Aussie researchers and ASX biotechs driving paediatric drug breakthroughs
CSL is a global leader in developing therapies for children with haemophilia and other serious blood disorders
Neurotech International is at forefront of developing cannabinoid-derived therapies for paediatric neurological disorders
Australia may lack the size and funding of larger global biotech hubs, but it consistently punches above its weight in paediatric drug discovery.
From life-altering treatments for rare genetic conditions to vaccines that have reshaped public health, Aussie researchers and ASX-listed companies have and continue to deliver breakthroughs improving and in some cases saving young lives globally.
Among these advancements is Australia's largest biotech CSL (ASX:CSL), which has become a global leader in developing therapies for children with haemophilia and other serious blood disorders.
Its paediatric haemophilia products — including Afstyla and Hemlibra — allow young patients to live more freely and safely.
CSL also played a pivotal role in bringing Gardasil to the world – the revolutionary human papillomavirus (HPV) vaccine co-developed by former Australian of the Year Professor Ian Frazer at the University of Queensland.
The vaccine is helping to protect millions of girls and boys from HPV-related cancers.
In the 1990s Dr Sandra Anderson and colleagues at Sydney's Royal Prince Alfred Hospital (RPAH) started research into how inhaled mannitol can be used to diagnose asthma.
Their work resulted in development of two new pharmaceutical products:
• Aridol – to identify bronchial hyperresponsiveness and assist in asthma diagnosis.
• Bronchitol – to treat patients with bronchiectasis and cystic fibrosis (CF) by helping clear mucus from their lungs.
In 2001 Pharmaxis now Syntara (ASX:SNT) licensed patents for respiratory products from the Central Sydney Area Health Service (which included RPAH).
Pharmaxis funded later stage clinical trials necessary to get Aridol and Bronchitol, which are now available world-wide, approved. Its shares peaked in 2007 at over $4 a share – for a roughly $800 million market cap – mainly on its Bronchitol hopes.
The company announced sale of its mannitol respiratory unit, which included Aridol and Bronchitol, in October 2023 along with its rebrand to Syntara and change of focus to haematological malignancies.
On the ASX, several healthcare players are cooking up the next-gen of paediatric therapeutics.
Neuren targets various rare diseases
In 2023 Neuren Pharmaceuticals (ASX:NEU) became the darling of the Aussie bourse after its US partner Acadia Pharmaceuticals secured US Food and Drug Administration (FDA) approval for trofinetide to treat Rett syndrome.
Marketed as Daybue, it is the first drug to treat the rare neurological disorder which emerges in infancy and is a profoundly debilitating neurological condition affecting predominately girls.
And Neuren hasn't stopped there. It's also advancing NNZ-2591 in clinical trials for a suite of other rare paediatric syndromes – including Angelman, Phelan-McDermid, and Pitt-Hopkins — for which there are currently no approved treatments options.
Neuren is also developing NNZ-2591 to treat neonatal hypoxic-ischemic encephalopathy (HIE).
"HIE is a devastating type of brain injury caused when a baby's brain does not receive enough oxygen or blood flow before or shortly after birth," CEO and managing director Jon Pilcher told Stockhead.
"It is one of the leading causes of neonatal death and neurodevelopmental disability.
He said Neuren aimed to provide a highly differentiated form of treatment continuing beyond the neonatal intensive care unit to target the long-term outcomes for these children.
"Neuren's whole business is aimed at trying to improve the lives of the children and their families that are so impacted by all of these conditions," he said.
"The whole team is highly motivated by that mission, building on scientific foundations from Australia and New Zealand."
Argenica Therapeutics (ASX:AGN) has been targeting neonatal HIE for nearly a decade, alongside other acute neurological conditions such as stroke and traumatic brain injury, with its novel therapeutics aimed at reducing brain tissue death.
Its first published paper out of the Perron Institute in Western Australia was in 2018. The study was run by Dr Adam Edwards, who leads Argenica's preclinical neonatal HIE program.
Both Argenica's therapeutic candidates ARG-006 and ARG-007 are being investigated for their respective safety and efficacy profiles in HIE, offering different therapeutic options for the treatment of the condition.
The FDA has granted both ARG-006 and ARG-007 ODD and Rare Paediatric Disease Designations (RPDD), qualifying Argenica for significant incentives.
Neurotech tackles paediatric neuro disorders
Neurotech International (ASX:NTI) is at the forefront of developing cannabinoid-derived therapies for paediatric neurological disorders, led by its proprietary oral formulation, NTI164.

Following the path of successful companies like Neuren, Neurotech is advancing treatments for Rett syndrome, having completed a phase I/II trial with promising results and an extension study currently underway.
NTI164 has been granted orphan drug designation (ODD) by both the US FDA and the European Commission, reinforcing its potential as a differentiated therapy in this rare disease.
Beyond Rett, Neurotech has completed multiple paediatric clinical programs with NTI164, including:
• A phase I/II and phase II/III trial in Autism Spectrum Disorder (ASD)
• Phase I/II trials in PANDAS/PANS (Paediatric Autoimmune Neuropsychiatric Disorders and Acute-Onset Neuropsychiatric Syndrome)

Dimerix tackles leading cause of kidney failure in children
Dimerix (ASX:DXB) is focused on the rare kidney disease focal segmental glomerulosclerosis (FSGS) with its lead phase III drug candidate DMX-200.
CEO and managing director Dr Nina Webster told Stockhead FSGS affects both adults and children damaging filtering units of the kidneys, leading to permanent damage and eventual organ failure.
'FSGS is one of the leading causes of kidney failure in children, with 20% of all presentations of nephrotic syndrome in paediatric patients caused by the disease,' she said.
'This is a disease for which there are no treatments available anywhere in the world and 60% of those who do get a transplant get reoccurring disease in the transplanted kidney.
'This is a very poor prognosis and in particular for children who will live with life-long dialysis.'
Dimerix's phase III study of lead drug DMX-200 in FSGS is titled Angiotensin II Type 1 Receptor (AT1R) & Chemokine Receptor 2 (CCR2) Targets for Inflammatory Nephrosis – or ACTION3 for short.
The second interim analysis of its ACTION3 trial is forecast in around mid-CY25.
DMX-200 has been granted ODD in the US and Europe, as well as the equivalent Innovative Licensing and Access Pathway (ILAP) designation in the UK.
Mesoblast gains world-first approval for treatment
Mesoblast (ASX:MSB) has developed Ryoncil (remestemcel-L) for the treatment of steroid-refractory acute graft versus host disease (SR-aGvHD) in paediatric patients two months and older, which is the first FDA approved mesenchymal stromal cell (MSC) therapy.
Resulting from bone marrow transplants, GvHD is a life-threatening condition with high mortality rates.
Mesoblast said in the US about 10,000 patients undergo a bone marrow transplant annually with donated tissue, 1500 of whom are children.
About half of them will develop GvHD and 50% of these won't respond to steroids.
Ryoncil applies to patients resistant to steroids, the standard-of-care treatment.
Mesoblast is also targeting the children's disorder hypoplastic left heart syndrome, for which it has rare paediatric disease designation from the US FDA for its allogeneic cell therapy Revascor.

Women hospitalised with lung injuries after dry needling at Bendigo massage clinic
Two Bendigo women have been treated for lung injuries after receiving dry needling treatments at a local massage clinic. Dry needling is the practice of inserting small needles into tight muscles to try and relieve them. It is different to acupuncture, which has lengthier and more rigorous training requirements. The ABC understands two women had massages at Miracle Health Massage in Eaglehawk on separate occasions in July and had dry needling included in their session. One of the women, Julie — her name changed for privacy reasons — said she began experiencing severe shortness of breath at the end of her appointment. She said staff at the massage centre called an ambulance and she was taken to Bendigo Health. Julie said doctors confirmed she had a pneumothorax, or collapsed lung, which required the insertion of a chest tube to drain her lung. She said doctors advised her not to drive or fly for at least 10 weeks. Bendigo Health said it could not comment on the matter due to patient privacy. A spokesperson for the Victorian Health Complaints Commissioner (HCC) confirmed to the ABC it was aware of two complaints about the same Bendigo centre. "The HCC has recently received complaints regarding pneumothorax as a result of dry needling and we are currently looking into it," the spokesperson said in a statement. "We do not hesitate to take action if we believe there is a risk to public safety. "If anyone has experienced pneumothorax after dry needling, or has information about unsafe practices, we encourage them to contact us directly." Zaparas Lawyers said it was exploring legal action on behalf of one of the women. "There is a level of care that members of the community expect will be adhered to and incidents like these should not happen to anyone," said senior associate Georgio Skoumbri. "We are committed to pursuing compensation for all those who have been injured as a result of negligent treatment." Miracle Massage Eaglehawk owner Lin Yang told the ABC she was still investigating the complaint. She said the staff member who did the dry needling was recently qualified but would not be continuing with the practice. Ms Yang said she was yet to speak directly with Julie, who she described as a regular customer. "If the client has any questions I hope we can resolve them together," she said. The ABC understands dry needling injuries are rare, with the HCC receiving low numbers of complaints in the past two years. Royal Australian College of General Practitioners rural chair associate professor Michael Clements said it was "very unlikely and very rare" a small needle, such as in dry needling, could cause pneumothorax. "But as soon as you're putting needles where they are not supposed to go, and penetrating that normal skin barrier, we do have to think about risks like that." Dr Clements said pneumothorax occurred when air escaped into a space between the lung and chest wall. He said it was also not unusual for emergency departments to deal with cases of spontaneous pneumothorax. "It can also occur after chest infections or if they have got underlying lung disease." But Dr Clements said if a person who presented with pneumothorax had had penetrating treatment with needles then doctors would probe that. "This would certainly be high on our list of potential causes," he said. Melbourne myotherapist and a former director of the Association of Massage Therapists, Ian Lim, said he was concerned but not surprised to hear of injuries from dry needling. Mr Lim said the treatment was often confused with acupuncture, which had more lengthy and rigorous training requirements. "[Acupuncture] practitioners must undergo years of formal education and licensing in Australia through the Chinese Medicine Board," Mr Lim said. "I am very conservative with my approach to dry needling and only use it as a last resort because of its risk profile." Massage and Myotherapy Australia chief executive Ann Davey said reports of injuries linked to dry needling were "very concerning". She said the massage industry was self-regulated and anyone could claim to be a therapist. Ms Davey urged the public to check the qualifications of their therapists before attending appointments. "The same principle should apply here. "Being a self-regulated industry doesn't mean clients can't or shouldn't ask to see someone's qualifications."

New ‘super-sized' insect species discovered in Qld rainforest
A giant new species of 'super-sized' stick insect has been discovered in a remote rainforest in North Queensland. The insect, weighing around 44g and measuring 40cm long, is believed to be the heaviest ever found in the country. Footage captured of the incredible insect shows its large wingspan displayed across someone's arm, measuring similar to the length of a small bird. The new species has been named Acrophylla alta – a nod to its high-altitude habitat in the Atherton Tablelands. It's roughly the same size as a small barn owl or a wood pigeon, and is 14g heavier than Australia's heaviest insect; the giant wood moth. James Cook University's Angus Emmott and south-east Queensland scientist Ross Coupland searched for the stick insect after they received a photograph of what they believed was an unknown species. Despite its elusive nature, the pair managed to find a female at an elevation above 900 metres in the Wet Tropics Wold Heritage Area. Professor Emmott said the creature's large size could be an evolutionary response to its cool, wet habitat. 'Their body mass likely helps them survive the colder conditions, and that's why they've developed into this large insect over millions of years,' said Professor Emmott. 'The remote habitat was probably also why it had remained undiscovered for so long.' It's restricted to a small area of high-altitude rainforest, and it lives high in the canopy. So, unless you get a cyclone or a bird bringing one down, very few people get to see them,' he said. The stick insect's distinctive eggs also helped scientists identify it as a new species, as no two species' eggs are the same. 'Every species of stick insect has their own distinct egg style,' said Professor Emmott. 'They've all got different surfaces and different textures and pitting, and they can be different shapes.' Two specimens are now being kept at the Queensland Museum in hopes they can aid future research. Currently, the insect's life span is still unknown.

James Cook University medical student allowed to continue studies after domestic violence assault
A Queensland university is under pressure to take disciplinary action against a medical student who has been sentenced for domestic violence assault. The James Cook University student pleaded guilty this week in the Townsville Magistrate's Court to assault occasioning bodily harm. He was sentenced to two years' probation and ordered to pay $500 compensation, with no conviction recorded. The university confirmed a staff member provided a character reference for the student in a personal capacity. The ABC understands the student is now on leave, with the university considering further action in the wake of the court decision. The James Cook University Medical Students' Association released a statement saying it was deeply disturbed at the reports. "There is absolutely no place for violence, abuse or intimidation in the medical profession," the statement read. "No-one in our community should have to experience harm." Townsville nurse Emma Coppens wrote an open letter to the university criticising it for allowing the student to continue their studies in the wake of the court action. Ms Coppens, who spoke with the victim, said she was alarmed the student was on track to graduate as a doctor. "I'm just a nurse, I'm just a mum, but I think I speak for so many people when I say that we've had enough of the soft reactions to these kind of things," Ms Coppens said. The ABC has seen a letter sent to medical students this week by College of Medicine and Dentistry dean Sarah Larkins, referring to a "negative media story" involving a student. "I would like to reiterate the position that there is no place for any form of violence, including domestic and family violence, in medicine or health care," she said. A spokesperson for the university said a complaint was investigated when the matter was first reported to JCU in August 2022, but its outcome was confidential. They said in view of the court decision, the university would continue to respond in accordance with its policies and procedures. A spokesperson for state Health Minister Tim Nicholls said the student was not currently working in "any capacity" for Queensland Health. Queensland Police have been contacted for comment on whether they will seek to appeal the student's sentence.
