Ajax had taken the lead through Anton Gaaei but were pegged back by Thom van Bergen in the second half.
Former Manchester United and Burnley striker Wout Weghorst would have thought his 68th-minute goal was going to be enough for a vital three points.
But it was the ten men of the hosts that struck the heartbreaking goal through Thijmen Blokzijl in the dying seconds of added time.
The result sees Ajax fall to second place and one point behind title rivals PSV.
Henderson and co were the favourites for their 37th title with just five games left of the season.
Ajax needed to pick up just six points to be confirmed as league champions.
However, they have stumbled to defeats against Utrecht and NEC as well as a draw with NEC and Groningen before the final game of the season.
Peter Bosz's side took full advantage of Ajax's slips to eradicate the nine-point gap, having won their previous six matches.
This means that with one game to go, the title is PSV's to throw away.
PSV face 11th-placed Sparta Rotterdam while Ajax will take on FC Twente.

Ajax will have to win and hope PSV slip up as their goal difference is significantly worse.
After Blokzijl's last-gasp goal fans on social media were left stunned by Ajax's drop off.
One posted: "How does Ajax give away a 9-point lead in 4 games? I have no words for it."
A second wrote: "Ajax have bottled it what is happening in the Netherlands."
A third commented: "In rap battles, we call that choking."
A fourth said: "How did Ajax bottle the league title?"
Another added: "As a fan, my head would be on Pluto until the last games bc HOW?"

Footage was captured from the PSV game of the club's supporters and coaches waiting to hear the result of the Ajax match.
Many appeared stressed and expecting the Amsterdam-based club to win but were quickly overjoyed when the late goal went in.
However, the race is not fully over as Ajax manager Francesco Farioli issued a rallying call.
He said: "It's never easy to speak after conceding a last-minute equaliser.
"It's not over yet. We've got one more match to play: let's go.
"We gave it our all, but unfortunately, it wasn't enough. Still, football teaches us that anything can happen."
Henderson, 34, joined Ajax back in January 2024 after leaving Saudi Pro League side Al-Ettifaq.
He has been given the captain's armband but is yet to lift a trophy with the club.

Nick Morgan living the dream with Stern John in St Lucia
The 44-year-old from St Julians has a management CV that features a raft of Gwent clubs but now he is part of St Lucia's set-up, working as an assistant for former Premier League striker Stern John. The 'Piton Boyz' will go up against Advocaat's Curacao, who boast players from the Netherlands' Eredivisie, in the early hours of Saturday UK time and then face Barbados on Tuesday night. Morgan has had the job since the autumn of 2024 after accepting an invite from John, who played in England for Nottingham Forest, Birmingham, Coventry, Derby, Sunderland, Southampton, Bristol City and Crystal Palace. The pair met on a Football Association of Wales A licence course and that led to the Welshman jumping at a chance to experience international football. Morgan initially did that while managing Westfields in Hellenic League Premier Division but he has now left the Hereford club and will devote more of his time to St Lucia, alongside his full-time job as a project manager with energy company SSE. COACH: Nick Morgan in St Lucia (Image: Nick Morgan) 'I'm senior team coach but now I will also be a liaison for the players in the UK, which I couldn't do when I had games on Saturdays,' he said. 'I'll go and watch those who have already committed to us but also work on a big list of those who maybe could in the future. 'Stern came over in May and we went to watch some players because we have seven professionals in the English leagues – the likes of Terell Thomas at Carlisle, Arkell Jude-Boyd at Cheltenham, Chris Forino-Joseph at Bolton.' St Lucia are 165 in FIFA's rankings and the World Cup is not a realistic aim but they did have a good crack at qualification for this summer's Concacaf Gold Cup. They finished second to Curacao in the group stages, that after stunning Advocaat's side 2-1 in their opener. The big aim is to make progress as a nation with Morgan relishing the chance to be part of a professional football environment again. He did that with Newport County AFC under good friend Michael Flynn in 2017/18 – a season that featured the FA Cup games against Tottenham at Rodney Parade and Wembley – but it was a stretch. 'I remember that we had Lincoln away on a Tuesday night and I had to be in Exeter for work at 8am. We got back to the training ground at 3am,' said Morgan, who has a wife Lucy and children Cian and Kayleigh. 'I was committed to that season but it was tough, I've been offered opportunities in full-time football but, with the level that I would have to go in at, I couldn't risk my job in the outside.' Morgan has been part of the set-ups at Newport Corinthians, Newport YMCA, Chepstow, Monmouth, Cwmbran Celtic and Abergavenny, plus he also worked with the Exiles' academy before stepping up to help with the first team. 'I've been in semi-professional football for the last 15 years since I stopped playing and it's always been something running alongside my job, working with some very good Welsh teams before going over to England,' he said. 'Now I've got this opportunity to go over to the Caribbean and represent a country, and I love the togetherness that we have as a squad and staff. 'This gives me the chance to get that experience of a professional football environment with the camps three or four times a year. I love it.'

Virgil van Dijk rules out one transfer when his Liverpool contract expires with the Anfield captain insisting 'I am very clear'
Virgil van Dijk has opened up about his retirement plans and ruled out one particular move before he hangs up his boots. The 33-year-old signed a new two-year contract with Liverpool in April, committing his future to the Reds until the end of the 2026/27 season. That means that the centre-back will be just shy of his 36th birthday when his current deal expires. And as he reported for international duty with the Netherlands, Van Dijk was questioned about how he plans to end his career. The Liverpool captain started his senior career with FC Groningen, having spent his youth career in the Willem II academy. He will play at Groningen's Euroborg stadium for only the second time since leaving the club when the Netherlands host Malta in their latest World Cup qualifier on Tuesday night. Van Dijk appears in good spirits as he trains with the Netherlands ahead of the clash with Malta Ahead of the game, Van Dijk was asked whether he would ever consider a return Eredivisie. But he quickly dismissed such a notion, insisting that he has no desire to finish his career back in his homeland. 'No, unfortunately not,' he said at a press conference. 'I am very clear about that. 'I will not return to the Netherlands. I started here and that was very important to me. 'I am very clear that I do not want to return to the Eredivisie.

Erik ten Hag tried to STOP Man Utd signing Antony but club ‘pushed £86million transfer through at all costs'
ERIK TEN HAG tried to block Manchester United from signing mega-money flop Antony, according to a sensational claim. Antony, 25, joined the Red Devils in 2022 for £85.5million in Ten Hag 's first summer at the club to follow the manager directly from Ajax. 2 The Old Trafford outfit originally did not want to pay as much for the star. However, according to widespread reports, Man Utd only pushed ahead with the deal after being encouraged by Ten Hag to complete it when he lost his first two games in charge. Then Man Utd chief John Murtough submitted three bids to take Antony from the Dutch giants, which was eventually accepted. And after a bright start, Antony failed to live up to his billing and quickly became a scapegoat for Ten Hag's failings as a Man Utd boss. The Brazilian eventually lost his place in the starting XI in his second season before he was sent out on loan to Real Betis halfway through his third season with Man Utd. Ten Hag now manages Bayer Leverkusen and has been linked with the winger once again. But in a stunning claim from Kees Vos, co-founder of Sports Entertainment Group (SEG) who are Ten Hag's agency, he says Ten Hag actually tried to BLOCK the move, with Man Utd being the ones wanting to "push it through at all costs". Writing in the book, Liverpool and the Dutch, Vos claims: "Ten Hag and Vos were not in favour of this and wanted to stop the transfer. "But the club wanted to push through at all costs, because Arsenal was also interested in Antony. "The risk of a stronger competitor would be great. Ultimately, Manchester United paid almost 100 million euros for Antony. 'With you, I started to smile again' - Tearful Antony announces Real Betis exit "Although Ten Hag did not support this transfer, it stuck with him throughout his entire time in Manchester that he took over the attacker from Ajax for this ill-fated amount, who was never able to fulfil his promise." Overall, Antony scored just 12 goals and added five assists in 96 games for the Red Devils and was often ridiculed for his flagrant skills which often yielded no end product. His loan at La Liga club Betis reminded people of skills as he contributed to 14 goals in 26 games. Man Utd chiefs are reportedly willing to take a hit worth more than half of what they originally paid in order to get him off the books, with a figure of around £40m being reported. SunSport have also revealed how Ten Hag's Bayer are considering a move for Jack Grealish after he was told his Manchester City career is over.
