Scientists Find Molecule That Blocks Brain Cell Death In Parkinson's, Alzheimer's

A team of Australian scientists has identified a small molecule that blocks cell death, an advance that could lead to new treatments for neurodegenerative conditions like Parkinson's and Alzheimer's.
The team from the Melbourne-based Walter and Eliza Hall Institute of Medical Research (WEHI) aimed to find new chemicals that block cell death, which could help treat degenerative diseases in the future.
The findings offer hope for treatments that could slow or stop the progression of neurodegenerative diseases.
After screening over 100,000 chemical compounds, the team found a small molecule that targets a killer protein called BAX.
By interfering with a well-understood cell death protein, the molecule effectively stopped cells from dying.
"We were thrilled to find a small molecule that targets a killer protein called BAX and stops it working," said Professor Guillaume Lessene from WEHI.
"While not the case in most cells, in neurons turning off BAX alone may be sufficient to limit cell death," Lessene added.
While drugs that trigger cell death are transforming the treatment of certain cancers, the development of cell death blockers -- that could be similarly game-changing for neurodegenerative conditions -- has proven challenging.
The new molecule targets a killer protein called BAX, which kills cells by damaging mitochondria, the powerhouse of cells.
"For the first time we could keep BAX away from mitochondria and keep cells alive using this molecule," said lead author and Dewson Lab researcher Kaiming Li.
"This could pave the way for next-generation cell death inhibitors to combat degenerative conditions," Li said, in the paper published in the journal Science Advances.
While drugs that trigger cell death are transforming the treatment of certain cancers, the development of cell death blockers - that could be similarly game-changing for neurodegenerative conditions -- has proven challenging.
The study demonstrates the potential to identify drugs that block cell death and may open a new avenue to find much-needed disease-modifying drugs for Parkinson's and Alzheimer's, said the researchers.

Nagpur: In a groundbreaking development, Nagpur-based doctors performed a rare and highly advanced heart procedure that saved a young woman's life — without open heart surgery. The patient, born with a complex congenital heart defect known as Blue baby syndrome (Tetralogy of Fallot or TOF), a condition wherein a baby's skin turns bluish due to lack of oxygen in blood, had undergone a major open-heart surgery as a child. Now an adult, she recently became severely unwell due to a leaking heart valve and needed another major surgery. Instead of undergoing another high-risk open-heart operation, she received a minimally invasive treatment — much like angioplasty — through a vein in her leg. Incredibly, the patient was able to walk and talk just an hour after the procedure and was ready to go home within 24 hours. This life-saving and rare procedure, known as Transcatheter Pulmonary Valve Implantation, was successfully carried out by Dr Manish Chokhandre, senior interventional paediatric cardiologist at Arneja Heart Institute. "Redo open-heart surgeries are extremely risky, especially in adults with childhood congenital heart conditions," explained Dr Chokhandre. "We carefully studied the patient's unique heart structure, custom-designed a special valve, and performed the entire procedure without opening the chest. " Welcoming the advancement, Dr Amar Amale, president of Cardiological Society of India (CSI), Vidarbha chapter, said, "Gone are the days when children had to undergo open-heart surgery with large chest incisions. Paediatric cardiology has advanced to a level where even a complex congenital heart condition in an adult, who earlier had childhood surgery, can now be treated through a simple leg vein — just like an angioplasty. It's a huge leap in cardiac care." "Paediatric cardiology needs more attention. One in every 100 children is born with a heart defect. Many go undiagnosed or are diagnosed too late," said Dr Chokhandre, who regularly performs heart procedures on newborns. Veteran cardiologist Dr Jaspal Arneja said, "Thanks to medical advances and new technologies, even complex heart problems can be treated more safely. With artificial intelligence (AI) helping us understand heart function better, we can now offer more accurate and effective treatment." Researchers found that adults with readily available listeners demonstrated better cognitive function, even with age-related brain volume loss. This social support could potentially slow cognitive aging and delay Alzheimer's symptoms. Keeping the brain sharp is crucial for overall well-being. From cognition, memory, and emotional regulation, a healthy brain plays a key role in how we function daily. While we often turn to puzzles, exercise, and diet to maintain brain health, scientists now say something much simpler, yet often overlooked, could be just as powerful: having someone who listens! A study published in JAMA Network found a link between having someone you can count on to listen to you when you need to talk, with improved cognitive resilience. A person who is all ears According to the study, adults who have access to a good listener, a person who is available most or all of the time to provide emotional support, show significantly better cognitive resilience. This interaction will help the brain from declining, even with aging or diseases like Alzheimer's. 'We think of cognitive resilience as a buffer to the effects of brain aging and disease. This study adds to growing evidence that people can take steps, either for themselves or the people they care about most, to increase the odds they'll slow down cognitive aging or prevent the development of symptoms of Alzheimer's disease—something that is all the more important given that we still don't have a cure for the disease,' lead researcher Joel Salinas said. About 5 million people in America are living with Alzheimer's disease, a progressive condition that affects mostly those over 65. This condition interferes with memory, language, decision-making, and the ability to live independently. Salinas added that the results of this study indicate that people younger than 65 would benefit from social support. For every unit of brain volume loss, people in their 40s and 50s who had fewer listeners showed cognitive abilities similar to someone four years older than those who had more listeners. 'These four years can be incredibly precious. Too often, we think about how to protect our brain health when we're much older, after we've already lost a lot of time decades before to build and sustain brain-healthy habits. But today, right now, you can ask yourself if you truly have someone available to listen to you in a supportive way, and ask your loved ones the same. Taking that simple action sets the process in motion for you to ultimately have better odds of long-term brain health and the best quality of life you can have,' Salinas said. The study Researchers analyzed data from 2,171 participants in the Framingham Heart Study, one of the longest-running community-based studies in the U.S., with an average participant age of 63. The participants self-reported about the various types of social support they have, including access to someone who listens, offers good advice, and provides emotional support. The cognitive resilience of the participants was measured as the relative effect of total cerebral brain volume on global cognition, using MRI scans and neuropsychological assessments. Lower brain volumes tend to be associated with lower cognitive function. The researchers examined the modifying effect of individual forms of social support on the relationship between cerebral volume and cognitive performance. They found that the cognitive function of individuals with greater availability of one specific form of social support was higher relative to their total cerebral volume. This particular form of 'social support' was the availability of a listener, and it was linked with increased cognitive resilience. These kinds of questions about a person's social relationships and feelings of loneliness can tell you a lot about a patient's broader social circumstances, their future health, and how they're really doing outside of the clinic,' Salinas said. Researchers noted that further study is required for further understanding of the biological mechanisms that link psychosocial factors to brain health. 'While there is still a lot that we don't understand about the specific biological pathways between psychosocial factors like listener availability and brain health, this study gives clues about concrete, biological reasons why we should all seek good listeners and become better listeners ourselves,' Salinas concluded. Government officials said the project aims to allow researchers from the international community to study the biology of Dada Guruji Maharaj, who walks from village to village to inspire spiritual devotion in people. Samarth Bhaiya ji Sarkar, popularly known as Dada Guruji Maharaj, is said to derive his energy from nature—trees, flowing rivers, as well as the earth and small pebbles he touches as he walks along the river banks. Bhopal: The Madhya Pradesh government has approved a proposal to conduct scientific research into the 'nature-based' life and extreme physical endurance of a Jabalpur-based spiritual leader, who claims to have given up food since October 2020, relying solely on water from the Narmada since then. 'Dada Guruji, who is a spiritual leader, has been surviving without any food and simply on water while undertaking the 'parikrama' (ritual of moving clockwise round an object of devotion) of the Narmada,' Deputy Chief Minister Rajendra Shukla told ThePrint. 'This was a topic of research and so a team of medical experts from Jabalpur's medical college carefully observed him for a specified duration and then submitted their report.' He said it had still not been decided who would be approached to conduct further research. Dada Guruji Maharaj's growing popularity for his extreme endurance caught the attention of the Mohan Yadav-led BJP government, which constituted a panel of experts from the state-run Netaji Subhash Chandra Bose Medical College in May 2024 to examine his case. The issue came up for discussion in the Cabinet on 3 June, nearly a year after the medical college in Jabalpur conducted a week-long comprehensive study between 22 and 29 May on the 'various body and mind parameters during prolonged fasting and continuous physical exertion'. Following the presentation of the committee's 124-page report, the Cabinet decided to send the proposal for further research. 'The government decided that this report will be sent to researchers, individuals and institutions so that researchers from the country and the world can investigate and research the nature-based life of Dada Guru,' panchayat and rural development minister Prahlad Patel said on 'X'. Shukla, who also holds the charge of health minister, added. 'It came up for discussion during the Cabinet meeting and it was felt further research should be allowed on how a human body can sustain without food for such a prolonged duration and still have normal vital functioning.' Marginal aberrations seen during observation During the week-long research, Dada Guruji was kept in a CCTV-monitored medical room at the Jabalpur medical college under police surveillance. Experts closely observed his physical, mental, biochemical and other parameters in the morning and evening, essentially before and after he completed a 20-25 km walk on the banks of the river. They found that Dada Guruji did not consume any food during this period and only had 300-1,000 ml of water from the Narmada at the end of the parikrama. Despite 40 to 45 degree temperatures, the medical team found that his body was fit and agile with no symptoms or complaints. 'There were only a few marginal aberrations in blood sugar, uric acid, blood urea, iron and ferritin, which were consistent after fasting for seven days. In our opinion, Dada Guruji's response to strict fasting and strenuous physical activity is unique and hitherto unreported in world medical literature,' the medical college said in its report to the health department. The report was prepared under the guidance of P. Panikar, a professor of medicine; R. Mahobia, a professor of patmedhology; and Dr R.S. Sharma, an AIIMS cardiologist. 'Based on existing evidence of humans and animal studies, we suggest that this unique response of Dada Guru is due to 'medical adaptation' and markedly increased 'endurance power' for fast and adverse conditions,' they said in the report. The report also quotes Dada Guruji saying that when doctors and scientists asked him why only he derived energy from the earth and the Narmada while so many others walking the banks didn't, he explained: 'One has to be selfless and pure of heart to get this energy.'
