Mammal lifespan linked to immunity genes

There may be an evolutionary thread linking big brains, long lifespans and immune-system genes in mammals, a new study finds.
An organism's lifespan depends partly on its genes, but scientists have yet to pinpoint all of the genes that turn the dial on longevity. Long-living mammals tend to have larger brains, leading scientists to suspect that the same genes that boost longevity may also build bigger brains.
Now, in a study published April 29 in the journal Scientific Reports, scientists compared the genomes of 46 mammal species — including Homo sapiens — revealing a bevy of immunity genes that could be linked to both characteristics.
The maximum lifespan of a species refers to how long its members would live if they escaped environmental threats, such as predators or infections, and died only of old age. These lifespans vary considerably across mammals, from less than one year for the common shrew (Sorex araneus) to up to two centuries for the bowhead whale (Balaena mysticetus). The oldest person lived to 122, but one study suggests humans can reach age 150.

Some genes linked to lifespan have already been found — for instance, elephants' genetics help guard against cancer. Due to their sheer size, elephants have more cells than other mammals and thus have greater odds of developing cancerous mutations. However, researchers discovered that these behemoths carry 19 additional copies of the gene for the tumor-preventing p53 protein, which enables them to live longer lives cancer-free.
Other genes for longevity remain to be discovered, so Araxi Urrutia, a geneticist at the National University of Mexico, and her colleagues set out to search for more. They wanted to focus on "families" of related genes.
Sometimes, when a cell copies its DNA and divides to form a new cell, it can accidentally copy and paste a gene to a new spot in the genome — a phenomenon called gene duplication. Over time, additional mutations cause the genes to become slightly dissimilar and adopt distinct functions.
If gene duplications occur multiple times within a group of related genes, they can produce a large family. One example is the beta-globin family of genes, which code for the proteins that make up hemoglobin, the oxygen-transporting molecule in blood. Urrutia's team wanted to assess if any of these duplicated-gene families contributed to longevity.
They looked at more than 4,100 gene families across 46 mammal species, including baboons (Papio anubis), cats (Felis catus), and dogs (Canis lupus).. Some mammals have higher-quality genome sequences available than others.
"Sometimes genomes are not sequenced in the same way, so they could introduce some noise" in the data, said lead study author Benjamin Padilla-Morales, a geneticist at the University of Bath in the U.K. To reduce the odds that any members of a given gene family were missing from the analysis, his team focused only on species that had at least 80% of their genome sequenced.
They also focused their analysis on gene families that were detected in at least 80% of the mammal genomes they studied, so they were common to many animals. This made it possible to see if the size of the gene family in a species was proportional to that animal's maximum lifespan.
They found that gene families involved in DNA repair and inflammation were small in short-lived mammals, whereas gene duplication had expanded those families in longer-living species. It may be that carrying more copies of DNA-repair genes can help prevent dysfunctional mutations from building up over time and thus promoting aging or cancer.

Meanwhile, genes involved in immunity may promote longevity by eliminating cancers early or fighting infections efficiently, said Maria Chikina, a computational biologist at the University of Pittsburgh who was not involved with the work. Some of the expanded genes in long-living species code for proinflammatory proteins, such as interleukin-6, which indirectly kickstarts antibody production. Chikina said these genes may also be involved in maintaining healthy tissue, such as by disposing of dead cells.
The immunity genes could additionally have roles unrelated to defense. "Many immune genes have been shown to be important in development, particularly in brain development," Urrutia said. Past research has linked brain size and longevity, and most of the immunity genes tied to lifespan in the recent study were also linked to bigger brains, she added.
Regarding humans, previous work pinpointed gene variants in centenarians — people who live to at least 100 — that may partially account for their long lives, and the new study revealed that many of these genes belonged to expanded gene families. This suggests these genes might be worth exploring further to understand how genes control human lifespan.

The study shows that the evolution of longer lifespans in mammals took place alongside the duplication of immunity genes. However, it wasn't possible to determine if these gene duplications caused the longer lifespans, Chikina noted. She suggested that, in the future, they could show that the association between these genes and lifespan is robust by repeating the experiment in nonmammals, like birds, to see if the connection holds true across more branches of the tree of life.
Chikina also proposed a way to test a causal link, to see if the genes actually drive up lifespan directly. "If you see there's some sort of interesting new gene popping up in long-lived species, you could put it in a mouse and see what happens," she said.
If further research can decipher if and how these genes affect longevity, scientists may one day be able to harness that information to develop anti-aging therapeutics or to predict the maximum lifespan of an individual based on their genetic makeup.

Sleep talking can be a slightly strange experience — whether you're the one doing the midnight chattering or you're sharing a bed with someone who is talking mid-snooze. But what is sleep talking and why do we do it? Is it our brain's way of communicating something important or just idle chatter? In this feature, we dive into the science behind sleep talking - from what triggers it to whether it's ever a sign of something more serious. You'll also find expert tips on how to reduce your chances of talking in your sleep, and when to seek help - plus the latest research on the parasomnia. What is sleep talking? Sleep talking, otherwise known as somniloquy, is a type of sleep parasomnia where someone talks while asleep, unconsciously. 66% of people will sleep talk at one point in their lives 'The talking can be anything from unintelligible murmurs to full sentences,' says neuroscientist Dr Caroline Leaf. 'It normally occurs during non-REM sleep when the non-conscious working mind is still sorting and stabilizing memories and reorganizing networks in the brain,' she adds. 'It is quite common and may be related to the nature and intensity of what is being processed when sleeping,' Dr Lead fact, according to a study in 2010, 66% of people will sleep talk at one point in their lives, and 17 per cent of us have done it in the last three months. Is sleep talking associated with your dreams? Most of us would assume that sleep talking is a way of processing dreams — but experts believe there isn't enough evidence to suggest this. Although some sleep talking does happen during REM sleep (when we dream), most of the time it's associated with non-REM sleep. 'This is where it's less about active dreaming and more about fragmented neural activity,' explains Dr. Leaf. 'The mind processes unresolved mental events or stressors from the day, activating high-energy states and neuroplasticity in the brain,' she says. Why do people talk in their sleep? 1. Stress and anxiety Stress has a massive effect on our sleep, including for those who are prone to sleep talking. 'Stresses and anxieties can interfere with regular sleep patterns and increase sleep fragmentation, resulting in parasomnias like sleep talking,' explains sleep doctor Saad Karim. 'This is because the brain is more agitated or active at night when we're anxious, which results in vocalizations.' 2. Sleep deprivation Regularly not getting enough sleep, also known as sleep deprivation, can impact our health, but also how we sleep. 'Not enough sleep can make sleep more intense and more disorganized when you do sleep, increasing the risk for strange things like sleep talking,' explains the sleep doctor. 'The brain's transitions between sleep stages can become less smooth, and parasomnias are more likely.' 3. Genetics Genetics plays a big role in many health conditions, and it's no different with sleep, says Dr. Leaf. 'If a family member sleep talks or experiences other sleep behaviors like sleepwalking or night terrors, there's a higher chance others in the family may too,' she explains. But, it's not always the way, 'genetics are not destiny, they simply create a probability or propensity,' Dr. Leaf adds. 4. Medications 'Medications, such as antidepressants or sleep aids, can affect the central nervous system,' the neuroscientist explains, meaning they can 'disrupt natural sleep patterns'. Research shows that certain medications can increase fragmented sleep, which raises the likelihood of parasomnias like sleep talking. 'This is especially true if the medications interfere with REM cycles, deepen non-REM transitions or shorten the periods between non-REM and REM sleep unnaturally,' she adds. How to stop talking in your sleep There are many lifestyle changes you can make to stop sleep talking, but if you're worried about your symptoms, talk to your doctor. 1. Improve sleep hygiene Sleep hygiene doesn't only refer to your habits and lifestyle that can impact your sleep. It also refers to your environment. If you've ever woken up to loud music outside or street lights, then you may need to take steps to create a calm, peaceful environment. These things can not just be annoying when you sleep, but also make sleep talking worse. 'Ensuring your sleeping place is cool, dark, and quiet allows your brain to transition from sleeping stages smoothly - avoiding parasomnias,' Karim explains. 2. Mind-managing and stress-reducing nighttime routine Dr. Leaf believes that this is one of the most crucial things to help sleep, as 'the mind is 99% of who you are and the brain and body are only 1%.' 'Your mind is your active thinking, feeling, and choosing, and it's driving your brain,' she says. So, how should you act to calm the mind? 'Engaging in a reflective, calming mental practice before bed, like journaling, meditation, or my 5-step Neurocycle method, can help deconstruct stress before sleep,' Dr. Leaf advises. 'This reduces the risk of mental spillover that triggers sleep talking,' she explains. 3. Cutting down on or avoiding alcohol or caffeine Yes, a glass of wine can feel like a nice way to relax after a hard day, but as Dr. Leaf says, 'it weakens the brain's built-in sleep rhythm, often causing parasomnias. Cutting alcohol close to bedtime gives your brain a better chance at deep, uninterrupted rest.' And what about that coffee you had at lunchtime, will it bring on or make sleep talking worse? Caffeine can stay in our system for up to 10 hours, and it 'can reduce melatonin production and delay sleep onset. It also keeps the brain in a hyper-alert state longer than we realize.' So, what's the solution? 'Removing it from your afternoon and evening routine could restore natural sleep transitions and reduce talking during sleep, but coffee affects everyone differently, so work out your unique response.' 4. Stick to a consistent sleep schedule When you have an erratic sleep schedule - i.e going to bed at different times every night, it can confuse your circadian rhythm. This is your internal body clock, which helps you naturally wake up and feel sleepy. 'Creating a consistent, calm, tech-controlled and curated bedtime routine sends a signal to the brain to wind down,' the neuroscientist explains. 'This helps regulate neurochemical balance, especially serotonin and GABA, which are critical for sleep stability.'

German Volume Training (GVT) could help you pack on muscle size and strength — here's how, according to a personal trainer

German Volume Training, or GVT, is a method that uses 10 sets and 10 reps of a compound exercise to maximize your efficiency (and gains) in the gym. With just 60 seconds between sets and as few as two exercises included per gym session, the training technique could help you pack on size pretty fast in just a matter of months. Recently, I spoke with Max Taylorson, a London-based personal trainer, who uses the method to help his clients hit their muscle mass targets with volume and overload. As a trainer myself, I had never heard of the method before, which Taylorson tells me was first used by the German national weightlifting team and later adopted by strength coach Charles Poliquin. You can certainly trust the Germans to be efficient — and this method is just that, trust me. Here's everything you need to know about GVT, including how it can help you pack on lean muscle size and take your muscles to their absolute limit. German Volume Training: What is it, and how to use it There's little room to maneuver when it comes to GVT, as you'll be rigidly sticking to 10 sets of 10 reps, taking just 60 seconds of rest between each set. Taylorson tells me you're looking to include two compound exercises in a session, like a squat and bench press. You'll lift at roughly 60-70% of your one-rep max (the maximum you can lift for that movement), and you'll know if you've hit the mark because it should feel like you could maybe achieve 15 to 20 reps at that weight. Remember, there are 10 sets to reach in total, so avoid increasing or decreasing the weight throughout your working sets. Each set will begin to feel more challenging, so keep aiming for 10 reps, if you can. Once you can comfortably hit all 10 reps for 10 sets, it's time to add weight incrementally. Doing so will help you break through plateaus and improve strength, muscle gain and endurance. It builds muscle quickly due to high volume. 'It builds muscle quickly due to high volume,' Taylorson explains. 'GVT uses a simple and easy-to-follow structure, which makes it easy to track progress and pushes muscles to their maximum potential, leading to growth.' Taylorson first discovered GVT for his own training regimen, having struggled to put on size previously. 'This type of program helped me increase volume,' he says. 'Focusing on big compound lifts, doing 10 sets of 10 reps, overloads the muscles. It was never time-consuming either, so great for my clients who are on a tight schedule.' Taylorson reminds me that less is more…despite the 'more is more' structure of GVT. By that, he means only pick two exercises per session. If you want to add accessory work after, you can, but only pick a couple of exercises to finish with, using a more typical weightlifting structure (so not 10 x 10). He also recommends opposing muscle groups during the session, like the lower and upper body, so that you don't overtrain a single muscle group. Tempo Try to focus on keeping movements controlled and working muscles through their full range of motion during each exercise. That means half reps don't count, and neither do rushed reps. Tempo plays into time under tension, an important part of hypertrophy, or the muscle growth process. Aim to count three to four full seconds during both phases of the movement, for example, the lowering and standing phase of a squat. Add a pause or beat at the bottom and top of the movements, too. Programming GVT Taylorson says programming depends on you and your workout goals, but try to train several times per week with rest days prioritized. As a trainer myself, I also recommend keeping a close eye on your form and stopping during a set before it, or you, go to absolute failure. Your results and recovery will also depend on your diet, so refueling with plenty of protein-rich foods is important. Here's how much protein you need to build muscle. An example GVT workout format: Day 1: Bench press and back squat (chest and legs) Day 2: Rest, core, or active recovery Day 3: Rest Day 4: Row and deadlifts (back and posterior chain) Day 5: Rest Day 6: Shoulder press and pull-ups (arms, shoulders, back) Day 7: Reset to day 1 Alternate between your exercises, resting after every set. Finish with any accessory work (if you want to, this is optional), and complete as many reps of the 10 as you can. Once you manage 10 sets of 10 quite comfortably, add load. Try to stick to a program for four weeks before switching things up or mixing in new exercises. Remember, the above is only a guideline. Beginners, the German Volume Training Method may not be suitable to start your weightlifting journey with, so if you do want to try it, work closely with a personal trainer who can keep you safe and tailor your program accordingly. But it stands on the shoulders of a giant: When NASA published the Hubble Ultra Deep Field image in 2004, it stunned the world of astronomy. A composite of 800 images from exposures totaling 11 days, the deep image of an otherwise unremarkable part of the night sky revealed nearly 10,000 galaxies, many among the most distant known. Now, JWST has observed that same patch of sky with different eyes — and found 2,500 more objects. Crucially, they're even more distant. JWST's new take on the Hubble Ultra Deep Field, named the MIRI Deep Imaging Survey (MIDIS), is the deepest-ever mid-infrared image of that part of the night sky. The extraordinary new image is the result of nearly 100 hours of observing time using the space observatory's Mid-Infrared Instrument (MIRI) and Near-Infrared Camera (NIRCam). It includes hundreds of extremely red galaxies, some of which may date back to less than a billion years after the Big Bang. Using just one of MIRI's filters, JWST took an exposure of the night sky for 41 hours — the longest single-filter observation it has performed of an extragalactic field to date. The plan was to capture galaxies in mid-infrared light — something neither Hubble nor human eyes can detect — which also revealed previously unseen regions of dust and old, red stars. Processing such images requires filters that assign a different color to each different wavelength of light. In this image, galaxies rich in dust and star-forming activity are orange and red, extremely distant compact galaxies are greenish, and galaxies bright in the near-infrared are blue and cyan. Researchers described the image in a paper in the journal Astronomy & Astrophysics, along with a slider tool, a pan video and a transition video with the Hubble Ultra Deep Field for comparison.
