Poxel Announces Positive Preclinical Data for PXL065 in Hypertrophic Cardiomyopathy To Be Presented at the ESC Congress 2025

PXL065 demonstrated significant benefits in a HCM mouse model preventing pathological myocardial remodeling, including hypertrophy and fibrosis in the heart Presentation of detailed results from preclinical study on Sept 1st, 2025, 11:15 am CEST, at the European Society of Cardiology 2025 Findings support the clinical development of PXL065 as a potential disease- modifying treatment for symptomatic and asymptomatic HCM patients LYON, France, May 26, 2025--(BUSINESS WIRE)--Regulatory News: POXEL SA (Euronext : POXEL - FR0012432516), a clinical stage biopharmaceutical company developing innovative treatments for chronic serious diseases with metabolic pathophysiology, including metabolic dysfunction-associated steatohepatitis (MASH) and rare metabolic disorders, today announces that the abstract featuring previously announced preclinical data demonstrating positive results for PXL065, the deuterium-stabilized R-enantiomer of pioglitazone, in hypertrophic cardiomyopathy1, has been accepted for presentation at the 2025 edition of the European Society of Cardiology (ESC) Congress (link), to be held jointly with World Congress of Cardiology, on September 1st, 2025 at 11:15 am CEST, in Madrid, Spain. Prof. Dr. Cordula Wolf, Director of the Center for Rare Congenital Heart Diseases at the TUM University Hospital German Heart Center, stated: "The compelling results obtained in this study illustrate the potential of PXL065 in HCM, the most common genetic cardiac disorder. Current treatments have important limitations in efficacy, safety, or patient applicability. There is a clear unmet need for safe and effective disease-modifying therapies." Thomas Kuhn, CEO of Poxel, added: "We are pleased to have the data with PXL065 in HCM be presented at one of the world's leading forums for cardiovascular science and medicine, which underscores both the quality and relevance of these findings. We look forward to further supporting PXL065 development for the treatment of HCM based on these promising results." Hypertrophic Cardiomyopathy (HCM) is a genetic disorder marked by myocardial hypertrophy, cardiac fibrosis, ventricular dysfunction, arrhythmias, and an increased risk of sudden cardiac death. It is caused by mutations in sarcomere protein genes, leading to altered cell metabolism, including oxidative stress and mitochondrial dysfunction. The estimated prevalence of HCM is 0.2%, or 1/500 adults, and its incidence is around 5 per 100,000 person-years. In connection with the mechanism of action of PXL065 on the inhibition of the mitochondrial pyruvate carrier (MPC) and on the inhibition of Acyl CoA Synthetase 4 (ACSL4) thus acting on oxidative stress, inflammation and fibrosis, PXL065 was successfully assessed in an established mouse model of hypertrophic cardiomyopathy. This preclinical study was funded by the German Center for Cardiovascular Research (DZHK) and conducted at the TUM University Hospital German Heart Center by leading HCM expert Prof. Dr. Cordula Wolf. Poxel and the TUM University Hospital German Heart Center collaborated on the pre-clinical study based on Poxel's existing data and patent portfolio on PXL065 and prior research conducted by Prof. Dr. Cordula Wolf and her group on the disease mechanisms and therapeutic use of TZD's in HCM. About Poxel SA Poxel is a clinical stage biopharmaceutical company developing innovative treatments for chronic serious diseases with metabolic pathophysiology, including metabolic dysfunction-associated steatohepatitis (MASH) and rare disorders. For the treatment of MASH, PXL065 (deuterium-stabilized R-pioglitazone) met its primary endpoint in a streamlined Phase 2 trial (DESTINY-1). In rare diseases, development of PXL770, a first-in-class direct adenosine monophosphate-activated protein kinase (AMPK) activator, is focused on the treatment of adrenoleukodystrophy (ALD) and autosomal dominant polycystic kidney disease (ADPKD). TWYMEEG® (Imeglimin), Poxel's first-in-class product that targets mitochondrial dysfunction, is now marketed for the treatment of type 2 diabetes in Japan by Sumitomo Pharma and Poxel expects to receive royalties and sales-based payments. Poxel has a strategic partnership with Sumitomo Pharma for Imeglimin in Japan. Listed on Euronext Paris, Poxel is headquartered in Lyon, France, and has subsidiaries in Boston, MA, and Tokyo, Japan. In pictures: Austria welcomes JJ back home after Eurovision win

As JJ, whose full name is Johannes Pietsch, walked through the gate, hundreds of fans on Sunday cheered. Some played his song, others hugged him. The 24-year-old countertenor, whose winning song combines operatic, multi-octave vocals with a techno twist, and who also sings at the Vienna State Opera, held up his trophy in one hand and a big bouquet of roses in the other. He smiled, wiped away tears and told the crowd 'that victory is for you.' JJ is Austria's third Eurovision winner, after Conchita Wurst in 2014 and Udo Jürgens in 1966. Austria's president, Alexander van der Bellen, celebrated JJ in a video posted on X. 'What a success! What a voice! What a show!' he exclaimed. 'All of Austria is happy.' Chancellor Christian Stoecker wrote on X: 'What a great success — my warmest congratulations on winning #ESC2025! JJ is writing Austrian music history today!' The Vienna State Opera also expressed joy over the win. 'From the Magic Flute to winning the Song Contest is somehow a story that can only take place in Austria,' opera director Bogdan Roscic told the Austrian press agency APA. Several Austrian cities were quick to show their interest in hosting next year's contest. Innsbruck Mayor Johannes Anzengruber told APA that 'not everything has to take place in Vienna... Austria is bigger than that,' and the towns of Oberwart in Burgenland and Wels in Upper Austria also threw their hats into the ring. JJ himself said he hoped that Vienna would get the next ESC which he would love to host together with his mentor, Conchita Wurst.

Austrian president issues nine-word statement after JJ's huge Eurovision win

JJ, whose full name is Johannes Pietsch, was Austria's third Eurovision winner, after bearded drag queen Conchita Wurst in 2014 and Udo Jürgens in 1966 Austria's JJ received a hero's welcome at Vienna airport this Sunday, after taking the title at the 69th Eurovision Song Contest with his chart-topping hit Wasted Love. The scene was one of celebration as hundreds of fans erupted into cheers as JJ made his grand entrance through the arrivals gate. Devotees of the 24-year-old countertenor, who is also a performer at the Vienna State Opera, were seen blasting his winning track, eager to embrace him and grab his autograph. Victorious and visibly moved, JJ, whose offstage name is Johannes Pietsch, beamed as he waved his trophy alongside a sizeable bunch of roses, declaring to all present that "that victory is for you." ‌ ‌ With three Eurovision wins under its belt, Austria celebrates JJ's triumph, joining ranks with past victors including the iconic Conchita Wurst and Udo Jürgens. Overwhelmed with emotion, JJ reflected on his incredible achievement post-victory in Basel, acknowledging the surreal nature of his success, stating, "This is beyond my wildest dreams. It's crazy,". Top officials didn't hesitate to share their pride; President Alexander van der Bellen heralded JJ's spectacular win online, proclaiming, "What a success! What a voice! What a show!" The entire nation basked in the glory, according to the president. Adding to the avalanche of praise, Chancellor Christian Stoecker took to social media, exclaiming, "What a great success - my warmest congratulations on winning #ESC2025! JJ is writing Austrian music history today!". The Vienna State Opera is over the moon with the recent win. Bogdan Roscic, the opera director, told APA: "From the Magic Flute to winning the Song Contest is somehow a story that can only take place in Austria,". ‌ Cities across Austria didn't waste any time showing their eagerness to host next year's contest. Innsbruck's Mayor Johannes Anzengruber suggested to APA that "not everything has to take place in Vienna. ... Austria is bigger than that," while Oberwart in Burgenland and Wels in Upper Austria also expressed their desire to host the event. JJ himself, fresh from his Saturday night victory, expressed his hope that Vienna would be chosen for the next ESC, an event he'd relish hosting alongside his mentor, Conchita Wurst. Controversial Israeli singer Yuval Raphael clinched second place, with the entrant's participation being hit with criticism over the country's bombardment of Palestine, which had resulted in over 52,000 deaths. In the glow of triumph at a press conference, JJ shared that the essence of his song about unrequited love is that "love is the strongest force on planet Earth, and love persevered. "Let's spread love, guys," declared JJ, expressing his pride at being the first Eurovision champion of Filipino heritage and a proudly queer winner.

BBC viewers slam 'cringe' Eurovision Song Contest 2025

Broadcasting live from St Jakobshalle in Basel, Switzerland, the show hit screens and airwaves via BBC iPlayer, BBC One, Radio 2, and BBC Sounds on Saturday (May 17). Presenters Hazel Brugger, Michelle Hunziker, and Sandra Studer were at the helm for the night's festivities as 26 acts battled it out for the coveted trophy. However, the trio's pre-recorded opening segment failed to impress, leaving viewers less than thrilled. It isn't the first time something appears to have gone wrong at Eurovision either. Fans quickly voiced their opinions on X, formerly known as Twitter, with one viewer commenting: "why are the presenters always cringe? #Eurovision.", reports Wales Online. Another chimed in: "Awful cringe hosting but loving to randomness of the hosts costumes #Eurovision." A third viewer remarked: "cringe opening #Eurovision." While a fourth questioned: "Why are the intros always so awkward? #Eurovision #ESC2025." It was a busy evening for Graham, who made a stunning cameo in Doctor Who as a hologram as part of BBC's Super Saturday. After informing Ncuti Gatwa's Doctor that the world is going to end next week in a stunning cliffhanger, Graham was on Eurovision commentary duties for the station. Talk show legend Graham has been commentating on the Eurovision for the BBC since 2009 when he took over the role from Terry Wogan.
