New high-resolution structures of measles virus enzyme could lead to protective measures

May 30—Using high-resolution cryo-electron microscopy (cryoEM), researchers at The Hormel Institute, University of Minnesota, have revealed the first high-resolution renderings of the measles virus's (MeV) polymerase. This enzyme is crucial for the virus's ability to hijack cells and make copies of itself, which is one aspect that makes the virus so effective at infecting people and spreading throughout the body.
For a virus that's been documented since at least the ninth century, there is still plenty we have to learn about the measles virus and how it operates, Associate Professor Bin Liu, PhD, explained as he discussed his new study published in Nature Communications.
"Even well-known viruses like measles still have uncharted molecular terrain, and illuminating its structure provides valuable insights for therapeutic development," Liu said.
By revealing measles' structure, Liu, along with Postdoctoral Researchers Dong Wang, PhD, and Ge Yang, PhD, have unlocked valuable insights that could help other researchers develop preventative and therapeutic measures to combat this deadly virus that can cause complications ranging from pneumonia to ear infections to encephalitis (inflammation of the brain).
"Although an effective vaccine is available, recent measles outbreaks highlight the urgent need for alternative antiviral treatments," Liu said. "Because the polymerase is essential for viral genome replication, it represents a critical target for antiviral intervention."
The study presents two new, distinct renderings of MeV polymerase complexes known as Lcore-P and Lfull-P-C.
According to Liu, one of the most intriguing findings is the structural role of the measles virus C protein in forming the Lfull-P-C complex with two other proteins, L and P. This is surprising because the C protein was traditionally seen as a regulatory protein, not part of its core replication machinery. Now, it's shown to physically bridge and modulate the L protein's activity, potentially influencing how efficiently the virus replicates.
Additionally, the study shows that the C protein widens the polymerase's RNA channel in the polymerase, possibly enhancing the processivity of RNA synthesis. That kind of physical alteration, revealed via cryoEM at near-atomic resolution, is a remarkable mechanistic insight. It suggests that the measles virus has evolved an elegant, multi-protein solution for efficient replication inside host cells. This kind of structural adaptation is a biological engineering marvel, and it highlights how even simple viruses can have complex, dynamic protein machinery.
By revealing detailed interactions within the Lfull-P-C complex, the paper opens doors for next-generation antiviral drug designs that halt viral replication.
"This shifts the measles conversation from 'solved by vaccines' to 'still relevant for therapeutic innovation,'" Liu concluded.

By Bhanvi Satija (Reuters) -More than one in four U.S. adults with diabetes used one of the injectable medicines that target the GLP-1 protein last year, the U.S. Centers for Disease Control and Prevention said on Thursday. The wildly popular and effective GLP-1 drugs Mounjaro from Eli Lilly and Ozempic from Novo Nordisk are approved in the United States for treating type 2 diabetes. They are sold as Zepbound and Wegovy, respectively, for weight loss and are being tested for other conditions. Use of the injectable drugs was highest among adults with diabetes aged 50–64 at 33.3%, reflecting the more significant disease burden in this group, the report said. A quarter or 25.3% of adults with diabetes aged 18 to 34 reported using a GLP-1 drug, while the adoption rate was 20.8% among those 65 and older. The data are from a nationally representative annual survey of U.S. adults aged 18 and older that was conducted in person and with follow-up by phone in 2024. In 2024, for the first time, participants in the annual survey who had diabetes were asked if they were using the Lilly or Novo blockbusters or other GLP-1 drugs to lower blood sugar or lose weight. The drugs mimic the activity of a hormone that regulates blood sugar levels, slows digestion and helps people feel full for longer. Drugmakers Lilly and Novo have faced criticism about the cost of the treatments, which carry a list price of about $1,000 for a month's supply. Roughly 31% of survey participants who reported using insulin also reported using GLP-1 drugs, as did about 28% of patients who were using oral drugs to control their blood sugar, according to the report, indicating that these treatments are being integrated into combination regimens. Hispanic adults with diabetes had the highest rate of GLP-1 use, at 31.3%, followed by Black non-Hispanic and White non-Hispanic adults, at 26.5% and 26.2%, respectively, the survey found. Historic Marin City neighborhood will be getting an $85M renovation

The shortage of affordable housing is a familiar story in the Bay Area, especially in pricey locations like Marin County. But one neighborhood, the county's only true African American enclave, is about to get an upgrade. And on Wednesday, Marin City unveiled a tribute to how it all got started 83 years ago. Today's housing shortage is nothing compared to Marin during WWII. America was suddenly thrust into battle, and a huge shipbuilding operation called Marinship went up seemingly overnight. And on Aug. 13, 1942, the first Greyhound Buses began arriving with African American workers from the deep South. "This is my mother-in-law and my father-in-law, Willie and Ollie Hector," said Connie Hector, pointing to the couple's picture displayed high up on a light pole. Connie's in-laws worked at the shipyards and are among a group of residents memorialized on new banners honoring the legacy of the Black workers, many of whom were able to escape, for the first time, the oppression of the Jim Crow South. "And so, coming to California enabled them to feel like there was something better," said Connie. The housing that was built in Marin City in 1942 was considered a model for the rest of the country. The current two-story townhouses and five-story towers became home to a new generation of Black Marin residents. Connie's husband, David Hector, was born in the development known as Golden Gate Village. It was built in about 1960 to replace the wartime worker housing for the shipyard. Many people moved away after the war, but the African American workers stayed, and at one time they comprised more than 90 percent of the Marin City population. "People would like to be able to stay here," he said. "Most of the people are second and third generations that live in Marin City. And I think it would be important to be able to maintain that history." The problem is that many of the apartments in Golden Gate Village have deteriorated over time. Promises were made for years without delivery. But now, there is hope. Funding has been announced to completely renovate every housing unit in the development. That would mean a lot for Angela Pate, who has a lot of memories in her apartment, along with the urn holding her father's ashes. "Christmas Eve, he left us. But I've got him right here to keep us safe," she said. Once work begins, groups of residents, including Angela, will be moved to temporary housing while the units are gutted and rebuilt. "It's kind of something that I'm looking forward to doing," she said. "But at the same time, I'm having to move 17 years' worth of stuff out for three months and moving it back in! But when I move back in it's going to be something that no one's ever lived in. And that's the greatest feeling of all. Look, I'm 60-years-old and I'm going to do cartwheels! It's going to be very, it's going to be awesome." The renovation won't be cheap, $85 million to renovate 88 existing units. It would have been cheaper to demolish everything and start fresh. But while some may question spending nearly a million dollars each on affordable housing units, those who see it as more than just an apartment complex would disagree. "Because it's history," said Connie. "I mean, a lot of our monuments throughout the nation, you don't just knock 'em down or you shouldn't. Just knock 'em down. They stand for a purpose! And I'm not even from here! But I fell in love with the history of Marin City." The Golden Gate Village buildings in Marin City now stand for a higher purpose. They're now a monument to people seeking a better life. And it's hard to put a price tag on that.
