American Brandon Hicklin had a clear lead, but Simbine, one of the all-time greats of anchor-leg running, hunted him down, taking him just before the line as he clocked a 37.61 sec world lead to claim victory by five-hundredths of a second.
Canada, the Olympic champions, were third in 38.11.
Simbine's run was reminiscent of his lightning bolt effort to give South Africa an ill-fated gold at the 2021 World Relays in Poland, a medal the team was forced to return after one of the members, Thando Dlodlo, failed a doping test.
It's taken four years to get into a position to take another shot at a World Relays gold and Simbine wasn't going to miss out again.
The men's 4x400m outfit, runners-up last year, stepped up to the top of the podium with a great all-round effort on Sunday.
Gardeo Isaacs went around the first lap in 45.39, and then matric pupil Udeme Okon, the 2024 under-20 world 400m champion, delivered a well-paced second lap in 44.24 before fellow schoolboy Leendert Koekemoer, the youngest in the team at 17, pulled the team into first place with a final burst on the home straight, completing his leg in 44.23.
Nene closed it out with an impressive 43.64.
In the women's 4x400m Shirley Nekhuibui started, handing over to Miranda Coetzee and Precious Molepo before Zeney van der Walt defended South Africa's position to claim a historic gong, only 12-hundredths behind the second-placed US.
And the South African mixed 4x400m team ended fifth in their final, clocking 3:16.29.
With the performances on Sunday, it's hard to imagine South Africa's eight-year world championship medal drought extending beyond the Japan showpiece in September.
The last time they won world championship silverware was at London 2017, missing out three times in a row at Doha 2019, Eugene 2022 and Budapest 2023.

'Damian and Cheslin have also recovered from their injuries, and with their experience and knowledge of the Wallabies, it was a logical choice to recall them. 'We are determined to get things right this week. 'Prop Boan Venter got a taste of what it's like to face Australia last week, and Ruan Nortje did well against them last year. 'It will be good for a few other guys to experience playing against one of the top teams in the world, especially as we continue to build our squad depth. 'We are pleased with this squad, and we are all aligned on what we have to do to lift our standards this week.' Erasmus said he expected another physical battle against the Wallabies, who will have momentum on their side. 'There's no doubt they'll take a lot of confidence from their victory last week, and especially since that marked their first victory at Ellis Park against us in the professional era,' he said. 'We are still hurting from that result, and the coaches and players are extremely disappointed with the way we played last week. 'The Boks know we didn't play according to our standards in the second half, and we must rectify that. 'We let ourselves and the country down, so we have all the motivation we need to make things right this week.' On-loan Mosele has Chippa on his mind

Chippa United on-loan player Goodman Mosele says his focus is on helping the club reach its target of finishing in a decent position in the league and being competitive in cup competitions. He was not overly concerned about the midfield competition at his parent club, Orlando Pirates, and how he could eventually break into its starting line-up, he said. The 25-year-old has struggled to secure a place in the Pirates squad since joining from Baroka FC in 2021 . Following minimal playing time at the Sea Robbers, he was loaned to the Chilli Boys for the 2023/2024 season . He found his rhythm at Chippa, participating in 25 matches throughout all competitions and netting one goal. That form earned him a place back at the Buccaneers, but a long-term injury set him back and he only played about 200 minutes for Pirates, scoring one goal. Now he finds himself back at the Eastern Cape-based outfit. 'It has been hard since the injury. I'm still working towards being the old Goodman that was at the peak of his game,' Mosele said at training in East London this week. 'It will take time before I get there, I know, and I'm pushing hard, I know I will get there. 'Pirates is my home, I know, but it is not on my mind currently. I want to help Chippa get a top eight in the league.' The last time he was at Chippa, he worked with Siphelele Luthuli, Brooklyn Poggenpoel, Ronaldo Maarman, Craig Martin and Sinoxolo Kwayiba in midfield. The Eastern Areas Bowling Association hosted a highly successful Ladies Mini Districts Tournament at Port Alfred Bowling Club at the weekend, drawing together seven districts from across the Eastern Cape for three days of thrilling competition. From Friday to Sunday, the greens were alive with exciting play as teams battled it out for top honours. Eastern Areas District made an early statement, holding the overall lead from day one. In the individual team standings, Dianne Ahrens from the Port Elizabeth District shone brightly as her side secured the top spot. Sunday proved decisive, with the Port Elizabeth District putting in a dominant performance by winning all their matches. This final push saw them lift the overall trophy, while Ahrens and her unbeaten team claimed Individual Team Gold. Port Elizabeth also secured the individual team silver, capping a remarkable weekend for the district. Despite their strong start, Eastern Areas District had to settle for the overall silver medal, while the other districts showcased excellent skill and spirit throughout.
