London – 28 April 2025 - Purespring Therapeutics, a pioneering company focused on transforming the treatment of kidney diseases, today announces that the European Medicines Agency (EMA) has granted orphan drug designation to PS-002, Purespring's lead programme, in IgA nephropathy (IgAN).
Orphan drug designation in the European Union (EU) is granted by the European Commission based on a positive opinion adopted by the EMA Committee for Orphan Medicinal Products. Orphan designation provides incentives to developers of medicines for limited patient populations, including market exclusivity, protocol assistance (guidance on study design and scientific evaluation) and regulatory fee reductions.
IgAN is a common, chronic autoimmune kidney disease that primarily affects young adults. In patients with the disease, a protein called immunoglobulin A (IgA) becomes trapped in the filters of the kidney, known as glomeruli, causing inflammation, damage and scarring to the whole kidney. There are currently no approved therapeutic options to stop disease progression, nor a cure, and about one third of high risk IgAN patients will go on to lose kidney function within five years and require a kidney transplant or dialysis.

Everest Medicines Presents Positive Results in Preliminary Analysis of Phase 1b/2a Clinical Trial of Novel BTK Inhibitor EVER001 at the 62nd Congress of the European Renal Association
EVER001 is a covalent reversible BTK inhibitor with potentially best-in-class characteristics for the treatment of primary membranous nephropathy (pMN) and other autoimmune renal diseases, including IgA nephropathy (IgAN), minimal change disease (MCD), focal segmental glomerulosclerosis (FSGS), and lupus nephritis (LN), offering treatment options for over 10 million patients worldwide. No drug has been approved globally for the treatment of pMN currently. There are approximately 2 million patients with pMN in China, and nearly 220,000 patients in the United States, Europe and Japan. As of December 17th, 2024, the ongoing Phase 1b/2a clinical trial of EVER001 includes longer-term data collected from some patients: 10 patients in the low-dose cohort completed 52 weeks of follow-up, and 10 patients in the high-dose cohort completed 24 weeks of treatment. Preliminary results showed that EVER001 was well-tolerated and effective in patients with pMN. These results support the potential of EVER001 as a treatment for proteinuric autoimmune glomerular diseases.- Compared to baseline, the least squares (LS) geometric mean levels of anti-PLA2R autoantibodies decreased by 62.1% in the low-dose cohort and 87.3% in the high-dose cohort at week 12. The reductions in both cohorts reached approximately 93% at week 24.- In the low-dose cohort, a 78.0% of reduction in proteinuria was observed by the end of 36 weeks of treatment. This reduction was sustained through week 52. In the high-dose cohort, a 70.1% of reduction in proteinuria at week 24 was shown.- EVER001 was generally safe and well tolerated. Preliminary results showed that EVER001 was well-tolerated and effective in patients with pMN. These results support the potential of EVER001 as a treatment for proteinuric autoimmune glomerular diseases. This initial unveiling of the preliminary data at an international congress focuses on pMN, which is the second most common cause of primary glomerulonephritis. EVER001 is a covalent reversible BTK inhibitor with potentially best-in-class characteristics for the treatment of autoimmune renal diseases. Compared to covalent irreversible BTK inhibitors, EVER001 offers improved selectivity while maintaining high potency, thereby potentially avoiding many of the side effects associated with earlier-generation BTK inhibitors. This oral presentation highlights an ongoing Phase 1b/2a clinical trial of EVER001 for the treatment of pMN, which is being conducted in China. The study is designed to evaluate the safety, efficacy, pharmacokinetics, and pharmacodynamics of EVER001 in Chinese patients with glomerular diseases characterized by proteinuria. A total of 31 patients with biopsy-proven pMN who tested positive for anti-PLA2R autoantibodies were enrolled into two cohorts. The total treatment duration was 36 weeks. The presentation includes longer-term data collected from some patients (10 patients in the low-dose cohort completed 52 weeks of follow-up, and 10 patients in the high-dose cohort completed 24 weeks of treatment). Preliminary results showed that EVER001 was well-tolerated and effective in patients with pMN. These results support the potential of EVER001 as a treatment for proteinuric autoimmune glomerular diseases. The trial results, based on data analysis as of December 17th, 2024, demonstrated that EVER001 was generally safe and well tolerated, with the most common TRAEs being Grade 1-2. No clinically significant adverse events commonly associated with covalent irreversible BTK inhibitors, such as bleeding, arrhythmia, severe infections, or severe liver function impairment were observed. Compared to baseline, the least squares (LS) geometric mean levels of anti-PLA2R autoantibodies decreased by 62.1% in the low-dose cohort and 87.3% in the high-dose cohort at week 12. The reductions in both cohorts reached approximately 93% at week 24. 76.9% of patients in the low-dose cohort and 81.8% in the high-dose cohort achieved immunological complete remission at week 24. For 24-hour proteinuria, the LS geometric mean decreased by 78.0% from baseline in the low-dose cohort at week 36 and the reduction was maintained for 16 weeks after the end of treatment. At week 36, 69.2% of patients in the low-dose group achieved clinical remission. In the high-dose cohort, proteinuria had already decreased by 70.1% at week 24, with 80.0% of patients achieving clinical remission. Patients in both cohorts maintained stable renal function during the treatment period. Novo's Ozempic Linked to Rare Cases Of Serious Eye Disorder, EU Regulator Says
(Reuters) -Novo Nordisk's popular weight-loss and diabetes drugs Wegovy and Ozempic may in very rare cases cause a serious eye condition that can lead to vision loss, the European Medicines Agency's safety committee said on Friday. In the past, studies in type-2 diabetes patients have linked Ozempic to the condition called non-arteritic anterior ischemic optic neuropathy (NAION). But this is the first time a regulator has confirmed the side effect. The condition may affect up to 1 in 10,000 people taking semaglutide, the active ingredient in Wegovy and Ozempic as well as in Novo's other diabetes drug Rybelsus, for at least one year, the regulator said. NAION is the second-most common cause of blindness due to optic nerve damage, after glaucoma. "This has been reported as a potential risk for some time, so I think the clinical community is relatively aware of it. I don't see this as making any major difference to prescribing patterns," said Barclays analyst Emily Field. U.S.-listed shares of the Danish drugmaker were up nearly 2.5% in early trading. The EMA, which started its review in December, has asked Novo to add NAION as a side effect of very rare frequency in the product information accompanying drugs that contain semaglutide. Novo said it would work with the EMA to update the labels, adding clinical trials and after-market studies did not suggest a reasonable possibility that the drugs caused the condition. The "benefit-risk profile of semaglutide remains favorable," the company said in a statement. Novo has recently faced investor concerns that it is losing its first-mover advantage in the highly competitive obesity treatment market, leading the company to oust CEO Lars Fruergaard Jorgensen in May. Wegovy and Eli Lilly's Zepbound currently dominate the weight-loss drug market, potentially worth about $150 billion by the next decade. The EMA said several large studies in type-2 diabetes patients have suggested that use of Novo's drugs could raise the risk of developing NAION by twofold. A study of nearly 350,000 diabetes patients published in March showed that the risk of developing NAION more than doubled after two years of treatment with Ozempic, compared to patients taking medicines from other classes. The U.S. Department of Health and Human Services did not immediately respond to a Reuters request for comment on whether the Food and Drug Administration was conducting a probe into the side effect. Nefecon Shows IgAN Benefits Regardless of Baseline eGFR
Nefecon, an oral targeted-release formulation of budesonide that targets drug release into the distal ileum, provided significant improvements in the loss of kidney function in patients with immunoglobulin A nephropathy (IgAN) at 9 months, regardless of baseline estimated glomerular filtration rate (eGFR) status, according to a new subanalysis of the phase 3 NeflgArd study. METHODOLOGY: Patients in the NeflgArd study with primary IgAN were randomly assigned to one of two groups of 182 patients each to receive either treatment with 16 mg Nefecon or placebo daily, along with supportive care with renin-angiotensin system inhibition. This was followed by a 15-month off-drug observational period allowing for continued supportive care. For the subanalysis, the patients were further stratified on the basis of eGFR deciles in relation to the overall study population, with levels above and below 38, 43, 47, 51, 55, 60, 66, 72, and 82 mL/min/1.73 m 2 . . The median eGFR overall was 55.49 mL/min/1.73 m2 and was balanced across the Nefecon and placebo groups. TAKEAWAY: The relative benefit of Nefecon in terms of eGFR was seen across most deciles and timepoints and showed a generally greater benefit in higher baseline eGFR groups (above 72 mL/min/1.73 m 2 ). ). Furthermore, the relative urine protein-creatinine ratio benefit was observed from month 9 with Nefecon vs placebo regardless of baseline eGFR, with a continued reduction to month 12, followed by a maintained treatment benefit to month 24. The eGFR benefits with Nefecon over placebo were observed among patients within the upper and lower deciles over 2 years. eGFR showed a pronounced on-treatment benefit and a delay in decline during the observation period. IN PRACTICE: 'This NefIgArd subanalysis demonstrated that the efficacy of a 9-month Nefecon treatment course in reduction of proteinuria and preservation of kidney function was independent of baseline eGFR,' said first author Jonathan Barratt, MD, of the College of Life Sciences, University of Leicester, UK. 'So, we should be confident about treating people with Nefecon across the full range of eGFR in terms of the eGFR saving we're going to see, but also the proteinuria reduction,' he added.
