Engineered ion channel offers precise, non-invasive control of brain activity

FAYETTEVILLE, GA, UNITED STATES, May 30, 2025 / EINPresswire.com / -- In a breakthrough advancement for neuroscience, researchers have developed RADICAL, a cutting-edge chemogenetic tool that allows for the precise manipulation of neuronal activity using a synthetic chemical, cyclohexanol ( CHXOL ). Unlike traditional methods that rely on invasive optics or slow-acting G-protein coupled receptors, RADICAL utilizes a modified TRPM8 ion channel to enable rapid and targeted control of calcium influx in neurons. This innovative tool has the potential to advance brain function research and open up new therapeutic possibilities for neurological disorders.
Current technologies for controlling neuronal activity—such as optogenetics and chemogenetics—have their limitations. Optogenetics requires invasive light delivery, while chemogenetic systems like DREADDs rely on slow and indirect cellular signaling pathways. Additionally, engineered ligand-gated ion channels, such as those based on nicotinic receptors, can result in unintended interactions with native proteins. These challenges have highlighted the need for a more efficient, non-invasive, and precise method of modulating neuronal excitability. In response to this gap, researchers sought to develop RADICAL, a novel chemogenetic tool that addresses these limitations.
In a letter (DOI: 10.1093/procel/pwae048 ) published on September 3, 2024, in Protein & Cell, a team from Zhejiang University unveiled RADICAL, an engineered ion channel activated by cyclohexanol (CHXOL). By introducing specific mutations to the TRPM8 ion channel, they created a system that responds with exceptional sensitivity and specificity to CHXOL. This innovation allows for precise neuronal control without interfering with the brain's native functions, marking a significant step forward in chemogenetics.
The key modification in RADICAL was the engineering of the TRPM8 ion channel, which is naturally expressed at low levels in the brain, minimizing potential disruptions to endogenous systems. The team introduced two critical mutations (I846F and I985K) to the TRPM8 ion channel. The I846F mutation restored CHXOL binding, while I985K enhanced voltage sensitivity, enabling robust activation even at hyperpolarizing potentials (-80 mV). Patch-clamp recordings and calcium imaging confirmed the double mutant, TRPM8-I846F-I985K's EC50 of 1.17 mmol/L for CHXOL at depolarizing potentials (+80 mV). In vivo, RADICAL demonstrated its potential: CHXOL administration enhanced fear extinction memory in mice by activating neurons in the infralimbic cortex (IL), and also increased locomotor activity when expressed in astrocytes of the ventral tegmental area (VTA). Importantly, the tool's calcium permeability and minimal cell death risk, as shown in HEK293T cells, suggest its suitability for studying calcium-dependent processes such as learning and memory.
Dr. Fan Yang, one of the co-corresponding authors of the study, said: RADICAL represents a major breakthrough in chemogenetics. Its ability to modulate neuronal activity with high precision and minimal off-target effects makes it a versatile tool for both basic neuroscience research and the development of therapeutic interventions.
With its non-invasive approach and high specificity, RADICAL has substantial potential in both research and clinical settings. It could enhance our understanding of neurological conditions such as memory disorders, addiction, and mood disorders by providing a precise way to manipulate neuronal circuits. Furthermore, future efforts to miniaturize the tool for adeno-associated virus (AAV) delivery could broaden its applicability in gene therapy. RADICAL's unique combination of speed, specificity, and safety positions it as a powerful platform for next-generation treatments of brain diseases.

Every little bit counts to up your protein intake and reach your goal. Enter: protein water. If this phrase sounds new to you, don't be intimidated—protein water is exactly what it sounds like. It's a type of water 'enhanced with some sort of supplemental protein,' says Dana White, RD, ATC, a registered dietitian and nutrition consultant specializing in sports nutrition. 'Protein water is basically water that's been infused with extra protein,' says Sohee Carpenter, CSCS, a fitness and nutrition coach, WH advisor, and author of Eat. Lift. Thrive. A lot of brands have between 10 and 20 grams of protein, and it's usually fruit-flavored, she says, adding that it's 'a really easy, convenient way to get in extra protein.' What options are available? Protein water is pretty similar to a protein shake, White says. 'Most of the products that I see have a slight boost of protein, usually from some whey-based product." There are also powders available that you can add right to your own water. Some current options include PWR Lift and Protein2o, which have whey protein. Can I drink protein water every day? 'I wouldn't say should—I would say you definitely can,' Carpenter says. It comes down to whether you actually need that protein, and when you eat it. 'What I see a lot as a dietitian is that many people consume enough protein, say, gram-for-gram based on what their needs are, but the timing of it is lousy,' White says, explaining that they might barely consume protein during the day and then have one large meal at night. 'It's much more efficient to spread out your protein,' especially for active individuals, she says. Ideally, every meal and snack has protein and adds up to your goal by the end of the day, and a protein water can certainly help you hit that target or distribute your protein better. What are the benefits of protein water? For starters, you need amino acids and protein for a variety of bodily functions, such as muscle health, immune system function, and acid-based balance, White explains. Protein water is a super convenient way to add that into your diet, and while it may not be enough of a meal or snack on its own, you can, say, pair it with a protein bar for a post-workout recovery snack. 'It's not going to solve everyone's problems in terms of protein needs, but they can certainly be an additional way to help with a protein boost here and there,' White says. Are there any downsides to protein water? There's the question of what else is in your protein drink, like a lot of sugar or an artificial sweetener that you personally might not want to consume regularly. If you want to drink one on a daily basis, weigh the pros and cons—yes, you could be consuming more protein, but you could also be 'doing yourself a disservice in another department,' White says. Also, it's not great to consume a super large dose of protein right before exercise. Who should drink protein water? Everybody needs protein, so anyone can drink protein water, White says. However, if you're highly active and/or struggling with consuming enough protein through whole foods, it might be especially helpful to try protein water, Carpenter adds. Maybe you're on the go a lot, you're vegan, or you don't cook—a protein supplement or water might be good to add into your rotation. As far as how much protein water you should drink a day, one bottle is fine, White says. When shopping for protein water, know that they're not one-size-fits-all, White says. Between all the high-protein snacks, shakes and protein powders, it's no surprise that protein is still a hot topic. If you like using supplements to help achieve your protein quota, then you know how heavy traditional protein shakes can sometimes be, even when mixed with a high-speed blender. Enter, clear whey protein. Clear whey protein is a version of whey protein isolate (a popular protein powder) that undergoes an ultrafiltration process to remove fat, lactose and other components to make it easily digestible. So instead of a thick protein shake, you end up with a nearly transparent beverage with a fruity profile that resembles a sports drink. But is it the right supplement for you and is there a right way to take it? We spoke to a pharmaceutical scientist to learn more about why this supplement is becoming popular and how it compares to its counterpart. Is clear whey protein isolate better than whey protein isolate? Although clear whey protein isn't new on the scene, TikTok has contributed to its recent popularity and sudden interest among active individuals. Seeing all the hype surrounding clear whey protein makes you wonder if it's better than whey protein isolate. Both come from whey protein, the proteins derived from the liquid released from milk. So it comes down to your preferences. 'Both are high-quality, complete proteins rich in leucine and other BCAAs,' explains John-Paul Andersen, a pharmaceutical scientist with a PhD in physiology and pharmacology, and chief science officer at Phi Health. Leucine is one of the three essential branched-chain amino acids (BCAAs), along with isoleucine and valine, that are vital to muscle growth and repair. He recommends clear whey for post-cardio or hot-weather training, while regular isolate is ideal when you want satiety or are blending it into a smoothie. 'You can rotate or combine them based on your routine and dietary goals," he says. Clear whey protein tends to be a bit more expensive than whey protein isolate because the extra filtration system is a more involved process. How should you take clear whey protein? Clear whey protein comes in powder form, similar to whey protein isolate. Some brands known to make it include GHOST, Myprotein, Isopure and Ascent. It should be mixed with water and ideally taken after a workout. Andersen points out that clear whey protein isolate is best post-workout because that's when amino acid uptake is highest and the body is primed to absorb them quickly to help with muscle repair and recovery. It's also hydrating, which makes it the perfect post-workout drink or a great choice on a hot summer day. Whatever you do, though, even if social media recommends it, don't dry scoop it. Dry scooping is when you down a mouthful of a powdered supplement like pre-workout without mixing it with water first. 'Dry scooping defeats the purpose of its refreshing formulation and can irritate the throat and cause GI discomfort,' warns Andersen. Additionally, if you're planning on taking it before or during your workout, Andersen advises pairing it with carbs for endurance. Otherwise, save it for after your sweat sesh. How to safely take clear whey protein It's safe to take one to two servings of clear whey protein daily for healthy individuals. Keep in mind, excessive intake of any protein source can strain the kidneys, particularly if you have underlying health conditions. Andersen also warns, 'Overconsumption can also cause GI issues like bloating, cramping or loose stools, depending on how the whey was filtered and what flavoring agents are included.' Who is clear whey protein best for? Clear whey protein is perfect for people who don't like protein shakes or their milky disposition when best if you're looking for a refreshing and light hydrating drink that won't leave you stuffed after a workout. Another scenario is if you know you usually don't have an appetite after exercising but want to replenish those nutrients anyway. As long as you don't have dairy allergies or sensitivities, a clear whey protein could be an option to have on hand on those hot summer days. However, sometimes a protein shake made up of whey protein isolate is what you're looking for after a lifting session because you know your next meal won't be for a while, so you need something to hold you over. If you aren't sure if clear whey protein is going to give you issues, some signs to look out for include bloating, gas, nausea or diarrhea. Sometimes the residual lactose, artificial sweeteners or flavor additives can also trigger your stomach. 'If symptoms occur, switching brands, reducing the serving size or using a plant-based alternative may help," recommends Andersen. Takeaway Clear whey protein is a refreshing alternative to regular whey protein, particularly if you want something easily digestible after your workout or if you're a heavy sweater. You can switch up the type of whey protein you take after your workouts, but like most supplements, clear whey protein should be taken alongside a well-balanced diet that works for your lifestyle and needs.
