Science seek to tap amazing healing powers of the mouth's interior

ST. PAUL, Minn., July 2 (UPI) -- The cellular action that enables the mouth's interior lining to heal quicky and without scarring has always been a mystery but a new study suggests it may be linked to a specific "signaling pathway."
That new knowledge is raising hopes the mouth lining's healing powers could be applied to other skin wounds, which take far longer to heal and often leave permanent, unsightly scars, according to a preclinical study published Wednesday by Cedars-Sinai Medical Center and University of California-San Francisco researchers.
The study on mice provides evidence that the "Gas6/AXL" protein/enzyme signaling pathway is likely responsible for the mouth's amazing ability to heal itself completely from even deep bite wounds within a matter of days with no leftover scarring, the authors wrote in the journal Science Translational Medicine.
Known as the oral mucosa, the mouth's interior lining heals fast and cleanly, even though it is continuously exposed to different types of microbes, abrasion from foods, motion and tension. This unique regenerative capacity is made possible by specialized gene programs activated during injury, but just how those mechanisms work has been unknown.
In the study reported Wednesday, a research team led by Dr. Ophir Klein, executive vice dean of Children's Health at Cedars-Sinai and executive director of Cedars-Sinai Guerin Children's in Los Angeles, found that by manipulating levels of the AXL enzyme they could affect how wounds healed.
For instance, if they inhibited AXL in mice, the healing of oral mucosa wounds worsened, making them more like skin wounds. Conversely, when they stimulated AXL in facial skin wounds, they healed more efficiently and with reduced scarring, much like the oral mucosa.
Boosting the levels of AXL suppresses the expression of another protein called Focal Adhesion Kinase, or FAK, which plays a significant role in scar formation during wound healing.
Should future research validate the benefits for humans, the new knowledge could aid in developing new therapies to heal skin wounds with reduced scarring, Klein said.
"We are actively thinking about harnessing AXL-driven repair in both the mouth and the skin, and perhaps in other tissues as well," he told UPI in emailed comments. "One application that comes to mind is to improve healing and reduce scarring in the skin for burn victims or those with other cutaneous disorders.
"Another is that there are a number of diseases of the mouth, many of them autoimmune, in which severe oral ulcers form and heal poorly or not at all. These are different from canker sores, also known as aphthous ulcers, which many people get and which typically heal on their own; the more severe ulcers are difficult to treat and cause very significant illnesses, and we currently do not have good therapies for them."
A related direction for research could focus on "obtaining a deeper understanding of the effects of AXL signaling on scarring and regeneration at a molecular level, in terms of other components of the signaling pathways," Klein added.
"It will also be interesting to explore the role of this pathway in other tissues and organs, and in the context of distinct injuries and perturbations --for example, physical cuts vs heat burns vs irradiation versus chemical insults."
Meanwhile, a pair of recent animal studies led by the University of Arizona in Tucson similarly have shown that the use of FAK inhibitors can make skin grafts look more like healthy skin and produce less scarring, raising hopes for a potential new treatment for survivors of blast wounds, burns and other major injuries.
Kellen Chen, an assistant research professor in the university's Department of Surgery and lead author of those papers, said the current Cedars-Sinai study appears to add to a mounting list of evidence that "FAK acts as a central mediator of scarring in the oral mucosa gives additional credence to the importance of this factor." Chen was not involved with the new study.
"A better understanding of how the oral mucosa heals without scarring through the FAK pathway will allow us to better develop therapies to target FAK for healing in the future," he told UPI.
Chen said his team has the funding to pursue Food and Drug Administration approval for a FAK inhibitor, or FAKI, hydrogel therapy and expects to conduct human trials soon.
The growing evidence that wound healing can be accelerated at the cellular level "could have strong clinical implications for the treatment of severe injuries for humans and animals, especially because there are zero FDA-approved treatments that reduce scar formation," he added.
"Patients with severe injuries or burns can suffer a lifetime of pain and disfigurement from scarring. The potential to heal burn injuries with regeneration, rather than scarring, would dramatically change the lives of these patients."

File Photo by Qilai Shen/EPA Aug. 1 (UPI) -- U.S. Department of Transportation Secretary Sean Duffy announced Friday that the Federal Railroad Administration will cancel two grants totaling more than $26 million for the Baltimore-Washington maglev project. The department's press release about the Superconducting Magnetic Levitation Project said it has seen "nearly a decade of poor planning, significant community opposition, tremendous cost overruns, and nothing to show for it." The release called the project a "boondoggle." As part of its analysis, the FRA also determined the project would result in "significant, unresolvable impacts to federal agencies and federal property, including national security agencies," the release said. "We want big, beautiful projects worthy of taxpayer dollars -- including high-speed rail. This project lacked everything needed to be a success from planning to execution. This project did not have the means to go the distance, and I can't in good conscience keep taxpayers on the hook for it," Duffy said in a statement. "We'll continue to look for exciting opportunities to fund the future of transportation and encourage innovation." The Northeast Maglev would eventually connect Washington, D.C., and New York City. The train would be able travel at speeds of more than 300 mph to make the trip one hour long. Maglev is a system of rail transport whose rolling stock is levitated by electromagnets rather than rolled on wheels, eliminating rolling resistance. Compared with conventional railways, maglev trains have higher top speeds, superior acceleration and deceleration, lower maintenance costs, improved gradient handling, and lower noise. But they are more expensive to build, cannot use existing infrastructure, and use more energy at high speeds. Indirect effects of this project also would impair critical infrastructure and ongoing agency missions, the release said. Government agencies harmed by this project would have included: the National Security Agency, U.S. Department of Defense and Fort George G. Meade, National Aeronautics and Space Administration, U.S Department of Agriculture, U.S. Secret Service, U.S. Department of Interior -- Fish and Wildlife Service and National Park Service, and the U.S. Department of Labor. In 2015, the federal government gave Maryland a grant of $27.8 million to study a high-speed maglev train line that could connect Baltimore and Washington, D.C., in 15 minutes. Duffy is now canceling that grant. The funding for such a grant was authorized in 2005, when Congress set aside $90 million for maglev projects. In 2021, China unveiled a maglev train that it said can travel 373 mph. In July 2020, the government said it planned to build a network with as many as nine maglev lines that include 620 miles of track.

NASA astronauts Zena Cardman and Mike Fincke, Japan Aerospace Exploration Agency astronaut Kimiya Yui, and Roscosmos cosmonaut Oleg Platonov, arrived at the launch complex, where SpaceX's Dragon spacecraft is ready for liftoff at 11:43 a.m. EDT Friday. Cardman is the commander, Fincke is the pilot, and Yui and Platonov are mission specialists. The launch was scrubbed on Thursday just before liftoff due to inclement weather at Kennedy Space Center in Florida. The mission was scheduled for 12:09 p.m. EDT from Launch Complex 39A at Kennedy Space Center aboard a Falcon 9 Rocket. Possible goals of these studies include ways to treat or prevent a group of eye and brain changes that can happen during long-duration space travel. Another experiment will measure how multiple systems within the human body change in space, using vision assessments, MRI scans, and other medical exams to give a complete overview of the body's response to long-duration spaceflight.

JTWNI, GARD join EarthJustice, other groups in legal action against EPA

Two Northwest Indiana community groups have joined EarthJustice to challenge the Environmental Protection Agency's actions to delay air pollution compliance in steel mills. 'The 2024 air standards — hard-won safeguards for fenceline communities — are being stripped away, as the agency grants emissions exemptions to steel mills and eliminates public input,' Lisa Vallee, organizing director of Just Transition Northwest Indiana, said in a Wednesday news release. 'This is environmental injustice: they are sacrificing our communities, gutting our social safety nets, and taking away our future. We will not stay silent.' Earlier this month, the EPA issued a final rule delaying air pollution protections for communities near steel mills, according to the Federal Register. Current rules have required steel mills to reduce emissions since last April, but the agency's new action delays compliance until April 2027, allowing pollution to continue at current levels. According to EarthJustice, the EPA published the final rule without notice, which did not allow the public to object to the extension before it went into effect. EarthJustice is a San Francisco-based nonprofit public interest environmental law organization that works with community groups nationwide. EarthJustice Senior Attorney Adrienne Lee said in a Wednesday news release that it's alarming for the EPA to delay protections without first accepting public input. The action is also inconsistent with the agency's responsibility to human health and the environment, Lee said. 'Communities living near steel mills in states such as Indiana and Pennsylvania deserve better,' she said. 'Each year, steel mills expose these communities to hundreds of tons of toxic air pollution which places them at increased risk of developing cancer and a variety of chronic health conditions.' An October report from Industrious Labs found that most residents in Gary are in the top 10% nationally for being most at-risk for developing asthma and at-risk of low life expectancy. In 2020, Indiana had a lung cancer rate of 72.5 per 100,000 people, with Lake County as one of the state's counties with the highest cancer mortality rates, according to the American Lung Association. A 2016 JAMA Network report also found Gary as one of the top five U.S. cities with the lowest life expectancy at one point. The EPA declined to comment on the legal action, saying in an email that it's longstanding practice not to comment on current or pending litigation. Dorreen Carey, president of Gary Advocates for Responsible Development, said in the news release that EPA's actions fail to protect Gary and surrounding Northwest Indiana communities' residents. Carey called out U.S. Steel's Gary Works facility specifically, saying pollutants have led to increased health issues in the city. 'The current outmoded and insufficient methods of hazardous air emission monitoring and control at Gary Works are not protective of human health and the environment,' Carey said. 'Residents must have a voice in EPA decisions and rulemaking and EPA must do its job to protect the residents and the natural resources of Gary…' A U.S. Steel spokesperson responded to the action in a statement Thursday. 'Environmental stewardship is a core value at U.S. Steel, and we remain committed to the safety of our communities as do our more than 3,400 Gary Works employees,' the statement said. JTNWI and GARD have previously expressed concerns with U.S. Steel's application for two-year exemptions from hazardous air pollutant rules for integrated iron and steel, coke and taconite iron ore process, according to Post-Tribune archives. U.S. Steel previously told the Post-Tribune in a statement that it challenged all three rules because they were not supported by science or law and would impose significant costs while setting technically unachievable standards. Seeking these exemptions doesn't mean U.S. Steel isn't 'supportive of revisions to regulations that are within (the EPA's) statutory authority, based on sound science and are technically feasible,' according to Post-Tribune archives. In March, the EPA announced corporations could apply for presidential exemptions to sections of the Clean Air Act. Exemption applications were due March 31, and if approved, can be extended for up to two additional years. 'Northwest Indiana is already home to some of the worst air pollution in this country,' a previous JTNWI statement said. 'In this region, we suffer from a cumulative, generational impact of exposure to industrial toxins in communities like Lake County, Indiana. These exemptions are a free pass for these polluters at the continued cost of our health and safety. We're adamantly opposed to this astonishing reversal of environmental law in no uncertain terms. Our communities and region are not expendable sacrifice zones for this administration or corporations.'
