Gene Therapy Partially Restores Sight in Legally Blind Children

A promising study out of the United Kingdom has partially restored the vision of multiple small children born legally blind. The experimental treatment uses gene therapy to resolve a retinal disorder called LCA4, which prevents the eye from distinguishing objects in a person's environment. Though limited, the study suggests that blindness caused by genetic defects could be curable.
LCA4—short for leber congenital amaurosis-4—is caused by a series of mutations in the AIPL1 gene, which is responsible for the function and survival of the retina's photoreceptors. When AIPL1 is defective, the eye can't see; at best, a person can only see vague spots of light. People with LCA4 experience blindness from infancy, with some losing even the ability to perceive light as they age.
Today, it appears gene therapy can help to restore at least some LCA4-related vision loss. In a study at University College London's Institute of Ophthalmology, researchers placed healthy copies of the AIPL1 gene within a virus engineered to target retinal cells without triggering actual illness. (This part isn't particularly new; engineered adeno-associated viruses have been used to deliver DNA to cellular targets for years now.) Once in the retinal cells, the "new" AIPL1 gene replaced the defective version, helping to slow the deterioration of the retina and even develop healthy photoreceptors. The study was published in the February issue of The Lancet.
In an effort to protect and build upon LCA4 patients' extremely limited sight before it was too late, the researchers selected children ages 1 through 3, who experienced severe retinal dystrophy but could still perceive small amounts of light. Each of four children received an injection of healthy AIPL1 in one eye while under anesthesia. Then the team monitored the children's health and vision, conducting different light and color perception tests as the children aged.

All four children showed "meaningful responses" to the therapy—in fact, while vision improved remarkably in every treated eye, each untreated eye lost its ability to perceive light entirely. The children gained the ability to sort different-colored crayons into cups, follow a pen light, locate tiny white objects on a dark background, navigate a normally lit corridor, identify doors, and even draw. According to the researchers, the children have each moved from "legally blind" status to "low vision" status, amounting to a massive quality-of-life upgrade.
Since the completion of this study, the researchers have begun working with a second cohort of seven children who have received treatment in both eyes. Results of this trial aren't quite ready yet, but they must be encouraging, as the team notes they are "exploring the feasibility of making the [treatment] more widely available."

As the industrial food system pushes ultraprocessed foods to every corner of the globe, new research shows that even seemingly healthy packaged meals may fall short of the benefits offered by whole, minimally processed foods. A fascinating new study published in Nature Medicine dove into the real-world effects ultraprocessed foods have on the human body. Following a six-month clinical trial involving 55 volunteers, researchers at the University College London found that eating minimally processed foods was not only potent fuel for weight loss, but quickly led to healthier biomedical readings across the board — a grim indictment of what's inexorably become the norm in global eating. To run the experiment, the team of over 20 researchers split the participants into two groups, each of which followed a strict diet for eight weeks. One group was assigned to eat only minimally processed whole foods, while the other was given the task of eating mainly hyper-processed slop. After the first diet was finished, the volunteers took a four-week break, then switched to the other chow. Given the direct comparison, the findings are noteworthy. Participants on the non-processed food regimen burned more fat, had lower systolic blood pressure, and lower levels of glycated hemoglobin — bloodsugar, essentially — compared to the processed one. And zooming in a bit, the researchers found that losses in fat mass, body fat percentage, visceral fat rating and total body water mass were "significantly lower" on the whole food diet, but not the ultraprocessed one. Participants on the processed diet also reported a greater number of "adverse events," like constipation, acid reflux, fatigue, and infections. Volunteers surveyed said that both diets were equally satisfying, likely because both tended toward "healthier"-seeming foods — both diets followed UK government dietary guidelines — though they reported having an easier time controlling their cravings on the non-processed diet, as noted by Gizmodo. Comparing the two menus is fascinating on its own. Participants on the non-processed diet enjoyed meals like breakfast cinnamon and apple overnight oats, sticky BBQ ribs with smoky rice and fruit, and salmon with herbed new potatoes and mixed vegetables. Their snacks were stuff like like blueberry, oat, nut and seed muffins, and pasta salad. Alarmingly, the ultraprocessed group were treated to foods that, on a surface level, looked comparable: commercial breakfast oat and fruit bars, premade ribs and rice with fruit snacks, and boxed salmon with potatoes and corn, and snacks made up of meal-replacement drinks, plant-based yogurt, and packaged wrapped oat bars. Though definitions vary, ultraprocessed food is typically identified as containing foodstuff rarely, if ever used in kitchens — like high-fructose corn syrup and hydrogenated oil — or which have been pumped full of synthetic additives like flavoring, dyes, sweeteners, or emulsifiers. Studies have shown that ultraprocessed food makes up over half of the dietary energy consumed in high-income countries like the US and the UK, the latter of which is where the clinical trial was run. Meanwhile, the ultraprocessed food market is projected to grow by an astonishing $856 billion over the next four years. It's not just about individual choices at the grocery store or restaurant, either: the class differences in diets are stark, with poor people forced to consume far more cheap ultraprocessed junk than their more affluent peers, meaning that to meaningfully improve health outcomes, the food supply needs to be restructured at a basic level. Joseph Lovett, TV producer who shed an early light on AIDS, dies at 80

In May 1983, when Mr. Lovett produced the first long-form news segment on AIDS for the ABC News program '20/20,' the disease had received slim coverage since being identified by the Centers for Disease Control and Prevention in 1981. Brandt Jr., assistant secretary of the Department of Health and Human Services; Dr. Anthony Fauci, who at the time was an immunology researcher at the National Institutes of Health; and playwright and AIDS activist Larry Kramer. After it aired, letters of support poured in from AIDS patients, health officials, and fellow journalists across the country. Mr. Lovett's reports were often crowdsourced from a vibrant network of activists and journalists he had cultivated in Lower Manhattan, where he lived. With his partner, Jim Cottrell, an anesthesiologist at SUNY Downstate Medical Center, he would host activists, journalists, and intellectuals at their home, on Fifth Avenue in Greenwich Village. After 10 years at the network, Mr. Lovett left ABC in 1989 and started his own independent production company. In the early 1990s, he created 'In a New Light,' a yearly series of star-studded specials aimed at demystifying the disease and raising prevention awareness. Airing from 1992 to 1996, it was hosted by celebrities including Elizabeth Taylor, Arsenio Hall, Paula Abdul, and Walters. Mr. Lovett drew heavily on personal tragedies in later films and documentaries. He staged a reenactment of his mother being crushed by a runaway car, an incident he witnessed as a 13-year-old, in the film 'The Accident' (1999). The death of his father and three siblings to cancer prompted him to produce and direct 'Cancer: Evolution to Revolution' (2000), a film about coping, treating, and learning to live with the disease; in one scene, he had a colonoscopy on camera. The documentary won a 2001 Peabody Award and was nominated for a Primetime Emmy in 2000. Advertisement Throughout his career, Mr. Lovett developed a frank approach to his productions. 'We don't talk about bodily fluids, we talk about sexual fluids,' he told Etcetera magazine, an LGBTQ+ digest based in Atlanta, before a new installment of 'In a New Light' in 1993. The youngest of five siblings, he moved in with his older sister, Trisha, after the death of his mother, Jeanne (Millman) Lovett, in 1958. His father, Samuel Lovett, a builder who worked on Cape Cod, had died of colon cancer in 1954, when Joseph was a child. He earned a bachelor's degree in English at Columbia University in 1967. That year, he enrolled in the film MFA program at the university, but, swept up by student protests, he dropped out in 1968. His first job after film school was casting actors for Amram Nowak Associates, a production company. He was hired as an editor for CBS in 1975 and later promoted to producer. His first long-form segment, 'Parents of Gays,' was aired in 1977 on 'Magazine,' a daytime program. Mr. Lovett met Cottrell in 1976. They married in Barnstable in 2004, on the first day that same-sex marriage was legal in Massachusetts. Mr. Lovett joined ABC as a producer in 1979. His '20/20' segments covering the AIDS crisis earned him a reputation as a crusading journalist, and he also became known as a gregarious mentor to young reporters. Among the projects he worked on was a one-hour ABC special on adoption, 'Born in My Heart' (2001), which he produced with Walters. He also produced and directed the documentaries 'Gay Sex in the 70s' (2005); 'Going Blind' (2010), which was prompted by his own glaucoma diagnosis; and 'Children of the Inquisition' (2019), which followed descendants of families who had fled the Spanish and Portuguese Inquisitions as they traced their roots. Cottrell is Mr. Lovett's only immediate survivor. One of his final projects, 'Something Terrible Happened to Joey' (2022), a short animated film about confronting childhood trauma narrated by Rosie Perez, follows a boy who experiences an unspecified 'terrible' thing and, as a result, has to navigate the world carrying a giant sack of coal. The boy lugs the coal around for years, but it begins to shrink only when he opens up to his friends about the terrible thing. NOVEL IMMUNOLOGIC SURVEILLANCE STUDY PROVIDES NEW INSIGHTS INTO POST-PANDEMIC RETURN OF RESPIRATORY VIRUSES

Ongoing clinical research study in US children documents the rebound of endemic respiratory viruses, builds foundation for expediting future vaccines and treatments AURORA, Colo., Aug. 7, 2025 /PRNewswire/ -- The first paper from a multi-year clinical research study has been published in The Lancet Infectious Diseases: Dynamics of Endemic Virus Re-emergence in Children in the USA Following the COVID-19 Pandemic (2022-2023): A Longitudinal Immunoepidemiologic Surveillance Study and demonstrates how the approach can improve modeling to better predict future outbreaks. The paper shares findings from a multicenter clinical research study, one of many studies that are part of the recently launched PREMISE (Pandemic Response Repository through Microbial and Immune Surveillance and Epidemiology) program, led by Dr. Daniel Douek at the National Institutes of Health's (NIH) Vaccine Research Center (VRC). Data collected during the first year of the PREMISE study, 2022-2023, shows for the first time how non-pharmaceutical interventions such as masking and distancing targeted towards SARS-CoV-2 during the pandemic also decreased circulation rates of and population immunity to common respiratory pathogens in children. The study provides new evidence-based insight into what was driving the large post-pandemic rebound in these diseases and enables more accurate predictions for the future. This study is a partnership between PREMISE and clinical research sites at academic institutions led by principal investigator Kevin Messacar, MD, PhD, infectious disease specialist at Children's Hospital Colorado, at the central site at University of Colorado Anschutz Medical Campus and Children's Hospital Colorado with additional study sites at University of North Carolina, Weill Cornell Medicine and the University of Alabama at Birmingham. The PREMISE study conducted immunologic surveillance on children younger than 10 years old, by enrolling them and following them for over a year. Through repeat blood sampling, the team could determine what children at varying young ages had immunity to and what they were susceptible to. Through respiratory sampling during illness, researchers were able to determine what infections they experienced. The data showed that most younger children lacked immunity to many normal respiratory viruses during the pandemic, suggesting they had not been exposed, as they typically would have, due to prevention measures in place. Following the lifting of pandemic measures, the level of immunity rose across all pathogens studied, reflective of the unprecedented widespread resurgence of these viruses in children after the end of the pandemic. While most research studies target a specific disease, samples from PREMISE were tested for many common and emerging respiratory viruses, including RSV, influenza and enterovirus D68 (EV-D68), which can cause the polio-like illness, acute flaccid myelitis. The data allowed experts to recreate past circulation patterns and model predictions for future outbreaks with greater accuracy and precision. They showed that PREMISE data from 2022-23 could be used to accurately predict the subsequent wave of disease of the emerging pathogen EV-D68 that occurred in 2024. "Four cohorts of almost 1,000 children have provided an invaluable bank of samples and data," said lead author Hai Nguyen-Tran, MD, infectious disease specialist at Children's Hospital Colorado and assistant professor at University of Colorado School of Medicine. "These are being used to develop 'on the shelf' medical countermeasures, such as antibody treatments and vaccines, for pathogens of interest. Instead of starting from scratch, this study gives us a head start to understand, predict and prepare for future pandemics." Samples and data from the PREMISE study will also be used to learn which parts of viruses the human immune system attacks to become immune, so teams can better design new antibody treatments and effective vaccines to mimic this response. "In the future, this type of immune surveillance can be used to better understand the impact of public interventions on population immunity and future waves of disease," said Dr. Messacar, who is also a professor at University of Colorado School of Medicine. "PREMISE is a great example of a successful research partnership between NIH scientists and clinical researchers in academia, leading to concrete deliverables such as vaccine candidates and monoclonal antibodies that can directly impact public health." This study is fully funded by a subcontract with Frederick National Laboratory for Cancer Research (FNLCR), currently operated by Leidos Biomedical Research, Inc. through Agreement 21X192QT1.
