Study reveals genetic insights about Saudi and Japanese populations

RIYADH: A new study using Saudi and Japanese pangenome samples exclusively has revealed genetic insights relating to these two populations.
'Up to 12 percent of patients with genetic disorders go undiagnosed due to reliance on reference genomes that don't reflect their population's genetic background,' said Malak Abedalthagafi, professor at Tufts Medical Center and one of the lead authors of the study.
'By constructing population-specific pangenome graphs, we improve variant calling and help close this diagnostic gap.'
The pangenome is considered a powerful reference tool to study individual and group DNA.
Similar to how a map shows a person's position in relation to their landscape, the pangenome allows researchers to compare a person's genetic makeup with the full spectrum of genetic variation found across a population.
'Having worked on the Saudi genome for several years, contributing to this project marks a meaningful step in my commitment to advancing representation in genomics and ensuring precision medicine serves diverse populations,' Abedalthagafi said.
Fellow author of the study and KAUST Professor Robert Hoehndorf explained that building the graphs, which the study calls JaSaPaGe (Japanese Saudi PanGenome), using samples from two distinctive populations offers new health insights.
'Japan and Saudi Arabia are pretty much at the opposite ends of Asia and have been separated for a long time. It gave us a chance to study the effects of population-specific pangenome graphs on variant calling when populations do not match,' Hoehndorf said.
JIHS' Yosuke Kawai, another author of the study, added that there were clinical benefits to be gained for both populations.
He said: 'The joint development of a population-specific pangenome graph for the Japanese and Saudi Arabian populations addresses a critical gap in global genomic representation.
'By integrating diverse data from both countries, we have created a powerful resource that not only improves variant detection accuracy but also holds great potential for advancing precision medicine tailored to each population's unique genetic landscape.'
The first human pangenome was reported in 2023, but none of the DNA samples collected were taken from individuals with Arab or Japanese descent, meaning it was constructed without representation from almost 10 percent of the world's population.
The study was carried out by King Abdullah University of Science and Technology, Tufts University and the Japan Institute for Health Security.

Speaking at a government press conference in Riyadh, Al-Benyan said the National Curriculum Center had developed 27 digital courses, converted 19 into interactive books, and reviewed 50 more, while updating over 6,700 digital learning resources. The reforms, he said, are designed to create a globally competitive generation, with efforts targeting 19 strategic objectives that include modernizing curricula, training teachers, embedding AI, and expanding technical training. The ministry has consolidated educational administrations from 46 to 16, and fully outsourced operations, maintenance, and environmental quality management to Tatweer Educational Transportation Services Co., enabling educators to focus on teaching. New initiatives also aim to strengthen school-family partnerships and instill values such as national identity, commitment, and tolerance among students. On infrastructure, the minister said 75 new school projects were launched, more than 15,000 buildings underwent maintenance, and 1,400 facilities were renovated. Early childhood enrollment among Saudi children has reached over 36%, while the number of identified gifted students has risen to more than 28,000. Specialized schools for gifted students, developed with the Saudi Data and Artificial Intelligence Authority and Tuwaiq Academy, are set to expand to five more regions next year. The ministry has also introduced AI curricula across all grades and a cybersecurity curriculum for high school students. Teachers and educational leaders received more than 5,200 training opportunities, including international programs such as professional master's degrees. Efforts to boost language skills include sending 100 teachers to study Chinese in China and expanding Chinese language teaching to more than 140 public middle schools. Al-Benyan said Saudi universities continue to climb global rankings, with 20 institutions now listed in the QS World University Rankings 2025 and three universities in the world's top 200. Since 2022, 3,388 Saudis have been sent to the world's top 30 universities under the King Salman Scholarship Program. In support of private sector engagement, the ministry launched the 'Madaris' platform, offering over 500 investment opportunities worth more than SR50 billion over the next five years.

Citing the 2025 Global Startup Ecosystem Report, he said Saudi Arabia's global ranking in entrepreneurship support jumped 60 places to 23rd, and five Saudi firms are poised to become unicorns. The Saudi Business Center and its online platform have delivered over 10 million services. In the industrial sector, the number of factories has grown from 7,200 before Vision 2030 to 12,500 by mid-2025, while industrial investment has climbed from SR955 billion to SR1.2 exports surged from SR178 billion to SR607 billion, with Saudi products now reaching more than 180 countries. The 'Made in Saudi' program has unlocked more than 3,000 export Saudi Industrial Development Fund issued SR4.5 billion in loans in the first half of the year, while the Saudi Authority for Industrial Cities and Technology Zones established around 2,200 ready-built factories to support investors and mining, Saudi Arabia advanced from 104th to 23rd in the global mining investment attractiveness index, with over 2,400 valid mining licenses issued by also highlighted health sector achievements, including Jeddah's recognition as the Middle East's first 'healthy city' of over one million residents by the World Health Organization, and the inclusion of seven Saudi hospitals in the 2025 global top 200 list. 'In the Kingdom, health is not just a sector but an investment in people and quality of life,' he announced that the Ministry of Media and Ministry of Education will launch a Media Scholarship Program to train Saudi students at leading universities and companies worldwide, with details to be revealed later.
