6,000-year-old skeletons found in Colombia have unique DNA

Scientists studying ancient human remains uncovered in Colombia have found that the people they were researching have no known ancestors or modern descendants.
In a study published May 30 in the journal Science Advances, a team of researchers reported on the genetic data of 21 individuals whose skeletal remains were found in the Bogotá Altiplano in central Colombia, some of whom lived as long as 6,000 years ago, that belonged to a previously unknown population.
Previous studies have proven the existence of two lineages, northern Native American and southern Native American, which developed after people first arrived on the continent across an ice bridge from Siberia and started to move south.
The latter split into at least three sub-lineages whose movements have been traced in South America, but scientists have not yet ascertained when the first people would have moved from Central America to South America.
The study helps to map the movements of the first settlers, who would have been nomadic hunter-gatherers, study author Andrea Casas Vargas, a researcher at the Universidad Nacional de Colombia, told CNN on Wednesday, but it also found that they have unique DNA.
Casas Vargas said the team were 'very surprised' to find that the remains did not share DNA with other people in the genetic record.
'We did not expect to find a lineage that had not been reported in other populations,' she said.
Casas Vargas underlined that Colombia's position as the entry point to South America makes it significant to our understanding of the population of the Americas.
'This study is very important because it is the first to sequence complete genomes in ancient samples from Colombia,' she said,
The results raise questions 'as to where they came from and why they disappeared,' said Casas Vargas.
'We are not certain what happened at that time that caused their disappearance, whether it was due to environmental changes, or if they were replaced by other population groups,' she added.
Further research will hopefully provide some answers, said Casas Vargas.
'Our next investigations will look for other archaeological remains from other regions of the country and analyze them at the genetic level and complement this first discovery,' she said.
Christina Warinner, a professor of scientific archaeology at Harvard University, told CNN that Colombia 'is a key region for understanding the peopling of South America… but until now it has been a blank spot in ancient DNA studies of the Americas.'
'This study highlights the deep history of population migration and mixing in the formation today's populations, and points to Central America as a key region that influenced the development of complex societies in both North and South America,' she added.

Photo courtesy Institute for Bioengineering of Catalonia Aug. 11 (UPI) -- A team of Spanish researchers announced Friday they have for the first time recorded video of a human embryo implanting itself in a simulated uterine wall, revealing never-before-seen details of how 5-day-old embryos carry out the mysterious process. Using advanced microscopy techniques allowing the scientists to record the human embryo in full color and 3D, the "astonishing" videos provide the first-ever, real-time glimpse of the implantation process and have provided key insights into how it actually works, they said. Researchers from the Institute for Bioengineering of Catalonia and Dexeus University Hospital in Barcelona, Spain, said the videos reveal for the first time that embryos exert "considerable force" and employ digging traction as they "invade" the uterine tissue, becoming completely integrated with it. The findings, published in journal Science Advances, found crucial differences between how mouse and human embryos move in connecting to the uterus wall, the authors said. An "ex vivo" platform they developed using an artificial uterine matrix made of gel and collagen which allows for implantation outside of a human uterus made the videos possible. The system could have a "significant impact" on efforts to counter infertility and help those who are unable to conceive naturally, they predicted. Failure of the implantation process is the main reason behind the relatively low effectiveness of assisted reproductive technologies, such as in-vitro fertilization, in which embryos are conceived in a lab and then transferred to the womb. Implantation occurs in only 25% to 30% of transferred embryos -- whether conceived in vivo or in-vitro -- with embryo quality cited as the most significant feature affecting implantation. "We've opened a window into a stage of development that was previously hidden," the co-authors said in a statement to UPI. "After Day 5, when an embryo has 100 to 200 cells, it must implant, but until now, doctors couldn't observe it again until an ultrasound weeks later. "With our system, we can test culture conditions or compounds that might improve implantation." For example, the scientists say they have already developed a protein supplement that can be used in clinics to enhance implantation rates, available through their spin-off company Serabiotics and in collaboration with the Spanish pharmaceutical major Grifols. "In short, this is a new tool for extending embryo observation and optimizing conditions for success," they said. The videos show a donated human embryo powerfully pulling on the uterine matrix and reshaping it as it goes, illustrating the importance of "optimal matrix displacement." Lead author Samuel Ojosnegros, principal investigator of IBEC's Bioengineering for Reproductive Health Group, said the initial real-time look at a human embryo implanting itself was a profound experience for him. "We had some experience making time-lapse movies of mouse embryos, but the first time we saw a human embryo implanting was truly astonishing," he said. "Everything was different, the size, the shape, the behavior. They were stronger, more forceful, digging a hole into the matrix in a remarkably invasive way. Every detail felt unique. "Watching it alive, in action, for the first time was absolutely mind-blowing." Embryo implantation is the "holy grail" of reproduction -- and unlike in the animal world, in humans it can be a problematic process, resulting in about 1 in 6 people around the world having trouble making a baby, noted Dr. Mark Trolice, a professor at the University of Central Florida College of Medicine and founder/director of The IVF Center, a full-service reproductive medicine clinic in Orlando. "Even though scientists have studied this for many years, they still do not fully understand how implantation works or what makes the uterus ready for an embryo," he told UPI. "One big mystery is why a woman's body can grow a baby made from sperm -- which is a 'foreign' tissue -- without rejecting it, as well as the ability to carry a donated egg." The new study, he said, "gives researchers a closer look at implantation. They used an ex vivo model, which means they studied the process outside the body. This let them watch how embryos interact with the uterine lining (called the endometrium) and measure the tiny pulling and pushing forces from both mouse embryos and donated human embryos." The videos showed for the first time that each species makes its own unique pattern of forces during implantation. Trolice noted that while there are "some limits" to the Spanish study, "this work could lead to new ways of adjusting the uterine environment, which might help more embryos successfully implant. "Before any treatment can be used, scientists will need to do human clinical trials. There are also important ethical and legal rules about using human tissues and embryos, which researchers must follow," he added.

Megathrust fault line off Haida Gwaii confirmed through new imaging

Scientists have confirmed that a fault line off the west coast of Haida Gwaii is of the megathrust variety, capable of generating large earthquakes and tsunamis. The conclusion was gleaned through hydrostatic imaging and measurements of the Queen Charlotte Fault where the Pacific tectonic plate meets the North American plate. The finding was published in Science Advances. The images show that instead of the plates sliding horizontally against each other, the Pacific plate is also colliding with and diving under the North American plate, in a process known as subduction. The friction created by subduction is what creates megathrust conditions, according to UBC professor and study co-author Michael Bostock. "For most of its length, the Queen Charlotte plate boundary is very much like the San Andreas fault — one plate is sliding by the other. But at the southern end, along the coast of Haida Gwaii and in particular southern Haida Gwaii, there is a component of convergence. So not only are they moving... side by side, but they're compressing each other." Bostock said the magnitude 7.7 Haida Gwaii earthquake of 2012 led scientists to understand there was a significant component of "under thrusting" or subduction along the fault line. The new research has now mapped it out in greater clarity. At approximately 300 kilometres long, the Queen Charlotte subduction zone is shorter than the 1,000-kilometre-long Cascadia subduction zone that runs from northern Vancouver Island to Northern California. The Cascadia subduction zone is where the "Big One" is expected — that being the oft-forecasted megathrust earthquake predicted to register magnitude 9 or higher. Experts believe it's just a matter of time before the Big One hits considering that stress between the subducting Juan de Fuca plate and the North American plate has been building up since its last major earthquake in the year 1700. In contrast, Bostock believes Haida Gwaii is not susceptible to a megathrust quake anytime soon. "In my opinion it's very unlikely we'd have another megathrust earthquake off Haida Gwaii within the next 100 years. We're not building stress up fast enough for us to have another one like we just experienced 12 years ago," he said. The western coast of Haida Gwaii has been the site of four earthquakes of 7.0 magnitude or more in the past century, including the largest in recorded Canadian history in 1949 that registered 8.1, according to Earthquakes Canada. University of Victoria professor and study co-author Kelin Wang said the new research helps understand and plan for a megathrust. "Certainly it will provide important input for our building codes and the type of ground shaking we would expect, also in terms of tsunami evacuation and preparedness," he said.

Myths, folk tales, and negative media coverage mean people often link these flying mammals to vampires or blame them for disease outbreaks. But bat populations around the world are in decline, and without them, ecosystems lose key benefits like pest control, pollination and seed dispersal. Through education, research and advocacy, Rodrigo Medellín, a senior professor of ecology at the University of Mexico, has made it his mission to change the way people see these animals. Medellín's fascination with bats began at the age of 13, when he held one in his hands for the first time. 'That's when I decided to dedicate my life to their study and protection,' he says. Since then, caves have become his happy place. 'The peace, the darkness, the silence apart from the bat squealing. I feel relaxed in a cave and all I do is I try to convey this feeling to the people that are with me,' says Medellín, who is part of the Rolex Perpetual Planet Initiative and has won multiple awards for his work. He also founded of the Latin American Network for Bat Conservation, and Global South Bats, a network of bat scientists. There are over 1,400 bat species, which account for around a fifth of all known mammal species across the globe. They are the only mammals capable of powered flight, rather than just gliding, which allowed them to spread across the globe. They use highly developed echolocation to navigate and find prey at night, with some able to sense objects as small as human hair. Although certain species live for over 30 years, bats reproduce slowly – typically one pup per year. This makes it difficult for populations to recover from decline. 'They are very mysterious. Many people fear them, attack them, or despise them. They are probably the most unfairly treated animals on Earth,' says Medellín. Bats are often seen as symbols of evil and darkness, largely due to their association with vampires and the supernatural in Western folklore. Throughout history in Christian Europe, the bat has been associated with the devil, evil spirits, and witches. Eastern cultures tend to view them much more positively. For instance, in Chinese culture, they are considered symbols of luck and happiness. Still, the perception of bats has worsened with the emergence of Covid-19, which some believe originated in bats and they are often viewed negatively due to their association with diseases. 'Bats are certainly not fuller of diseases than your dog or cat. That has been horribly exaggerated,' Medellín explains. One of the most impactful services bats provide is pest control. Medellín notes that just one species along Mexico's northern borders numbers up to 30 million individuals, collectively devouring about 300 tons of insects every night. But they do much more than keep insects in check. Fruit-eating bats play a crucial role in spreading seeds as they fly long distances in search for food. 'By dropping seeds far from the parent tree, they help regenerate forests, maintain plant diversity, and support the life cycles of countless other organisms. The restoration of forests relies very heavily on bats,' Medellín says. Bats are also key pollinators for a wide range of plants, according to Medellín, including agave, the plant used to make tequila. Despite their critical ecological roles, populations worldwide face many challenges, most of which are caused by human activity. Habitat loss, wind turbines, pesticide use, and most recently, white nose syndrome – a fungal disease – have all taken a serious toll. As a result, many species are now listed as endangered or threatened. 'Imagine what happens if we lose bats overnight,' Medellín says. 'Without them, crops would be ravaged by hungry insects, and mosquito populations would surge, drastically changing our way of life.' He is committed to convincing people that bats are important for their everyday wellbeing and believes that greater awareness could transform attitudes and turn fear into fascination. Through initiatives like bat-friendly agave farming, long-distance migration tracking and international conservation networks, he's working to protect bat populations while highlighting their role in pollination, pest control and ecosystem health. 'I give people facts, images, evidence, and automatically they fall in love with bats,' he says. 'In my experience, anyone who had any contact with bats maintains that contact and starts expanding it.' 'If there is anybody who is still afraid of bats, I would like to invite you to learn a bit more,' he adds. 'They are going to win your heart.'
