC). This is the second most common type of skin cancer.
The woman also had many wart-like growths in her mouth and on her skin. These were attributed to a human papillomavirus (HPV) infection.
Human papillomavirus is a common group of viruses that can infect the skin and other parts of the body. While HPV often does not cause any problems or symptoms in most people, in some cases it can cause warts and is even linked to certain types of cancer – such as cervical cancer.
The woman in the latest report was referred by her doctor to the team of researchers who conducted the case study. She had already undergone multiple surgeries and rounds of immunotherapy to remove a large squamous cell carcinoma that repeatedly grew back on her forehead. The patient's doctor believed this might be due to a condition that made it more difficult for her immune cells to fight off the tumours.
The researchers performed a genetic analysis on this recurrent tumour to understand why it continued to grow back. Under normal circumstances, SCC tumours have a genetic signature that shows their mutations were caused by ultraviolet radiation. These mutations usually drive their growth.
However, this patient's cancer didn't have these signature mutations. Instead, the researchers found that the HPV infection living on her skin had integrated itself into the DNA of the tumour on her forehead. It seemed that it was the virus that was actually driving the cancerous growth.
There are more than 200 different types of HPV, only a few of which have been associated with cancers. HPV19, which infects skin, had not previously been linked to cancer. But in this case, it had gone rogue and caused the carcinoma.
Unique case
This recent case study is unique, it should be said. There were many factors that made it possible for the HPV infection to drive the recurrent growth of skin cancer.
The patient had a long history of health problems beginning in early childhood. This had brought her to the attention of researchers who were studying people who had problems with their immune system. A 2017 case report on her revealed that she had inherited mutations in two genes that play a role in immune function.
One of the mutated genes was ZAP70, which is involved in the normal function of a type of immune cell called a T-cell. This cell plays an essential role in helping the body successfully fight infections.
Inherited changes in ZAP70 that prevent it from working were previously known to cause a condition called severe combined immunodeficiency. This condition is usually diagnosed in infancy and, if not treated with a stem cell transplant, leads to death within the first couple of years of life. Being in her late 20s at that time, the woman became the oldest patient ever to be diagnosed with a ZAP70 immune condition.
The second mutated gene, RNF168, is involved in repairing damage to DNA.
The new team decided to investigate whether it was the unique combination of mutations in both genes that was allowing the HPV infection to cause cancer. However, they concluded that the mutated RNF168 gene was a red herring.
The research team found that the patient's RNF168 mutation was relatively common in the wider American population and wasn't linked to any health issues. Further investigation of her cells also revealed that her DNA repair processes were functioning normally.
They then moved on to the ZAP70 gene. Here, they found that although the patient's ZAP70 gene was mutated, it still partly worked. This explained why she hadn't succumbed to severe combined immunodeficiency in childhood. However, the mutation still made her immune system less effective. So because her T-cell response wasn't fully functional, her body was unable to recognise and eliminate HPV-infected cells.
After receiving a stem cell transplant that replaced her immune cells with fully functioning ones from a donor, the woman made a complete recovery. The new T-cells were able to recognise and destroy the HPV-infected cells, including the skin cancer. Hopefully, she will now remain cancer-free for years to come.
Immune health and cancer
This story highlights how important our immune system is in protecting us against cancer. Without it, even innocuous viruses that usually harmlessly co-exist on our skin can drive the formation of aggressive cancers.
It also demonstrates how modern genomic technology is transforming our understanding of disease. Without genetic sequencing, doctors would still be none the wiser about why this unfortunate woman had so many aggressive skin tumours.
But this study also raises questions about whether HPV-driven skin cancer could be a wider, previously unrecognised problem. The authors suggest that in the future, patients with aggressive and recurrent squamous cell carcinomas should be profiled for T-cell function and the presence of HPV infections. Like the woman in this story, they too might benefit from immune-boosting therapies to treat their cancers.

The US health and human services (HHS) secretary, Robert F Kennedy Jr, has fended off an attack by conservative firebrand and Donald Trump influencer Laura Loomer by issuing a statement of fealty to the president which calls it 'a flat-out lie' that he is running for the White House in 2028. Kennedy, 71, had been under pressure since Loomer, 32, expressed concern in a recent Politico interview that Stefanie Spear, a top aide of the HHS secretary, was trying to 'utilize her position to try to lay the groundwork for a 2028 RFK presidential run'. Loomer's vigilante pressure campaigns within the White House have cost a number of Trump administration figures their jobs, including customs and border protection official Monte Hawkins as well as Food and Drug Administration vaccine regulator Vinay Prasad. Hawkins had been accused by Loomer of having an 'anti-Trump, pro-open borders and pro-[diversity, equity and inclusion, or DEI] bias'. And she had labelled Prasad a 'progressive leftist saboteur' before he was later reinstated by the White House chief of staff, Susie Wiles. Loomer told Politico that while she is realistic about neutralizing Kennedy, his deputies were vulnerable. 'I'm not naive enough to think that the president is going to get rid of RFK, but I will say that … there are concerns about some of the staffing decisions over at HHS,' she remarked. A White House official told the outlet that they 'would not be surprised if [Kennedy is] thinking about' running again after his 2024 candidacy prior to aligning himself with Trump. But the official claimed they 'don't think anyone thinks it's a real threat'. Kennedy responded on Friday, saying he would not strive for the presidency in 2028. The Kennedy family scion ran in 2024 for the Democratic party nomination before switching to become an independent candidate – and then cast his lot with Trump. Trump – who in the run-up to his second presidential election victory dismissed Kennedy as a 'radical left liberal' – rewarded him with a cabinet level post as well as his 'make America healthy again' (Maha) mandate. 'The swamp is in full panic mode,' Kennedy Jr said in an X post. 'DC lobby shops are laboring fiercely to drive a wedge between President Trump and me, hoping to thwart our team from dismantling the status quo and advancing [the Maha] agenda.' Kennedy added that the so-called swamp, a Republican term for an entrenched Washington bureaucracy, was 'pushing the flat-out lie that I'm running for president in 2028'. 'Let me be clear: I am not running for president in 2028,' he added. 'My loyalty is to President Trump and the mission we've started.' And he defended Spear. He said 'attacks on my staff, especially Stefanie Spear – a fierce, loyal warrior for Maha who proudly serves in the Trump administration and works every day to advance President Trump's vision for a healthier, stronger America – are proof we're over the target.' Kennedy also offered an overt expression of obeisance to his White House boss and political patron. The crossover of the administration's anti-DEI campaign into healthcare came after an essay in the American Conservative magazine recommended the removal of taskforce members, saying it was embedded 'left-wing ideological orthodoxy'. Among the points it raised was the taskforce's use of term 'pregnant persons' and mention of a 'lasting psychological impact and stigma of enslaved Black women being forced to act as wet nurses'. HHS announced earlier in August it was halting $500m in mRNA vaccine research. And it has also moved to revive a taskforce on childhood vaccine safety, though vaccine injuries are known to be extremely rare. Known as 'Trump's Rasputin' in some circles, Loomer views Kennedy's vaccine skepticism as surging from the left – and not in pure ideological terms. She disputes that he views the issue correctly as a rightwing one, though the two may act in confluence. She has previously labelled Kennedy, in the New York Times, as 'a very problematic person' who 'is running a shadow presidential campaign' from his office. 's mysterious health journey has not only gone viral - it's grabbed the attention of top dermatologists, with one well-known doctor insisting her issue could get worse if not properly treated. The Real Housewives of Beverly Hills alum has claimed that a parasite left her with facial disfigurement, a health issue she's been battling for two years now. She now says her face was left burned and with red patches after she tried using Nair, a hair-removal cream, earlier this month. Dr. Nicholas Perricone, a top dermatologist, spoke to the Daily Mail about Brandi's facial issues while promoting his new book, The Beauty Molecule: Introducing Neuroceuticals, the Breakthrough for Ageless Beauty. The Connecticut-based doctor has not treated or examined Brandi, but he has heard of the issue she's facing, and he said, 'Many people never get diagnosed and there's this low grade inflammation. 'You don't see it, you can't feel it, but it's very bad and it could also park itself in the central nervous system, which is not good.' When asked what her mysterious health condition could be, the doctor - who's known for being a pioneer of the anti-inflammatory movement - suggested, 'There's something we call a differential diagnosis. 'But the point is this, that anything that's going to create inflammation in the body is bad for us. It accelerates the aging process.' Dr. Perricone, who spoke to the Daily Mail at Impact Wealth Magazine's Annual Hamptons Celebration and Cover Party on August 10, suggested that the reality star should follow an 'anti-inflammatory diet,' and take 'pharmacologic agents' (certain substances used in medicine to treat diseases) that are 'very mild and very effective.' 'It's got to be taken care of. You just can't let it go... There's no excuse,' he insisted. 'The medications are so easy, they're benign, and of course of a few weeks makes all the difference in the world.' When asked how long it normally takes to fix what appears to be her problem, he responded, 'It doesn't take a long time. It really doesn't. 'You have to be on a proper dose [of pharmacologic agents], but you also follow other things that are important.' 'What's important about this is people have to just understand that [people are] in control, and so we take care of things,' he continued. 'If it's a medical problem, get it done. Anti-inflammatory diet [or] lifestyle is really amazing.' Dr. Perricone is even offering to help the 52-year-old get back to normal, saying he'll 'definitely' reach out to her to help because he believes the solution is 'so simple and there's no reason for this.' Dr. Perricone spoke with the Daily Mail at Impact Wealth Magazine's cover party for Stephen Ross in the Hamptons on August 10 (pictured with the Governor of Georgia Brian Kemp) A doctor who has notably reached out to the former model to assist with her health journey is Dr. Terry Dubrow, the husband of Real Housewives of Orange County's Heather Dubrow and star of E!'s Botched Presents: Plastic Surgery Rewind. Dr. Dubrow biopsied parts of her face earlier this year and said that it can take 'weeks' to get the results. They have yet to be disclosed to the public. In a recent interview with the Daily Mail, the Newport Beach plastic surgeon said that her diagnosis is 'very complicated' and that there's not an exact answer. 'I can tell you, I think she's on a path with some real experts right now that may help her,' he shared. This came just days after she made her priorities crystal clear while delivering a wild health update on her mysterious facial disfigurement. Brandi confessed that she couldn't wait to make out 'with a hot boy or girl' as she listed all she was looking forward to once she overcame her health ailment. She made the revelation as she publicly thanked Dr. Michael Scoma, an infectious disease immunology expert who 'delivers proactive treatments to Long COVID and [Myalgic Encephalomyelitis] ME patients.' She blamed her facial disfigurement on the COVID-19 vaccine and a mysterious facial parasite caught in Morocco - which is something her The Real Housewives Ultimate Girls Trip co-star Vicki Gunvalson has also claimed. The RHOC alum recalled during an interview on SiriusXM's Jeff Lewis show last month that the cast 'all got sick' when they went to Morocco to film. She told host Jeff Lewis, 'My stomach's bloated, I don't feel good. I feel tired.' Vicki added: 'Brandi is in so much pain. She's got a very large parasite... It moves. It goes into her mouth. It's bad.' The star revealed she's been taking Ivermectin (medication primarily used to treat certain parasitic infections in humans) 'twice a month' in order to kill the parasite. She's also been doing colonics treatment 'three times a week' and has taken medicines like 'Duralax' and 'Metamucil old lady stuff' to help her gut issues since she claimed the parasites have caused constipation as well.
