Pfizer will end development of weight-loss pill over fears about its impact to the liver
The move comes after a patient in a trial suffered a liver injury potentially caused by the oral GLP-1 drug known as danuglipron.
The patient, who was not identified, did not experience any symptoms and their injury 'resolved' after they stopped taking the drug.
'While we are disappointed to discontinue the development of danuglipron, we remain committed to evaluating and advancing promising programs in an effort to bring innovative new medicines to patients,' Pfizer's Chief Scientific Officer Dr. Chris Boshoff said in a statement.
The Swiss company reviewed all clinical data for danuglipron and recent input for regulators before making their decision.
Its statement also suggested the patient's liver enzymes were elevated. That often is indicative of damage to cells in the organ, according to the Cleveland Clinic. Pfizer told CNBC that the trial rapidly increased the dose of the pill over a short span of time.
The company had previously scrapped the development of a twice-daily version of the drug in 2023 after trial patients experienced vomiting and nausea.
A once-daily pill could have had a competitive edge over other weight loss drugs taken twice a day.
Several companies are developing weight loss pills after surging popularity of the shots that target an intestinal hormone called GLP-1. Ozempic and Wegoy are popular injectables related to CLP-1. Oral drugs are also considered to be easier to make than injectables, as well as easier to take.
Obesity is a serious and common chronic disease. In the U.S. more than two in five adults have obesity.
The investment bank Morgan Stanley said the global market for obesity drugs is expected to reach $105 billion by 2030 — and as high as $144 billion. Comparatively, it noted that sales of branded obesity drugs had totaled $6 billion in 2023.
Pfizer said that it would continue development of an experimental oral drug targeting a different hormone.
'Cardiovascular and metabolic diseases including obesity remain important areas of unmet medical need, and we plan to continue applying our global capabilities to advance a pipeline of investigational treatments that have the potential to fill critical gaps in patient care, including continued development of our oral GIPR antagonist candidate and other earlier obesity programs,' he added.

This herb can improve brain function, slash stress, and even reduce inflammation
It's a herb with several names. Some know it as brahmi. Others call it water hyssop. It's also referred to as bacopa, or bacopa monnieri. But, many don't know 'the herb of grace' at all. Still, the perennial herb has been used for centuries in Indian Ayurveda traditional medicine, according to the Cleveland Clinic, and boasts brain boosting benefits, among others. 'Bacopa monniera protects the brain through antioxidants,' registered dietitian Sarah Thomsen Ferreira previously told the clinic. 'It helps to stimuli enzymes in our body's own antioxidant defense system.' Antioxidants include vitamins and minerals that prevent cell damage and are found in many foods. A diet high in antioxidants can help to reduce the risk of chronic disease. Bacopa's brain impacts haven't been studied in humans, but trials in animals have 'established a biological rationale for protective properties,' according to the Alzheimer's Drug Discovery Foundation. The small trials have made it challenging to draw firm conclusions, but a systematic review of the research reported at least one statistically significant improvement on at least one neuropsychological measure in the majority of trials it analyzed, the non-profit said. But, that's not the only reason you may want to use the tropical herb in your next cup of tea or protein shake -- after consulting with a doctor, of course. Bacopa is an adaptogen, the clinic notes. Adaptogens are groups of plants and herbs that can reduce stress. It is believed they work by interacting with the hypothalamic-pituitary-adrenal axis: a group of three endocrine system organs that initiate the body's stress response, according to UCLA Health. The organs include the brain's hypothalamus and pituitary gland and the kidneys' adrenal glands. All of the organs regulate hormones, which cause stress. Lastly, several studies have found that bacopa can reduce inflammation in the central nervous system and related pain. Inflammation, which is our immune system's natural response to invaders, can be harmful when it persists. Chronic inflammation has been linked to heart disease, respiratory illness, autoimmune disease, arthritis, dementia, cancer, and other conditions, according to the Cleveland Clinic. Bacopa is generally considered to be safe, but not all of its risks have been studied and may vary by individual. The foundation warns that some sources could be unsafe, depending on where the plant is grown. 'Consider choosing supplements whose content has been verified by a third party,' it said.

Could GPL-1 weight loss soon come in pill form? What we know
Eli Lilly, the pharmaceutical company behind GLP-1 injection Zepbound, is working to bring its weight loss drug to the market in pill form. In an Aug. 7 earnings call, Eli Lilly CEO David Ricks told investors that the company plans to submit the drug, orforglipron, to the FDA for approval by the end of the year. Dan Skovronsky, chief scientific officer and president of Lilly Immunology, said that the company was "very pleased" with the safety and efficacy of orforglipron as seen in trials so far. Unlike injectable obesity drugs, which are peptides designed to mimic the appetite-controlling GLP-1 hormone, orforglipron is a small molecule pill that is easier to manufacture and package, said Kenneth Custer, president of Lilly's cardiometabolic health division. Lilly views the once-daily pill as a promising alternative to injections that could be used for early intervention and long-term disease management, Custer said. Here's what we know so far about the proposed weight loss pill. What have the trials found? In a clinical trial, the company's weight loss pill resulted in a nearly 12% weight loss at 72 weeks for participants who took the highest dose of the medication. On average, participants lost 7.8% to 12.4% of their body weight, and 40% of people on the highest dose lost more than 15%, Eli Lilly reported. The most commonly reported side effects included gastrointestinal issues consistent with other GLP-1s, such as nausea, vomiting, and diarrhea. Five to ten percent of patients discontinued the drug across doses, the company said, which was one of two late-stage trials the drug must pass before being submitted for federal approval. While Eli Lilly saw the results as indicators of success, investors on the call expressed concerns. Some had hoped that its efficacy would more closely resemble that of existing injections on the market — which can result in up to a 20% body weight loss — while others noted the trial drop-out rate of 12% due to side effects or personal reasons. When will the weight loss pill hit the market? While there is not yet an official timeline, Eli Lilly executives have said they plan to submit orforglipron for federal approval by the end of the year.
