Understanding cell signals could change the way hair loss is treated, study finds

A study titled 'Pathophysiological Mechanisms of Hair Follicle Regeneration and Potential Therapeutic Strategies' published in Stem Cell Research & Therapy (2025) explains how hair loss, especially androgenetic alopecia (AGA), is caused by problems in the body's natural hair growth signals -- not just hormones or ageing. The review focuses on five important biological pathways -- Wnt/β-catenin, Sonic Hedgehog (Shh), BMP, Notch, and AKT/MAPK that help control when hair grows, falls and regrows.
Study highlights
The study found that in people living with AGA, the signals that help hair grow (like Wnt and Shh) are blocked, while signals that stop growth (like BMP and TGF-β) are more active. This leads to smaller, weaker hair follicles that stop producing hair over time. 'These signals work together like a team,' Debraj Shome, co-founder and director at The Esthetic Clinics and Depti Bellani, senior manager medical affairs and research at QR678 and lead authors of the study explain. 'To fix the problem, we need to focus on all of them -- not just one.'
Current treatments like minoxidil (topical solution that works by increasing blood flow to the scalp and extending the hair's growth phase) and finasteride (oral medication that reduces the production of dihydrotestosterone (DHT), a hormone that contributes to hair follicle miniaturisation and hair loss) can slow down hair loss or help with some regrowth, but they do not treat the real reason why follicles stop working.
The study highlights new types of treatments that are being tested, such as small molecules that can restart growth signals, stem cell therapy and gene editing. Formulations such as QR678, developed by the authors' research team, are also part of this new approach.
Shift in hair loss treatment
Experts say this could mark a big shift in how hair loss is treated. Instead of just covering up the symptoms, future treatments may be able to restart the hair follicle's natural growth process. However, the study also warns that activating these signals too much, especially Wnt or AKT -- can be risky and must be done carefully.
With over 50% of men and 25% of women affected by AGA by the age of 50, the authors say it's time to look at hair loss not just as a cosmetic problem, but as a health issue that can be treated by understanding how the body's regenerative systems work. 'This isn't just about growing hair,' they said. 'It's about waking up the body's natural repair systems and helping hair follicles work the way they're meant to.'
The authors also stress the importance of collaboration across medical specialties to move this research forward. Since hair loss is linked to hormonal changes, immune responses, and genetic factors, a combined approach involving dermatologists, endocrinologists, geneticists, and regenerative medicine experts is essential. 'Personalised treatment plans based on each person's biological profile will likely give better results,' the authors note. They add that wider clinical trials, safety studies and shared research efforts will be key to turning these scientific findings into real, effective therapies for patients.
Various forms of hair loss and regenerative medicine
The study also points out that other forms of hair loss, such as scarring alopecia, autoimmune types like alopecia areata and hair loss caused by chemotherapy are linked to similar problems in the body's regenerative signalling systems. This suggests that the same scientific approach could help treat a wider range of hair loss conditions. The authors say that the hair follicle is more than just a cosmetic structure, it functions like a mini-organ and offers valuable insight into how tissues heal, how the immune system protects certain cells, and how stem cells behave.
By studying hair follicles, researchers can also learn more about the body's broader ability to repair itself, making hair loss an important area of focus in regenerative medicine.

A drug designed to treat coeliac disease -- an autoimmune disorder primarily affecting the small intestine -- has shown promise for children affected with severe post-Covid syndrome, according to a small study. Covid infection, although rare in kids, leads to multisystem inflammatory syndrome in children (MIS-C) -- a serious condition that presents as high fevers, gastrointestinal symptoms, and life-threatening cardiac injury. The trial results, published in the journal Science Translational Medicine, showed that larazotide supported children in returning to normal activities more rapidly following Covid. "While our study is small, its results are powerful and have implications not only for MIS-C, but potentially for long Covid," said lead author Lael Yonker, co-director of the Cystic Fibrosis Center, at Mass General Brigham. "Our findings suggest that larazotide is safe and quickly resolves symptoms in children with MIS-C. We are now running a clinical trial to test whether larazotide may also be a useful therapy to treat patients with long Covid," Yonker added. Current MIS-C treatments are limited. Some patients receive general anti-inflammatory drugs, but many experience a rebound of symptoms after completing a course. Such drugs are not designed to target the sticky SARS-CoV-2 viral particles that may persist in the gut. On the other hand is the orally administered Larazotide, which targets the gut and strengthens intestinal barriers to limit the number of materials -- like SARS-CoV-2 viral particles -- that exit the intestines and enter circulation. To test larazotide's efficacy and safety as an MIS-C treatment, researchers conducted a double-blind clinical trial with 12 children experiencing early-stage MIS-C. Each patient randomly received either a placebo or larazotide four times daily for 21 days, then was tracked over six months of recovery. Children who received larazotide showed faster resolution of gastrointestinal symptoms, faster clearance of SARS-CoV-2 viral particles, and more rapid return to normal activities, the team said.

SB43154 drug used in cancer research shows promise against COVID-19 in preclinical studies: Scientists

After a long battle and research, Indian scientists have found out that a drug used in cancer studies has the potential to provide substantial protection against the coronavirus infection in preclinical studies. Remarkably, even after more than 50 consecutive generations exposed to the drug, the virus was unable to develop resistance. Scientists from the Indian Council of Medical Research (ICMR) and Indian Council of Agricultural Research (ICAR) have jointly found that the drug -- SB431542, an ALK5 inhibitor -- is highly effective against the coronavirus. Study findings This discovery was a collaborative effort between the National Centre for Veterinary Type Culture in Hisar and National Institute of Virology (NIV) under the ICMR in Pune, said NIV Director Dr Naveen Kumar. The study was first released as a preprint on bioRxiv and has since been accepted for publication in the Journal of Virology. If future human trials confirm its effectiveness, the drug could become a groundbreaking tool in the fight against COVID-19. Since December 2019, when the coronavirus emerged from China's Wuhan, it has spread to nearly every country in the world. Since 2020, more than 30 anti-Covid vaccines have received the World Health Organization's (WHO) approval for global use. "However, health experts believe vaccines alone are not enough. The virus continues to mutate, and many existing treatments like Remdesivir and monoclonal antibodies will quickly lose effectiveness," Kumar said. In this context, Indian scientists have been screening small molecule inhibitors that mainly target cellular kinases and phosphatases. One such compound, SB431542 -- originally developed to block TGF-β (Transforming Growth Factor-beta) signalling and commonly used in preclinical cancer, inflammation and fibrosis research -- has now been found by Indian researchers to also inhibit the coronavirus effectively. Kumar explained that SB431542 fights the virus in three fronts. It first blocks the virus from entering human cells by targeting the TGF-β/Smad pathway. Secondly, it disrupts the ability of the virus to assemble inside cells by interfering with ORF3a-related lysosomal dysfunction. Finally, it prevents the virus from killing the infected cell (a process called apoptosis), which helps stop the virus from escaping and spreading to other cells. This three-pronged approach has not been observed in any existing antiviral drug. Interestingly, the drug acted through both direct and indirect mechanisms -- it directly targets the viral ORF3a protein and indirectly hinders the virus by blocking the host cell's TGF-β signaling pathway. SB431542 was also tested against the chicken coronavirus (Infectious Bronchitis Virus or IBV) in specific pathogen-free chicken embryos. Embryos treated with the drug developed normally and showed no mortality, while those in the untreated control group failed to develop properly and did not survive. Kumar emphasised that even after exposing the virus to the drug over 50 successive generations (passage), it failed to develop resistance. This suggests that the chances of the virus evolving a drug-resistant variant against this compound are extremely low. This is a major advancement over existing antivirals like Remdesivir, against which the virus builds resistance quickly.

However, frequent and unmindful usage of lipsticks can lead to skin concerns, says a dermatologist on the occasion of World Lipstick Day. Prolonged use of low-quality lipsticks, particularly those containing synthetic dyes, preservatives, or fragrances, can contribute to issues like allergic contact cheilitis or chronic chapping, especially if the product isn't removed thoroughly at the end of the day,' dermatologist Dr Rinky Kapoor, Director and Co-Founder of The Esthetic Clinics, tells HT Lifestyle. This is why it is crucial to pick your lipstick wisely and refrain from any low-quality lipsticks. She suggests on choosing high quality lipsticks, preferably, apply with SPF infused lip balm to protect them from the UV rays. Are lipsticks safe to use daily? Most dermatologically-tested lipsticks are safe for daily wear. According to Dr Kapoor, these days, many modern formulas include hydrating agents like vitamin E, squalene, or natural oils that are safe to use. However, repeated application, especially without a hydrating base, can cause dryness or irritation in some users. She also claims that a few lipsticks may also have low levels of heavy metals like lead or cadmium in certain products, which is unsafe for your lips in the long run. 'That's why I advise using lipsticks from transparent, well-regulated brands and paying attention to how your lips respond," says Dr Kapoor. Are natural or organic lipsticks better for lips? 'Not always,' says the expert. 'The terms 'natural' and 'organic' are appealing, but they don't guarantee safety. While organic lipsticks may avoid synthetic dyes and preservatives, they are not immune to causing irritation. The real benefit lies in simpler ingredient lists and fewer additives, which can help those with sensitivities," she adds. In a nutshell, the safest lipstick, as per her is not the most botanical, it's the one that's well-tested, well-formulated, and suited to your individual skin type. How to maintain lip care while using lipstick regularly? Avoid over-applying drying, matte formulas, and always remove lipstick completely at the end of the day. 'Healthy lips require more than just choosing the right lipstick, it begins with a protective base, a hydrating, non-irritating balm, to reduce moisture loss and irritation,' she asserts. In addition, Dr Kapoor also recommends gentle exfoliation that helps prevent flaking and improves product application. She also suggests using an overnight lip mask or thick balm to support skin repair. 'Also consider SPF-infused lip products to prevent sun-induced pigmentation, which is often overlooked', says Dr. Rinky. What to look for in a dermatologically safe lipstick? 'Look for lipsticks labelled hypoallergenic, fragrance-free, and dermatologically tested. These markers indicate lower risk of irritation," explains Dr Kapoor. Also look for hydrating ingredients like shea butter, jojoba oil, ceramides, or vitamin E in your lipstick. 'Avoid vague terms like parfum, harsh preservatives, and overly bright synthetic dyes unless they are certified for cosmetic use." And when in doubt, patch test! Understanding ingredient labels: Spotting irritants and allergens Reading ingredient labels can feel daunting, but a few common red flags make it easier. 'Irritants often include synthetic dyes like Red 7 Lake, parabens, artificial fragrances, and preservatives like methylisothiazolinone," she says. The fact is that even natural ingredients like peppermint oil, lanolin, or citrus extracts can trigger reactions in sensitive users. Dr Kapoor recommends to look for full INCI (International Nomenclature of Cosmetic Ingredients) listings and certifications like ECOCERT or EWG Verified. Rise of conscious beauty: How consumers can make safer choices The conscious beauty movement has shifted focus from appearance to overall wellness and ingredient integrity. This is especially important for products like lipstick, where ingestion of trace ingredients is a possibility. Today's consumers are more informed and demand transparency, which is a welcome change. 'To make safer choices, read labels, know your sensitivities, and opt for brands that provide full ingredient disclosures and undergo clinical testing. Ethical production and environmental responsibility are added benefits—but they must go hand in hand with dermatological safety. Informed beauty is beautiful, and far safer in the long run,' she says. Though, these above-mentioned points will help you find the perfect lipstick without damaging your lips in the long run, it is crucial to consult a dermatologist or skin expert if you have persistent skin issues or are allergic to any ingredient. In a nutshell, applying lipsticks is fine and healthy unless you are using a high quality product, and keep your lips nourished to prevent dryness.
